[reaction: see text] Ester-containing nitrones, including 5-tert-butoxycarbonyl-5-methyl-1-pyrroline N-oxide 5, have been reported to be robust spin traps for superoxide (O2*-). Using a chiral column, we have been able to isolate the two enantiomers of nitrone 5. With enantiomerically pure nitrone 5a and 5b we explored whether one of these isomers was solely responsible for the EPR spectrum of aminoxyl 6. Data obtained demonstrate that the spin trapping of O2*- by nitrone 5a and nitrone 5b affords the identical EPR spectra and lifetimes in homogeneous aqueous solution and exhibits the same ratio of cis and trans isomers. Quantum chemical modeling in vacuo also finds no difference, aside from the expected optical activity, arising from the difference in stereochemistry. 相似文献

In early pregnancy, the placenta anchors the conceptus and supports embryonic development and survival. This study aimed to investigate the underlying functions of Shh signaling in recurrent miscarriage (RM), a serious disorder of pregnancy. In the present study, Shh and Gli2 were mainly observed in cytotrophoblasts (CTBs), Ptch was mainly observed in syncytiotrophoblasts (STBs), and Smo and Gli3 were expressed in both CTBs and STBs. Shh signaling was significantly impaired in human placenta tissue from recurrent miscarriage patients compared to that of gestational age-matched normal controls. VEGF-A and CD31 protein levels were also significantly decreased in recurrent miscarriage patients. Furthermore, inhibition of Shh signaling impaired the motility of JAR cells by regulating the expression of Gli2 and Gli3. Intriguingly, inhibition of Shh signaling also triggered autophagy and autolysosome accumulation. Additionally, knockdown of BECN1 reversed Gant61-induced motility inhibition. In conclusion, our results showed that dysfunction of Shh signaling activated autophagy to inhibit trophoblast motility, which suggests the Shh pathway and autophagy as potential targets for RM therapy.Subject terms: Embryology, Endocrine reproductive disorders, Autophagosomes 相似文献

The design of high-efficiency CO2 adsorbents with low cost, high capacity, and easy desorption is of high significance for reducing carbon emissions, which yet remains a great challenge. This work proposes a facile construction strategy of amino-functional dynamic covalent materials for effective CO2 capture from flue gas. Upon the dynamic imine assembly of N-site rich motif and aldehyde-based spacers, nanospheres and hollow nanotubes with spongy pores were constructed spontaneously at room temperature. A commercial amino-functional molecule tetraethylenepentamine could be facilely introduced into the dynamic covalent materials by virtue of the dynamic nature of imine assembly, thus inducing a high CO2 capacity (1.27 mmol·g−1) from simulated flue gas at 75 °C. This dynamic imine assembly strategy endowed the dynamic covalent materials with facile preparation, low cost, excellent CO2 capacity, and outstanding cyclic stability, providing a mild and controllable approach for the development of competitive CO2 adsorbents. 相似文献

To extend the application range of L-asparaginase in food pre-processing, the thermostability improvement of the enzyme is essential. Herein, two non-conserved cysteine residues with easily oxidized free sulfhydryl groups, Cys8 and Cys283, of Acinetobacter soli L-asparaginase (AsA) were screened out via consensus design. After saturation mutagenesis and combinatorial mutation, the mutant C8Y/C283Q with highly improved thermostability was obtained with a half-life of 361.6 min at 40 °C, an over 34-fold increase compared with that of the wild-type. Its melting temperature (Tm) value reaches 62.3 °C, which is 7.1 °C higher than that of the wild-type. Molecular dynamics simulation and structure analysis revealed the formation of new hydrogen bonds of Gln283 and the aromatic interaction of Tyr8 formed with adjacent residues, resulting in enhanced thermostability. The improvement in the thermostability of L-asparaginase could efficiently enhance its effect on acrylamide inhibition; the contents of acrylamide in potato chips were efficiently reduced by 86.50% after a mutant C8Y/C283Q treatment, which was significantly higher than the 59.05% reduction after the AsA wild-type treatment. In addition, the investigation of the mechanism behind the enhanced thermostability of AsA could further direct the modification of L-asparaginases for expanding their clinical and industrial applications. 相似文献

Yttrium-90 (90Y) microspheres are widely used for the treatment of liver-dominant malignant tumors. They are infused via catheter into the hepatic artery branches supplying the tumor under fluoroscopic guidance based on pre-therapy angiography and Technetium-99m macroaggregated albumin (99mTc-MAA) planning. However, at present, these microspheres are suspended in radiolucent media such as dextrose 5% (D5) solution. In order to monitor the real-time implantation of the microspheres into the tumor, the 90Y microspheres could be suspended in omnipaque contrast for allowing visualization of the correct distribution of the microspheres into the tumor. The radiochemical purity of mixing 90Y-microspheres in various concentrations of omnipaque was investigated. The radiochemical purity and feasibility of mixing 99mTc-MAA with various concentrations of a standard contrast agent were also investigated. Results showed the radiochemical feasibility of mixing 90Y-microspheres with omnipaque is radiochemically acceptable for allowing real-time visualization of radioembolization under fluoroscopy. 相似文献

