研究了抗癌新药米托蒽醌(MXT)的电化学行为及与脱氧核糖核酸(DNA)的相互作用,推导了适用于研究不可逆电活性分子与DNA相互作用的电化学公式,运用该公式可以简便、快速地测定靶向分子与DNA的结合常数和结合位点数。实验发现,MXT与小牛胸腺DNA的结合以蒽醌母核的嵌插作用为主,同时,烃氨基侧链与骨架磷酸基团之间的静电吸引对母核起稳定作用,使化合物易于嵌入DNA的平面结构。MXT与DNA相互作用引起的峰电流的变化可以用于分析测定DNA。 相似文献

Ribavirin, the only small molecule available so far for treating hepatitis‐C‐virus infection, was recently used in an emergency context to treat patients with severe acute respiratory syndrome (SARS) in the early stages of the disease. To study the mechanisms responsible for the antiviral effects of ribavirin by using a photolabeling approach, we designed, synthesized, and characterized the azidotriazole nucleosides 1 and 2 as photolabeling probes of ribavirin. These probes were synthesized either by performing nucleophilic substitution of the corresponding bromotriazole nucleoside with NaN3 (Scheme 2) or by directly coupling the azidotriazole with the protected ribose sugar (Scheme 4). The azidotriazole nucleosides 1 and 2 showed a fast, clear‐cut photochemical reaction, which suggests that they are promising candidates for use in photolabeling studies. 相似文献

OPLS-AA force field and direct integration of intermolecular radial distribution functions (RDF) were employed to calculate absolute binding constants of pyridine molecules to amino group (NH2) and amide group hydrogen atoms in and first generation poly(amidoamine) dendrimers in chloroform. The average errors in the absolute and relative association constants, as predicted with the calculations, are 14.1% and 10.8%, respectively, which translate into ca. 0.08 and 0.06 kcal/mol errors in the absolute and relative binding free energies. We believe that this level of accuracy proves the applicability of the OPLS-AA, force field, in combination with the direct RDF integration, to reproducing and predicting absolute intermolecular association constants of low magnitudes (ca. 0.2-2.0 range). 相似文献

A bio-mimic bulk acoustic wave (BAW) sensor was fabricated by coating the caffeine (CAF) template molecular imprinting polymer (MIP). This sensor exhibited high selectivity and a sensitive mass response to CAF. The response range of the sensor was between 5.0 x 10(-9) and 1.0 x 10(-4) M with a detection limit of 5.0 x 10(-9) M at pH 8.0. Recoveries were 96.1-105.6%. Influencing factors were investigated in detail and optimized. A novel compound, [Co(phen)3][V10O26]·H2O, was hydrothermally synthesized and characterized by single-crystal X-ray diffraction. This compound crystallizes in the orthorhombic space group Ccca with a=13.447(3), b=29.936(6), c=23.252(5) Å, V=9360(3) Å3, Z=8 and R=0.0285. Data were collected on a Rigaku R-AXIS RAPID IP diffractometer at 293 K in the range of 1.36<θ<24.99°. The structure of the compound consists of vanadium oxide layers, which are built up from the infinite VO4 chains by corners and edges sharing. The [Co(phen)3]2+ complexes occupy the interlayer space and contact each other via π−π stacking interactions of the phen groups to form infinite one-dimensional chains. 相似文献
