We report a potential means of selectively delivering matrix metalloproteinase (MMP) inhibitors to target tumour sites by use of a bioreductively activated Co(III) carrier system. The carrier, comprising a Co(III) complex of the tripodal ligand tris(methylpyridyl)amine (tpa), was investigated with the antimetastatic MMP inhibitor marimastat (mmstH(2)). The X-ray crystal structure of [Co(mmst)(tpa)]ClO(4) x 4H(2)O was determined and two-dimensional NMR revealed the existence of two isomeric forms of the complex in solution. Electrochemical analysis showed that the reduction potential of the complex is suitable for it to be bioreductively activated at hypoxic tumour sites. In vitro assays confirmed the stability of the prodrug in solution prior to reduction and revealed very low cytotoxicity against A2780 cells. In vivo testing in mice showed a higher level of tumour-growth inhibition by the complex than by free marimastat. Both free marimastat and and its Co(III) complex increased metastasis in the model used, with the complex significantly more active.  相似文献

Magnetically aligned bicelles are increasingly being used as model membranes in solution- and solid-state NMR studies of the structure, dynamics, topology, and interaction of membrane-associated peptides and proteins. These studies commonly utilize the PISEMA pulse sequence to measure dipolar coupling and chemical shift, the two key parameters used in subsequent structural analysis. In the present study, we demonstrate that the PISEMA and other rotating-frame pulse sequences are not suitable for the measurement of long-range heteronuclear dipolar couplings, and that they provide inaccurate values when multiple protons are coupled to a 13C nucleus. Furthermore, we demonstrate that a laboratory-frame separated-local-field experiment is capable of overcoming these difficulties in magnetically aligned bicelles. An extension of this approach to accurately measure 13C-31P and 1H-31P couplings from phospholipids, which are useful to understand the interaction of molecules with the membrane, is also described. In these 2D experiments, natural abundance 13C was observed from bicelles containing DMPC and DHPC lipid molecules. As a first application, these solid-state NMR approaches were utilized to probe the membrane interaction of an antidepressant molecule, desipramine, and its location in the membrane.  相似文献

All possible combinations of mixed pentaarylantimony compounds bearing p-methylphenyl and p-trifluoromethylphenyl groups were synthesized; ArnTol5-nSb (n=0-5: Ar=p-CF3C6H4, Tol=p-CH3C6H4): Tol5Sb (1), ArTol4Sb (2), Ar2Tol3Sb (3), Ar3Tol2Sb (4), Ar4TolSb (5), and Ar5Sb (6). Compounds 2-5 are the first well-characterized examples of mixed acyclic pentaarylantimony species. The structures of 2-6 were determined by X-ray crystallography to feature trigonal-bipyramidal (TBP) geometry with the more electronegative p-trifluoromethylphenyl substituents selectively occupying the apical positions. Consideration of the chemical shifts of the ipso carbons of the aryl and tolyl groups suggested that the solution structures of 1-6 were also TBP, although their pseudorotation could not be frozen even at -80 degrees C. Ligand-exchange reactions (LERs) took place between 1 and 6 at approximately 60 degrees C in [D6]benzene and all six species 1-6 were found in the equilibrium mixture. The relative stabilities of 1-6 were determined quantitatively by comparison of the observed molar ratios of 1-6 in equilibrium with calculated statistical molar ratios, and Ar2Tol3Sb (3) was found to be the most stable. The ligand-coupling reactions (LCRs) of 2-5 in solution were greatly accelerated by adding Cu(acac)2 or Li+TFPB- (TFPB: [3,5-(CF3)2 C6H3]4 B), whereby the rate becomes comparable to the LER. The use of flash vacuum thermolysis (FVT) allowed the LCR to occur with very little ligand-exchange; the exception ArTol4Sb had very fast ligand-exchange. The selectivities of the LCRs were calculated from the yield of the biaryls synthesized by using FVT. These results were highly consistent with reactions catalyzed in solution, in which bitolyl was not obtained at all. The experimental results suggested that the LCR of pentaarylantimony compounds proceeds in the manner of apical-apical coupling.  相似文献

The 3,5-dimethylphenylcarbamates of cellulose and amylose bearing a small amount of 3-(triethoxysilyl)propyl residues were synthesized by a simple process and efficiently immobilized onto a silica gel support by intermolecular polycondensation of the triethoxysilyl groups. The obtained chiral packing materials (CPMs) were evaluated by high-performance liquid chromatography. The polysaccharide derivatives containing about 1-2% of the 3-(triethoxysilyl)propyl residue were efficiently immobilized with a high chiral recognition. The immobilized CPMs could be used with the eluents containing chloroform and tetrahydrofuran, which cannot be used with the conventional coated-type CPMs. By using these eluents, the chiral recognitions for many racemates could be improved.  相似文献

A novel catalytic activation of the leaving group in the S(N)2 reaction is achieved as an extension of our mercuric triflate-catalyzed reactions. Derivatives of anilinoethyl 4-pentynoate reacted smoothly with catalytic amounts of Hg(OTf)(2) to give indoline derivatives in excellent yield with efficient catalytic turnovers under very mild conditions. The reaction of optically pure secondary alcohol derivatives resulted in inversion of stereochemistry, which is a definitive feature of the S(N)2 reaction. The procedure is applicable for benzoazepine synthesis.  相似文献

Addition reaction of two geometrical isomers of 1-chlorovinyl p-tolyl sulfoxides, derived from unsymmetrical ketones and chloromethyl p-tolyl sulfoxide, with lithium enolate of tert-butyl acetate gave single isomers of the adduct, respectively. Treatment of each diastereomer with i-PrMgCl resulted in the formation of magnesium carbenoids. Highly regiospecific 1,3-CH insertion reaction was found to take place from the magnesium carbenoids to afford cyclopropanes in high yields. Stereochemistry of the adducts, reaction mechanism, and origin of the regiospecificity are discussed.  相似文献

Crystallization in n-alkane ultrathin films supported by solid substrates is investigated by molecular dynamics simulation. We consider a relatively short n-alkane, undecane C11H24, on a flat substrate of varied degree of attraction. By the use of the united atom model for n-alkane, we reveal several characteristics of the thin film crystallization. It is found that the crystalline films consist of thin crystalline lamellae where chains are either parallel or perpendicular to the substrate. The relative amount of both types of lamellae changes systematically with film thickness, substrate attraction, and crystallization temperature; thicker films on substrates of higher attraction comprise dominant parallel lamellae, while thinner films on substrates of weaker attraction prefer the perpendicular lamellae. A clue to the morphogenesis is suggested to be the marked preference of the chain ends to locate on the free surface and on the effectively repulsive substrate. It is also shown that the perpendicular crystals, both on the free surface and on the solid substrate, have melting points higher than that of the bulk.  相似文献

Phenyl furyl sulfides ( 3a‐j ) and phenyl furyl ethers ( 3k‐n ), which are useful in synthesizing furocondensed 3‐ring compounds, can be synthesized by nucleophilic substitution of nitrofurans having electron withdrawal groups. In our experiments using 5‐nitrofurans having electron withdrawal groups ( 2a‐i ), nucleophilic substitution readily occurred with the benzenethiolate anion of thiosalicylic acid ( 1a ), the benzenethiolate anion of thiosalicylate ester ( 1b ), and the phenylate anions of salicylate esters ( 1c‐d ) to yield phenyl furyl sulfides ( 3a‐j ) and phenyl furyl ethers ( 3k‐n ).  相似文献
