Human pluripotent stem cells (hPSCs), such as embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs), provide a powerful model system for studies of cellular identity and early mammalian development, which hold great promise for regenerative medicine. It is necessary to develop a convenient method to discriminate hPSCs from other cells in clinics and basic research. Herein, a simple and reliable biosensor for stem cell detection was established. In this biosensor system, stage-specific embryonic antigen-3 (SSEA-3) and stage-specific embryonic antigen-4 (SSEA-4) were used to mark human pluripotent stem cells (hPSCs). Antibody specific for SSEA-3 was coated onto magnetic beads for hPSCs enrichment, and antibody specific for SSEA-4 was conjugated with carboxyl-modified tDNA sequence which was used as template for strand displacement amplification (SDA). The amplified single strand DNA (ssDNA) was detected with a lateral flow biosensor (LFB). This biosensor is capable of detecting a minimum of 19 human embryonic stem cells by a strip reader and 100 human embryonic stem cells by the naked eye within 80 min. This approach has also shown excellent specificity to distinguish hPSCs from other types of cells, showing that it is promising for specific and handy detection of human pluripotent stem cells.  相似文献

Efficient DNA nick sealing catalyzed by T4 DNA ligase was carried out on a modified DNA template in which an intercalator such as azobenzene had been introduced. The intercalator was attached to a D-threoninol linker inserted into the DNA backbone. Although the structure of the template at the point of ligation was completely different from that of native DNA, two ODNs could be connected with yields higher than 90% in most cases. A systematic study of sequence dependence demonstrated that the ligation efficiency varied greatly with the base pairs adjacent to the azobenzene moiety. Interestingly, when the introduced azobenzene was photoisomerized to the cis form on subjection to UV light (320-380 nm), the rates of ligation were greatly accelerated for all sequences investigated. These unexpected ligations might provide a new approach for the introduction of functional molecules into long DNA strands in cases in which direct PCR cannot be used because of blockage of DNA synthesis by the introduced functional molecule. The biological significance of this unexpected enzymatic action is also discussed on the basis of kinetic analysis.  相似文献

A new flow injection catalytic spectrophotometric method for on-line preconcentration and determination of total iron in natural water is described. The method is based on a combination of iron-catalyzed oxidation of diaminoditolyl by potassium bromate and the use of on-line preconcentration of iron onto 8-hydroxy-quinoline immobilized on silica gel. The corresponding calibration graph is linear over the range of 2.0–110ngmL–1 for Fe(III) using a time-based technique for 5min preconcentration. The relative standard deviation of 11 measurements of 60ngmL–1 Fe(III) was 0.67%. The method was applied to the determination of iron in natural water. The results obtained by the proposed method were compared with those obtained by ICP-AES. The t-test showed no significant differences between the two methods at a confidence level of 95%.  相似文献

为验证亚毫米波辐射计在弹药末制导中的应用可行性,计算了不同天气条件下天空亮温和视在温度,利用360 GHz辐射计对地面金属目标和涂层隐身目标进行了模拟探测试验。计算和试验表明,在寒冷干燥的季节,辐射计可以探测到地面金属目标,随着温度和湿度的升高,辐射计很难再探测到目标;对于高温涂层隐身目标,辐射计可以探测到,而不受天气的限制。  相似文献

Comparative studies of lipase-catalyzed hydrolysis of soy oil in solvent-free system were carried out in shaking bath and in ultrasonic bath. A suitable ultrasonic power of 1.64 W cm(-2) was determined to guarantee satisfactory hydrolysis extent and lipase activity. The influence of temperature, pH, enzyme concentration and water/oil ratio was investigated subsequently. Compared with that in shaking bath, optimum temperature and inactivation temperature of lipase in ultrasonic bath were about 5-10 degrees C higher, while pH effect in ultrasonic bath was similar; ultrasound also led to a smooth increase of reaction rate at relatively higher enzyme loading and less use of water to saturate hydrolysis substrate. In optimum conditions, the overall hydrolysis reaction rate in the ultrasonic bath process was above 2-fold than that in the shaking bath process.  相似文献

