Two ladder‐like polysilsesquioxanes (LPS) containing side‐chain maleimide groups have been synthesized successfully by reacting N‐(4‐hydroxyphenyl)maleimide (HPM) with LPS containing 100 mol % of chloropropyl groups (Ladder A ) and 50 mol % of each methyl and chloropropyl group (Ladder B ). HPM was synthesized by reacting maleic anhydride with 4‐aminophenol, and the resulting amic acid was imidized using p‐toluenesulfonic acid as a catalyst (Scheme 1 ). The LPSs were characterized by Fourier transform infrared (FTIR), 1H nuclear magnetic resonance (NMR), proton‐decoupled 13C NMR, 29Si NMR, wide‐angle X‐ray diffraction (WAXD), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). Characterization indicated that these polymers had ordered ladder‐like structures with possible defects. These polymers were soluble in common solvents at ambient temperature, which suggested that they were not crosslinked. Both the polymers and the HPM were cured, and their kinetics were followed by dynamic DSC. The Ozawa and Kissinger methods were used to calculate activation energies for curing. The binding of lomefloxacin to bovine lactoferrin (BLf) in a dilute aqueous solution was studied using fluorescence spectra. The binding constant (K) and the number of binding sites (n) were obtained by a fluorescence quenching method. The binding distance (r) and energy-transfer efficiency (E) between lomefloxacin and bovine lactoferrin were also obtained according to the mechanism of Fo?rster-type dipole-dipole nonradiative energy-transfer. The effect of lomefloxacin on the conformation of bovine lactoferrin was also analyzed by synchronous fluorescence spectroscopy. The interaction between lomefloxacin and bovine lactoferrin is strong. Lomefloxacin can affect the conformation of bovine lactoferrin to some degree.  相似文献

Thymus immunosuppressive pentapeptide (TIPP) is a novel anti-inflammatory peptide with high efficacy and low toxicity. This study aims to establish a selective LC-MS/MS method for analyzing the analyte TIPP in biological samples, laying the foundation for further PK and PD studies of TIPP. Protein precipitation was conducted in acetonitrile supplemented with 2% formic acid and 25 mg/mL dithiothreitol as a stabilizer, which was followed by backwashing the organic phase using dichloromethane. The chromatographic separation of TIPP was achieved on a C18 column with a gradient elution method. During positive electrospray ionization, TIPP was analyzed via multiple-reaction monitoring. The linear relationships between the concentration of TIPP and peak area in murine plasma cell lysates, supernatants, and the final cell rinse PBS were established within the ranges of 20–5000 ng/mL, 1–200 ng/mL, 10–200 μg/mL, and 0.1–20 ng/mL, respectively (r2 > 0.99). Validated according to U.S. FDA guidelines, the proposed method was proved to be acceptable. Such a method had been successfully applied to investigate the pharmacokinetics of TIPP in mice via subcutaneous injection. The plasma half-life in mice was 5.987 ± 1.824 min, suggesting that TIPP is swiftly eliminated in vivo. The amount of TIPP uptake by RBL-2H3 cells was determined using this method, which was also visually verified by confocal. Furthermore, the effective intracellular concentration of TIPP was deduced by comparing the intracellular concentration of TIPP and degrees of inflammation, enlightening further investigation on the intracellular target and mechanism of TIPP.  相似文献

Nano-hydroxyapatite (HA)/poly(l-lactide) (PLLA) composite microspheres with relatively uniform size distribution were prepared by a solid-in-oil-in-water (s/o/w) emusion solvent evaporation method. The encapsulation of the HA nanopaticles in microshperes was significantly improved by grafting PLLA on the surface of the HA nanoparticles (p-HA) during emulsion process. This procedure gave a possibility to obtain p-HA/PLLA composite microspheres with uniform morphology and the encapsulated p-HA nanoparticle loading reached up to 40 wt% (33 wt% of pure HA) in the p-HA/PLLA composite microspheres. The microstructure of composite microspheres from core-shell to single phase changed with the variation of p-HA to PLLA ratios. p-HA/PLLA composite microspheres with the diameter range of 2–3 μm were obtained. The entrapment efficiency of p-HA in microspheres could high up to 90 wt% and that of HA was only 13 wt%. Surface and bulk characterizations of the composite microspheres were performed by measurements such as wide angle X-ray diffraction (WAXD), thermal gravimetric analysis (TGA), environmental scanning electron microscope (ESEM) and transmission electron microscopy (TEM).  相似文献

Based on the independent, continuous and mapping (ICM) method and homogenization method, a research model is constructed to propose and deduce a theorem and corollary from the invariant between the weight filter function and the corresponding stiffness filter function of the form of power function. 本文报道了用溶胶 -凝胶 (sol- gel)方法 ,在 La Al O3 (10 0 )衬底上制备 L a1-x Cax Mn1.0 3 O3 外延薄膜的输运性质 .Ca含量 x在 0 .2~ 0 .6 2 1范围内 ,电阻率随温度的变化关系 ,是从类半导体行为向金属导电行为转变 ,在 x≥ 0 .5的 4个样品中并没有发现电荷有序绝缘体 (COI)和反铁磁绝缘体 (AFI)现象 .Tc最高达 2 75K,MR最大可达 10 4 % (x =0 .2 ,H =1.5T) .在 T >Tp 或 Tc的温区内 ,电阻率ρ∝exp(- E/ k T) ,说明载流子的迁移是以热激活方式进行的 ,热激活能为 0 .32~ 18.3me V.当 T 0 .4 ) .  相似文献

We report a method for measuring shear wave velocity in soft materials using phase stabilized swept source optical coherence tomography (PhS-SSOCT). Wave velocity was measured in phantoms with various concentrations of gelatin and therefore different stiffness. Mechanical waves of small amplitudes (??10 ??m) were induced by applying local mechanical excitation at the surface of the phantom. Using the phase-resolved method for displacement measurement described here, the wave velocity was measured at various spatially distributed points on the surface of the tissue-mimicking gelatin-based phantom. The measurements confirmed an anticipated increase in the shear wave velocity with an increase in the gelatin concentrations. Therefore, by combining the velocity measurements with previously reported measurements of the wave amplitude, viscoelastic mechanical properties of the tissue such as cornea and lens could potentially be measured.  相似文献