Two-dimensional (2D) transition metal dichalcogenide nanosheets (TMDC NSs) have attracted growing interest due to their unique structure and properties. Although various methods have been developed to prepare TMDC NSs, there is still a great need for a novel strategy combining simplicity, generality, and high efficiency. In this study, we developed a novel polymer-assisted ball milling method for the efficient preparation of TMDC NSs with small sizes. The use of polymers can enhance the interaction of milling balls and TMDC materials, facilitate the exfoliation process, and prevent the exfoliated nanosheets from aggregating. The WSe2 NSs prepared by carboxymethyl cellulose sodium (CMC)-assisted ball milling have small lateral sizes (8~40 nm) with a high yield (~60%). The influence of the experimental conditions (polymer, milling time, and rotation speed) on the size and yield of the nanosheets was studied. Moreover, the present approach is also effective in producing other TMDC NSs, such as MoS2, WS2, and MoSe2. This study demonstrates that polymer-assisted ball milling is a simple, general, and effective method for the preparation of small-sized TMDC NSs. 相似文献

The shuttle effect is understood to be the most significant issue that needs to be solved to improve the performance of lithium–sulfur batteries. In this study, ultrathin two-dimensional Fe–Co bimetallic oxide nanosheets were prepared using graphene as a template, which could rapidly catalyze the conversion of polysulfides and inhibit the shuttle effect. Additionally, such ultrathin nanostructures based on graphene provided sufficient active sites and fast diffusion pathways for lithium ions. Taking into account the aforementioned benefits, the ultrathin two-dimensional Fe–Co bimetallic oxide nanosheets modified separator assembled lithium–sulfur batteries delivered an incredible capacity of 1044.2 mAh g−1 at 1 C and retained an excellent reversible capacity of 859.4 mAh g−1 after 100 cycles. This work was focused on the rust conversion of proanthocyanidins (PC) for goethite (α-FeOOH), akaganeite (β-FeOOH) and lepidocrocite (γ-FeOOH), trying to show the potential of PC as an eco-friendly corrosion inhibitor and rust converter for archaeological steel conservation. The experiment used a rusted steel screw from Lingzhao Xuan of the Forbidden City in the Qing Dynasty and three kinds of pure iron oxyhydroxides as research samples. By means of micro-Raman, FTIR, XRD, XPS, SEM and EIS, PC had the ability to chemically react with iron oxyhydroxides in the rust, forming amorphous PC-FeOOH with a marked signal about 1384 cm−1 as phenolic-Fe in infrared properties. The original relatively stable iron oxides were not induced to phase transformation and still remained. The converted rust layer could be more stable in the corrosive medium and increased the corrosion potential more effectively. Both the rust layer resistance and the charge transfer resistance of the archaeological samples were improved by at least 3 times with 5.0 g/L of PC, which could reasonably stabilize the archaeological rust and hindered external corrosive penetration into the core. It was a mild protection material that showed satisfactory performance for archaeological steel cultural heritage and has a good application prospect. 相似文献

Dehydrocostus lactone (DL) is among the representative ingredients of traditional Chinese medicine (TCM), with excellent anticancer, antibacterial, and anti-inflammatory activities. In this study, an advanced strategy based on ultra-high-performance liquid chromatography–quadrupole-Orbitrap high-resolution mass spectrometry (UHPLC–Q-Orbitrap HRMS) was integrated to comprehensively explore the metabolic fate of DL in rats. First, prior to data collection, all biological samples (plasma, urine, and feces) were concentrated and purified using solid-phase extraction (SPE) pre-treatment technology. Then, during data collection, in the full-scan (FS) data-dependent acquisition mode, FS-ddMS2 was intelligently combined with FS-parent ion list (PIL)-dynamic exclusion (DE) means for targeted monitoring and deeper capture of more low-abundance ions of interest. After data acquisition, data-mining techniques such as high-resolution extracted ion chromatograms (HREICs), multiple mass defect filters (MMDFs), diagnostic product ions (DPIs), and neutral loss fragments (NLFs) were incorporated to extensively screen and profile all the metabolites in multiple dimensions. As a result, a total of 71 metabolites of DL (parent drug included) were positively or tentatively identified. The results suggested that DL in vivo mainly underwent hydration, hydroxylation, dihydrodiolation, sulfonation, methylation, dehydrogenation, dehydration, N-acetylcysteine conjugation, cysteine conjugation, glutathione conjugation, glycine conjugation, taurine conjugation, etc. With these inferences, we successfully mapped the “stepwise radiation” metabolic network of DL in rats, where several drug metabolism clusters (DMCs) were discovered. In conclusion, not only did we provide a refined strategy for inhibiting matrix effects and fully screening major-to-trace metabolites, but also give substantial data reference for mechanism investigation, in vivo distribution visualization, and safety evaluation of DL. 相似文献
