We observed by using atomic force microscope (AFM) phospholipid (1,2-dipalmitoyl-sn-glycero-3-phosphocholine) monolayers on mica being annealed and cooled to a selection of temperatures through steps of 2-4 degrees C/min. The annealed phospholipid monolayers started to disappear at 45-50 degrees C and disappeared completely above 60-63 degrees C under AFM observation. The phospholipid monolayers reformed when the samples were cooled below 60 degrees C and developed from fractal into compact monolayer films with decreasing temperatures. Simultaneously the height of the reformed phospholipid films also increased with decreasing temperatures from 0.4 nm to the value before annealing. The observed thermal features are attributed to a phase-transition process that upon heating to above 45-50 degrees C, the lipids condensed in the monolayers transform into a low-density expanded phase in which the lipids are invisible to AFM, and the transformation continues and completes at 60-63 degrees C. The lipid densities of the expanded phase inferred from the dissociated area of the condensed phase are observed to be a function of the temperature. The behavior contrasts with a conventional first-order phase transition commonly seen in the Langmuir films. The temperature-dependent height and shape of the reformed phospholipid films during cooling are argued to arise from the adjustment of the packing and molecular tilting (with respect to the mica surface) of the phospholipids in order to accommodate more condensed phospholipids. 相似文献

For the molecule KCs the potential energy has been calculated for the 72 lowest molecular states Omega. Using an ab initio method the calculation is based on nonempirical pseudopotentials within the range of 5.0a0-34.0a0 of the internuclear distance R. Gaussian basis sets have been used for both atoms and spin-orbit effects have been taken into account through a semiempirical spin-orbit pseudopotential added to the electrostatic Hamiltonian. The spectroscopic constants for 60 states have been calculated by fitting the calculated energy values to a polynomial in terms of the internuclear distance R. The components of the spin-orbit splitting for (1,2,5,6) 3Pi and (1) 3delta have been identified. The comparison of the present results with those available in the literature shows a very good agreement, while the other results, to the best of our knowledge, are given here for the first time. 相似文献

Since hydration forces become very strong at short range and are particularly important for determining the magnitude of the adhesion between two surfaces or interaction energy, the influences of the hydration force and elastic strain energy due to hydration-induced layering of liquid molecules close to a solid film surface on the stability of a solid film in a solid-on-liquid (SOL) nanostructure are studied in this paper. The liquid of this thin SOL structure is a kind of water solution. Since the surface forces play an important role in the structure, the total free energy change of SOL structures consists of the changes in the bulk elastic energy within the solid film, the surface energy at the solid-liquid interface and the solid-air interface, and highly nonlinear volumetric component associated with interfacial forces. The critical wavelength of one-dimensional undulation, the critical thickness of the solid film, and the critical thickness of the liquid layer are studied, and the stability regions of the solid film have been determined. Emphasis is placed on calculation of critical values, which are the basis of analyzing the stability of the very thin solid film. 相似文献

A new program to characterize polyethylene glycol-modified (PEGylated) proteins is outlined using capillary zone electrophoresis (CZE). PEGylated ribonuclease A and lysozyme were selected as examples. Five separation procedures were compared to select out the mixed buffer of acetonitrile-water (1:1, v/v) at pH 2.5 as the best to characterize the PEGylated proteins without sample pretreatment. Polyethylene oxide (PEO) with a high molecular mass of 8 x 10(6) was applied to rinse the capillary to form a dynamic coating which would decrease the undesirable proteins adsorbed to the inner wall of the silica. The electroosmotic flow (EOF) mobility of the five procedures was determined, respectively. It is found that acetonitrile is mainly responsible for the good resolution of PEGylated proteins with the help of PEO coating in the semi-aqueous system. The low EOF mobility and current in the semi-aqueous system might also have some responsibility for the high resolution. The semi-aqueous procedure described in this paper also demonstrates higher resolution of natural proteins than aqueous ones. 相似文献

