The reproducibility, the small scale as well as the large scale variability of137Cs extracted sequentially from the soil by using a modified Tessier procedure was investigated at several grassland sites in Bavaria/Germany and in the Chemobyl area. Because undisturbed grassland soils are never homogeneous with respect to their soil properties, all sequential extractions at the German sites were carried out at each plot separately for different soil layers (e.g., 0–2, 2–5, 5–10, 10–15, 15–20 and 20–30 cm). The results show that the coefficients of variation (CV) for the reproducibility of the extraction procedure for137Cs was (with some exceptions) around 10–20% for all fractions. For the small scale variability of137Cs (samples within an area of 10×10m2) the values for theCV were (again with a few exceptions) in the same range. Compared to that, the large scale variability of extractable137Cs (random soil samples within an area of 100×200 km2) was higher for all fractions, even though only moderately. The implications of these results with respect to a sampling design are discussed.  相似文献

Heme oxygenase (HO) catalyzes the oxygen-dependent degradation of heme to biliverdinIXalpha, CO, and free iron ion via three sequential monooxygenase reactions. Although the distinct active-site structure of HO from cytochrome P450 families suggests unique distal protein machinery to activate molecular oxygen, the mechanism and the key amino acid for the oxygen activation have not been clear. To investigate the functionality of highly conserved polar amino acids in the distal helix of HO-1, we have prepared alanine mutants: T135A, R136A, D140A, and S142A, and found drastic changes in the heme degradation reactions of D140A. In this paper, we report the first evidence that D140 is involved in the oxygen activation mechanism in HO-1. The heme complexes of HO mutants examined in this study fold and bind heme normally. The pK(a) values of the iron-bound water and autoxidation rates of the oxy-form are increased with R136A, D140A, and S142A mutations, but are not changed with T135A mutation. As the wild-type, T135A, R136A, and S142A degrade heme to verdohemeIXalpha with H(2)O(2) and to biliverdinIXalpha with the NADPH reductase system. On the other hand, D140A heme complex forms compound II with H(2)O(2), and no heme degradation occurs. For the NADPH reductase system, the oxy-form of D140A heme complex is accumulated in the reaction, and only 50% of heme is degraded. The stopped flow experiments suggest that D140A cannot activate iron-bound dioxygen and hydroperoxide properly. To investigate the carboxylate functionality of D140, we further replaced D140 with glutamic acid (D140E), phenylalanine (D140F), and asparagine (D140N). D140E degrades heme normally, but D140N shows reactivity similar to that of D140A. D140F loses heme degradation activity completely. All of these results indicate that the carboxylate at position 140 is essential to activate the iron-bound dioxygen and hydroperoxide. On the basis of the present findings, we propose an oxygen activation mechanism involving the hydrogen-bonding network through the bridging water and D140 side chain.  相似文献

Porphyrins react readily with organolithium reagents under substitution of free meso positions. As this method has proven to be very versatile for the preparation of a wide range of meso substituted porphyrins, a mechanistic study of the reaction was undertaken using 5,15-diaryl- and dialkyl substituted porphyrins, 2,3,7,8,12,13,17,18-octaethylporphyrin, and the respective nickel(II) complexes. A combination of deuteration experiments, electronic absorption spectroscopy of the reactive intermediates, trapping of intermediates with organic electrophiles, and reaction at different pH values showed significant differences in the reaction pathways of free base porphyrins and metalloporphyrins. In both cases the reaction proceeds initially under formation of phlorin like intermediates which are stable in water. For the Ni(II)phlorins a mesomeric carbanionic form with a highly distorted structure exists that can react as a nucleophile with electrophiles such as RI, H+, or D+. In the latter case a protonation-deprotonation equilibrium involving porphodimethen intermediates has to be assumed. Free base phlorins do not react as nucleophiles but can undergo H/D exchange reactions in strongly acidic media.  相似文献

Volatile Au species in an acidified medium were generated at room temperature by reduction with NaBH4 in acidified aqueous medium using a flow-injection chemical vapor-generation atomic absorption spectrometric (FI-CVG-AAS) system in the presence of micro amounts of sodium diethyldithiocarbamate (DDTC). Precision of 2.0% RSD (n = 11, 2.0 mg L(-1) level) was obtained at a sample throughput of 180 h(-1). A detection limit of 24 ng mL(-1) (3sigma) was obtained with 300 microL sample solution. The method was used for the determination of gold in ore sample digests, and the results obtained agreed well with those obtained by flame AAS.  相似文献

The power-time curves of the growth of three strains of petroleum bacteria at different temperatures have been determined. A novel equation of a power-time curve has been proposed in this paper. The general formula to calculate the rate constant of the bacterial growth has been derived. The rate constants of the bacterial growth at different temperatures, the heat production per newly formed bacterium, the bacterial number at the end of the bacterial growth and the deceleration rate constant of the bacterial growth at 50.00°C, have been calculated. The optimum growth temperatures of the three strains have been obtained.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献
