小分子阿特拉津和罂粟碱检测的免疫芯片技术研究   总被引:5,自引:0,他引:5  
采用蛋白芯片竞争法对小分子半抗原的污染物进行检测。在获得特异性抗体的前提下,首先将阿特拉津半抗原进行了衍生化,然后将该衍生物和氨基罂粟碱分别与载体蛋白质卵清蛋白(OVA)进行偶联。实验证明新合成的完全抗原能够与其相应抗体发生特异性的结合。实验还对蛋白芯片检测阿特拉津进行了条件优化,其抗体固定化时间为2h,用卵清蛋白为封闭液的封闭时间为1h,样品稀释液pH值为8.0。并对阿特拉津及罂粟碱进行了定性、定量实验,结果表明:荧光信号强度随待测物浓度的降低而增强,有一定的线性趋势,阿特拉津检出限为0.001mg/L,罂粟碱检出限为0.01mg/L。  相似文献

合成了2-羟基-萘甲醛缩对甲氧基苯甲酰腙,采用元素分析和红外光谱对该化合物结构进行了表征。详细研究了该化合物在固态、不同溶剂中和有锌离子存在下的荧光光谱,探讨了溶剂极性和锌离子的存在对其荧光光谱的影响。结果表明:该化合物的固体和溶液都有很强的荧光,溶剂对其荧光性质有较大的影响,其DMF溶液在485nm处发光最强。金属锌离子具有荧光增敏性。  相似文献

Reaction of transition metal formate M(HCOO)(2).2H2O (M = Mn, Co, Ni) with 4,4'-bpy (4,4-bipyridine) has led to four new compounds with the formula M(HCOO)2(4,4'-bpy).nH2O (M = Mn, Co (1.Mn, 2.Co), n = 0; M = Co, Ni (3.Co, 4.Ni), n = 5). Compounds 1.Mn and 2.Co are isomorphous and crystallized in the tetragonal crystal system with the chiral space group P4(1)2(1)2. They are of three-dimensional diamondoid structure connected by anti-anti formate with 4,4'-bpy in the cavities of the framework reinforcing the intermetallic connections; the diamond-like net was observed also in their azide analogue (Mn(N3)2(4,4'-bpy)). Compounds 3.Co and 4.Ni are isomorphous also but crystallized in the monoclinic crystal system with the space group Cc. Both structures are uninterpenetrated 3D "CdSO4" type with big channels, constructed by anti-anti formate and 4,4'-bpy. This type of net was not observed in their azide analogue. Residing in the channels, water molecules form a new type of 1D tape constructed by vertex-sharing cyclic pentamers. Magnetic measurements were performed on all of these four compounds. 1.Mn and 2.Co are weak ferromagnets with the critical temperature Tc = 5.3 and 7.4 K, respectively. 3.Co is an antiferromagnet with Neel temperature TN = 3.0 K, and 4.Ni is a weak ferromagnet below 20 K. Hysteresis loop can be observed for 2.Co and 4.Ni at 1.8 K. As an analogue of azide, formate can be used to construct molecular architectures, which structurally and magnetically have great similarities to and also differences from those of azide. This offers a promising method for the design of new molecular architectures with formate.  相似文献

A new, practical approach to a variety of highly electrooptically active polymers for device development is described. It involves the use of a new thermally cross-linkable, hyperbranched oligomer containing nonlinear optical (NLO) chromophores as a macromolecular dopant in a common host polymer. A series of NLO polymeric blends were readily formulated and showed large and stable electrooptic (EO) coefficients (up to 65 pm/V). In comparison with previously studied linear NLO polyimides and guest-host polymers doped with molecular chromophores and even linear NLO analogous oligomers, this new approach offers clear advantages for device development in terms of improved poling efficiency, larger EO coefficients, good temporal stability, and versatile material formulation.  相似文献

In the title complex, [Mn(SO4)(C3H7NO)(H2O)2]n, each MnII ion has a distorted octahedral geometry formed by three O atoms of three different sulfate groups, one O atom of a dimethylformamide ligand and two water molecules. 5‐Sulfosalicylic acid (5‐SSA) and 3‐aminopyridine (3‐APy) crystallize in the same solvent system, resulting in two kinds of 1:1 proton‐transfer organic adduct, namely 3‐aminopyridinium 3‐carboxy‐4‐hydroxybenzenesulfonate monohydrate, C5H7N2+·C7H5O6S·H2O or 3‐APy·5‐SSA·H2O, (I), and the anhydrous adduct, C5H7N2+·C7H5O6S or 3‐APy·5‐SSA, (II). pH-zone-refining counter-current chromatography was successfully applied to the separation of cichoric acid from Echinacea Purpurea (L.) Moench. A 3.0 g quantity of sample was separated using the following two-phase solvent system: MtBE-CH3CN-water (4:1:5, v/v), 10 mM trifluoroacetic acid in organic stationary phase and 10 mM ammonia in aqueous mobile phase. The obtained fractions were analyzed by high-performance liquid chromatography and negative ion electrospray ionization mass spectrometry. Double separations were performed with the same solvent system yielding 563 mg cichoric acid at 95.6% purity.  相似文献

An electrospray ionization mass spectrometry based assay was developed for kinetic measurements and inhibitor screening of yeast hexokinase. There is considerable discrepancy in the literature as to the accuracy of kinetic data obtained for hexokinase. In the assay described herein, the product, glucose 6-phosphate was directly monitored by ion trap mass spectrometry and quantified using an internal standard, 2 deoxy-glucose 6-phosphate. The kinetic parameters, K(M) and V(max) for the two substrates were determined without using a coupling enzyme as is normally employed in the traditional spectrophotometric assay for systems lacking a chromophore. In addition, hexokinase was successfully immobilized onto an amino-link gel, and a mock library was screened against the immobilized enzyme for the identification of possible inhibitors. After comparing the mass spectra of the library before and after incubation, trehalose 6-phosphate, ADP, and oxidized glutathione were differentiated from other weak or non-inhibitors. Inhibition behavior of ADP with respect to ATP was further evaluated with the ESI-MS assay and the value of K(i) was determined. This ESI-MS assay was demonstrated to be both accurate and precise for determining kinetic constants and for identifying enzyme inhibitors.  相似文献
