Prompted by extensive theoretical interest in the role of tunneling in the intramolecular 1,5-hydrogen shift in 1,3(Z)-pentadienes and the large uncertainty in the published values of the theoretically relevant kinetic deuterium-isotope effect and its dependence on temperature, we have examined a degenerate bicyclic version, 2-methyl-10-methylenebicyclo[4.4.0]dec-1-ene, which is locked into the rearrangement-competent cisoid conformation, in the hope of obtaining more precise and accurate values. From rate constants determined over a range of 33 degrees C from 167.7 to 201.6 degrees C, Arrhenius parameters, Ea = 32.8 +/- 0.4 kcal mol(-1) and log A = 11.1 +/- 0.2, were obtained. An average kinetic isotope effect of 4.2 +/- 0.5 obtained from all values for kH/kD and k-H/k-D may be compared with a value of 5.0 +/- 0.3, recalculated from data in the pioneering publication of Roth and K?nig. From a highly problematic extrapolation of the temperature dependence, a value of kH/kD of 16.6 (standard error between 6.5 and 43) is calculated for the kinetic isotope effect at 25 degrees C (Roth and K?nig: 12.2). With curvature in Arrhenius plots being one of the three types of experimental evidence considered indicative of tunneling, the kinetic study of the previously published rearrangement of 1-phenyl-5-p-tolyl-1,3(Z)-pentadiene has been extended over a period of 339 days to a range of 108 degrees C (77-185 degrees C) without discerning any deviation from a straight-line Arrhenius plot: Ea = 28.7 +/- 0.5 (kcal mol(-1)) and log A = 9.41 +/- 0.30.  相似文献

We report a method of coupled CE-LIF detection with flow cytometry for high-throughput determination and quantitation of fluorophores in single intact K562/S (KS) cells. The membrane properties of KS cell including fluophore transport rate and apparent permeability coefficient were further quantitatively characterized. The method has advantages for accurate quantitation and unique capacity of high-throughput analysis. The strategy will be useful for the quantitation of fluorophores in the intact cells, such as measurement of multidrug resistance, quantitation of specific protein expression, and quantitative characterization of protein and enzyme functions.  相似文献

Poly-3-hydroxybutyrate (PHB) can be produced by various species of bacteria. Among the possible carbon sources, both methane and methanol could be a suitable substrate for the production of PHB. Methane is cheap and plentiful not only as natural gas, but also as biogas. Methanol can also maintain methanotrophic activity in some conditions. The methanotrophic strain Methylosinus trichosporium IMV3011 can accumulate PHB with methane and methanol in a brief nonsterile process. Liquid methanol (0.1%) was added to improve the oxidization of methane. The studies were carried out using shake flasks. Cultivation was performed in two stages: a continuous growth phase and a PHB accumulation phase under the conditions short of essential nutrients (ammonium, nitrate, phosphorus, copper, iron (Ⅲ), magnesium or ethylenediamine tetraacetate (EDTA)) in batch culture. It was found that the most suitable growth time for the cell is 144 h. Then an optimized culture condition for second stage was determined, in which the PHB concentration could be much increased to 0.6 g/L. In order to increase PHB content, citric acid was added as an inhibitor of tricarboxylic acid cycle (TCA). It was found that citric acid is favorable for the PHB accumulation, and the PHB yield was increased to 40% (w/w) from the initial yield of 12% (w/w) after nutrient deficiency cultivation. The PHB produced is of very high quality with molecular weight up to 1.5 × 10^6Da.  相似文献

巯基聚合物利用二硫键的形成, 以共价键黏附于黏膜表面, 可以延长药物在黏膜上的滞留时间, 有利于药物分子的吸收. 本文合成了一种新型巯基聚合物——N-乙酰基-L-半胱氨酸-g-壳聚糖(CS-NAC), 并进行了表征, 同时对材料的溶胀度、黏附性和细胞毒性进行了测试. 结果表明, 这种巯基聚合物具有较高的巯基含量, 最高可达到589.3 μmol/g; 具有快速的溶胀性能; 黏附性显著增强, 黏附时间和黏附力分别是壳聚糖的30和3倍; 无明显的细胞毒性. 因此, CS-NAC是一种很有应用前景的生物黏附性材料.  相似文献

以SiO2表面的富水吸附层为纳米反应器制备CuO纳米粒子. 不同水量下的吸附量测定、溶剂置换和反应后吸附对比实验结果表明, SiO2表面存在吸附水层, 并且该吸附水层是反应的主要场所. 根据吸附数据, 认为Cu2+在体系中具有三区域浓度分布特点, 体系中水浓度和氢氧化钠浓度的增加均会使得Cu2+向吸附层区域迁移. XRD分析结果表明, 反应温度的升高和NaOH浓度的增大有利于生成更小的CuO晶粒, 其中温度的升高使吸附层厚度减小, 限制了晶粒生长; NaOH浓度的增大则会增大层内溶质过饱和度, 导致晶核形成速率加快.  相似文献

An improved method for preparing melamine cyanurate (MCA) based flame retardant polyamide 6 (FRPA6) materials has been proposed. This processing method, i.e., improved in situ polymerization, was used to synthesize flame retardant PA6. In situ formed MCA nanoparticles were supposed to be linked to PA6 chains in the ε-caprolactam hydrolytic polymerization system to obtain startype polymers for the first time. Through TEM photographs, it can be found that the in situ formed MCA nanoparticles with diametric size of less than 50 nm, are nanoscaled, highly uniformly dispersed in the PA6 matrix. Synthesized flame retardant PA6 have good fire performance which can achieve UL-94 V-0 rating at 1.6 mm thickness with the presence of 7.34 wt.% MCA in the matrix.  相似文献

采用超滤浓缩、强阴离子交换、疏水作用和凝胶色谱等方法, 对毕赤酵母表达的rGlip进行分离和纯化, 对离子交换色谱中rGlip与固相结合的最佳pH值进行了考察, 并对纯化产物的活性进行了鉴定. rGlip在215 nm处有强的紫外吸收, 经激光解析电离时间飞行质谱鉴定其相对分子量为12722, 经反相液相色谱鉴定纯度≥97%. 设计rGlip的疏水作用色谱, 有效地去除色素. 凝血实验结果表明, rGlip可以凝集绵羊血红细胞, 但对人血A, B, AB和O型等红细胞无凝集作用, 有类似凝集素的生物学活性.  相似文献

A new method of extracting essential oils from dried plant materials has been studied. By adding a microwave-absorption medium (MAM) to a reactor, solvent-free microwave extraction (SFME) was improved and can be used to extract essential oils from dried plant material without pretreatment. With a microwave irradiation power of 85 W it took only approximately 30 min to extract the essential oils completely. The whole extraction process is simple, rapid, and economical. Three types of MAM, iron carbonyl powder (ICP), graphite powder (GP), and activated carbon powder (ACP), and two types of dried plant material, Illicium verum Hook. f. and Zingiber officinale Rosc., were studied. The results were compared with those obtained by use of conventional SFME, microwave-assisted hydrodistillation (MAHD), and conventional hydrodistillation (HD), and the conclusion drawn was that improved SFME was a feasible means of extracting essential oils from dried plant materials, because there were few differences between the composition of the essential oils extracted by improved SFME and by the other methods.  相似文献