分别以绕丹宁和噻唑烷-2,4-二酮单元为端基、IDT为中心核设计合成了一个新型不对称结构的有机小分子受体IDT-2,并通过与两端均以绕丹宁或噻唑烷-2,4-二酮受体单元的对称小分子受体IDT-1和IDT-3进行对比,探讨了分子结构与性能之间的关系。研究发现,从IDT-1到IDT-3,随着两端的绕丹宁基团被噻唑烷-2,4-二酮基团逐步取代,这类小分子受体的吸收光谱显著蓝移,光学带隙E_g~(opt)逐步增大,LUMO和HOMO能级也逐渐抬升。随后我们分别以这三个小分子为受体、P3HT为给体共混构建活性层而制备了有机太阳能电池,结果表明,以两端均为绕丹宁单元的对称结构小分子受体IDT-1构建的电池器件具有最高的光电转换效率(PCE),相应的J_(sc)和FF值也最大,而V_(oc)则最低;而以两端均为噻唑烷-2,4-二酮基团的对称结构小分子受体IDT-3的电池器件,其V_(oc)最高,但其J_(sc)和FF则最低,PCE值也最小。对于IDT-2而言,由于分子只有一个绕丹宁单元被噻唑烷-2,4-二酮所取代,其V_(oc),J_(sc)和PCE均介于IDT-1与IDT-3之间。由此说明,尽管噻唑烷-2,4-二酮基团的引入能有效提升器件V_(oc),但却不利于改善其J_(sc)和FF,因此受体的分子设计中如何平衡电池器件的几种光伏性能参数而获得高的光电转换效率仍是十分重要的研究课题之一。  相似文献

In this paper we reported that low temperature plasma may reverse the direction of a chemical reaction. The thermodynamically forbidden reaction between H 2 and AlCl 3 was able to take place with the assistance of low temperature plasma, yielding metal Al. The plasma chemistry of the reaction was investigated by optical emission spectroscopy, which suggested that the dissociation of H 2 and AlCl 3 molecules by plasma led the reaction to a thermodynamically favorable one by creating reaction channels with low Gibbs free energy change. The addition of Ar promoted the reaction kinetics dramatically, which was attributed to the enhanced dissociation of AlCl 3 molecules by excited Ar species.  相似文献

A new photoswitch for DNA hybridization involving para‐substituted azobenzenes (such as isopropyl‐ or tert‐butyl‐substituted derivatives) with L ‐threoninol as a linker was synthesized. Irradiation of the modified DNA with visible light led to dissociation of the duplex owing to the destabilization effect of the bulky substituent on the trans‐azobenzene. In contrast, trans‐to‐cis isomerization (UV light irradiation) facilitated duplex formation. The direction of this photoswitching mode was entirely reversed relative to the previous system with an unmodified azobenzene on D ‐threoninol whose trans form turned on the hybridization, and cis form turned it off. Such reversed and reversible photoswitching of DNA hybridization was directly demonstrated by using fluorophore‐ and quencher‐attached oligonucleotides. Furthermore, it was revealed that the cis‐to‐trans thermal isomerization was greatly suppressed in the presence of the complementary strand owing to the formation of the more‐stable duplex in the cis form.  相似文献

Metal-organic framework materials are a class of novel crystalline porous materials with regular pore structures formed by covalent bonding between metal centers and organic functional groups. Metal-organic framework materials have attracted great interest in analytical chemistry due to their unique properties such as good stability and permanent porosity. In this work, D-histidine was used to carry out chiral modification of zeolitic imidazolate framework-90 under mild conditions, and the D-histidine modified zeolitic imidazolate framework-90 coated capillary column was prepared. This chiral capillary column was used to separate epinephrine, norepinephrine, terbutaline, and tryptophan enantiomers. Under optimum conditions, baseline separations were achieved. The intra-day, inter-day, and inter-column relative standard deviations (n = 3) of the four pairs of enantiomeric migration times were 0.15%–0.56%, 0.74%–2.40%, and 1.93%–3.18%, respectively. Moreover, the D-histidine modified zeolitic imidazolate framework-90 coated capillary could be reused for at least 150 runs without significant changes in the separation efficiency and migration time.  相似文献
