Dye-ligand chromatography for the resolution and purification of restriction endonucleases

The resolution of restriction endonucleases from the same microorganism is conventionally achieved by lengthy fractionation protocols. We now report effective single-step procedures that exploit dye-ligand chromatography for the resolution and purification of restriction enzymes. After suitable initial screening, we demonstrated that resolution of two restriction activites can be achieved in one chromatographic step, and further purification can subsequently be effected using selected dye-adsorbents. Accordingly, we resolved in one step, Hpa I from Hpa II, Hind II from Hind III, and Sac I from Sac II. Furthermore, a three-step Chromatographic procedure has been developed to purify EcoRV suitable for commercial exploitation, as judged by the “overdigestion” and “cut-ligate-recut” quality control tests.     

Determination of sulfur dioxide in vitriol plant wastewater by using a polyNiMe4TAA electrochemically modified Pt microelectrode

A Pt microelectrode modified with nickel(II) polytetramethyldibenzo[b,i]tetraaza[14]annulene was prepared by electropolymerization of nickel(II) tetramethyldibenzo[b,i]tetraaza[14]annulene monomers and applied to determine sulfur dioxide in vitriol plant wastewater. For determination of SO2 with this electrochemically modified Pt microelectrode, the linear range was from 9.6 x 10(-6) to 2.4 x 10(-4) mol L-1, the sensitivity was 1.8 x 10(-4) A L mol-1, the detection limit was calculated to be 4.8 x 10(-6) mol L-1 (S/N = 3), the response time was less than 20 s and the relative standard deviation was found to be 2.1% on analyzing 4.8 x 10(-5) mol L-1 SO2 solution repeatedly (n = 7). These results demonstrated good accuracy compared with those obtained by the conventional iodimetric method.     

Simultaneous determination of p-hydroxybenzaldehyde, p-hydroxybenzyl alcohol, 4-(beta-D-glucopyranosyloxy)-benzyl alcohol, and sugars in Gastrodia elata blume measured as their acetylated derivatives by GC-MS

A method for the simultaneous separation and determination of the active constituents and three sugars in the roots of Gastrodia elata Blume (GE), which is used as a famous Chinese traditional herbal medicine, by gas chromatography-mass spectrometry is established. The samples are acetylated with pyridine-acetic anhydride. The contents of 4-hydroxybenzaldehyde, 4-hydroxybenzyl alcohol (HA), fructose, glucose, 4-(beta-D-glucopyranosyloxy)-benzyl alcohol (GA), and sucrose in GE are 0.004%, 0.03%, 1.36%, 1.12%, 1.97%, and 4.25%, respectively, and the detection limits are 1.5, 3.0, 11.0, 5.0, 33.0, and 35.0 pg, respectively. The contents of HA and GA in the urine and brain of a mouse are also determined. This method is simple, reliable, and quick for the simultaneous determination of the active constituents and sugars in GE.     

Synthesis of 3-epi-6,7-dideoxyxestobergsterol A

3-Epi-6,7-dideoxyxestobergsterol A (2), an analogue of xestobergsterol A, has been synthesized from dehydroepiandrosterone (3) in 15 steps. The key synthetic intermediate, 15beta,16alpha-dioxypregn-17(20)E-ene derivative 8, was prepared from the corresponding 15beta,16beta-epoxide 6 by treating with acetic acid and titanium tetraisopropoxide. The 23-oxo side chain was constructed stereoselectively by orthoester Claisen rearrangement of 8 followed by introduction of an isobutyl group. Basic treatment of the 15,23-diketone 12 followed by deprotection gave the title compound 2.     

A unified and quantitative receptor model for the microtubule binding of paclitaxel and epothilone

[formula: see text] Paclitaxel and epothilone represent the two major classes of antimicrotubule agents that promote tubulin polymerization and, presumably, mitotic arrest during cell division. A common minireceptor binding site model at beta-tubulin has been constructed for these structurally divergent compounds. Utilizing 20 amino acids identified in photoaffinity labeling experiments, the 3-D model correlates measured and predicted Ki's with r = 0.99 and rms(delta Gcalc-delta Gexp) = 0.2 kcal/mol. In addition, the model predicts the affinity of compounds not used in the training set and explains much of the SAR for the paclitaxel and epothilone families.     

Plasticization and Crystallization of Poly(ethylene Terephthalate) Induced by Water

Commercial poly(ethylene terephthalate) (PET) was treated at R. H.>80% and room temperature for a set time. The glass transition temperature (Tg) decreases with the time of exposure to high humidity. The decrease in Tg is a result of plasticization. Our data indicate that the Tg of dry PET of 76-78°C may decrease to as low a temperature as 65-67°C when it is wet. Induced crystallization of PET in the presence of water reduces the cold crystallization temperature (Tc). The structure of water-induced crystals is imperfect and can be improved in perfection by annealing. This revised version was published online in July 2006 with corrections to the Cover Date.     

Phenotyping of erythrocyte acid phosphatase and esterase D by high field strength isoelectric focusing on cellulose acetate membrane

Phenotyping of erythrocyte acid phosphatase (EAP) and esterase D (ESD) by cellulose acetate membrane isoelectric focusing (CAM-IEF) under a nonequilibrium condition is described. In an attempt to improve the method of CAM-IEF, we shortened the electrode distance to provide a higher field strength at a given (low) voltage. Various carrier ampholytes for EAP typing and various chemical separators for ESD typing were also tested. Good separations were obtained after 30 min IEF for EAP typing and 25 min for ESD typing. When applied to blood stains and stored for various periods at room temperature, the stains up to 8 months old could still be phenotyped for EAP and those up to 4 weeks old for ESD. CAM-IEF is suitable for routine forensic work of EAP and ESD phenotyping.