Abstract— In this study we compared the plasma distribution and arterial accumulation of a photosensitizer, benzoporphyrin derivative (BPD), in two models of atherosclerosis: the spontaneous lesions of the Watanabe heritable hyperlipidemic (WHHL) rabbit and induced lesions of the balloon-injured, cholesterol-fed New Zealand white (NZW) rabbit. Selective uptake and retention of a photosensitizer by the abnormal portion of a vessel is a necessity in order for photodynamic therapy to become a successful modality for inhibition of intimal hyperplasia, selective removal of atherosclerotic tissue or imaging of diseased arteries. Liposome-based formulations were compared to freshly isolated native low density lipoprotein (LDL) and acetylated-LDL (Ac-LDL) as delivery vehicles for BPD. Plasma distribution of the photosensitizer was analyzed by KBr density gradient ultracentrifuga-tion. Although the delivery vehicle influenced plasma distribution immediately postinjection, BPD subsequently partitioned according to the plasma concentration of the lipoproteins. Photosensitizer level in plaque and normal artery specimens was determined by ethyl acetate extraction and spectrofluorometric measurement. The measurement of BPD in normal and atherosclerotic arterial tissue demonstrated a selective accumulation in atherosclerotic tissue. Preassociation with LDL and Ac-LDL enhanced accumulation of BPD in atherosclerotic tissue when compared with normal artery (mean ratios of 2.8 and 4.1 were achieved, respectively). These results indicate that the preferential uptake of BPD by atherosclerotic plaque can be enhanced by preassociation with plasma lipoproteins, suggesting that light activation could lead to a highly selective destruction of diseased vascular tissue. 相似文献

The surface shear viscosity of a myelin mimetic Langmuir monolayer is investigated upon adsorption of myelin basic protein (MBP). We measure an increase of the surface shear viscosity at picomolar concentrations of the protein, suggesting that the globular conformation of MBP changes upon adsorption at the monolayer. The conformational change enables hydrodynamic interactions of the proteins, with a typical separation of hundreds of nanometers. This unfolding is essential for the compactification of the myelin sheath, serving an enhanced saltatory signal transduction in vertebrates. The viscometry used extends the sensitivity of standard surface viscometers toward lower viscosities. 相似文献

Irradiation techniques are compared with conventional acid digestion procedures for the release of bound heavy metals in natural waters and in blood, before their determination by anodic stripping voltammetry. Ultra-violet irradiation of acidified water with a 550-W mercury vapour lamp releases bound zinc, cadmium, lead and copper after 4 h. The same results can be achieved with a 30-Mrad dose of high-energy γ-irradiation. These techniques are also effective for the release of metals in whole blood and blood plasma, where sample volumes as small as 200 μl are adequate in analyses for zinc, copper and lead. By comparison with acid digestion and solvent extraction methods, irradiation treatments offer the advantages of minimum sample manipulation and negligible reagent blanks. 相似文献

The UV-induced photochemical reactions of pentacarbonyliron with ethene in a low temperature nitrogen matrix were studied by means of the Mössbauer technique. Fe/CO/4/C2H4/ was produced by UV-irradiation of penfacarbonyliron in close proximity to ethene molecules in a pure ethene matrix, or a homogeneous cocondensed matrix. The effect of phenyl substituents on the release rates of esterase-sensitive coumarin-based prodrugs
A coumarin-based prodrug system has been recently developed in our laboratory for the preparation of esterase-sensitive prodrugs of amines, peptides, and peptidomimetics. The drug release rates from this prodrug system were found to be dependent on the structural features of the drug moiety. In certain cases, the release can be undesirably slow for drugs that are secondary amines with relatively high pKa's. Aimed at finding ways to manipulate the release rates to suit the need of different drugs, we have examined the effect of the phenyl ring substitutions on the release kinetics of such prodrugs and found that appropriately positioned alkyl substituents on the phenyl ring could help to facilitate the release by as much as 16-fold. Therefore, introduction of alkyl substituents on the phenyl ring should allow us to manipulate the release rates and, therefore, time profiles for different drugs. 相似文献
