Microfluidic trapping methods create significant opportunities to establish highly controlled cell positioning and arrangement for the microscale study of numerous cellular physiological and pathological activities. However, a simple, straightforward, dynamic, and high-throughput method for cell trapping is not yet well established. In the present paper, we report a direct active trapping method using an integrated microfluidic device with pneumatic microstructures (PμSs) for both operationally and quantitatively dynamic localization of cells, as well as for high-throughput cell patterning. We designed and fabricated U-shape PμS arrays to replace the conventional fixed microstructures for reversible trapping. Multidimensional dynamics and spatial consistency of the PμSs were optically characterized and quantitatively demonstrated. Furthermore, we performed a systematic trapping investigation of the PμSs actuated at a pressure range of 0 psi to 20 psi using three types of popularly applied mammalian cells, namely, human lung adenocarcinoma A549 cells, human hepatocellular liver carcinoma HepG2 cells, and human breast adenocarcinoma MCF-7 cells. The cells were quantitatively trapped and controlled by the U-shape PμSs in a programmatic and parallel manner, and could be opportunely released. The trapped cells with high viability were hydrodynamically protected by the real-time actuation of specifically designed umbrella-like PμSs. We demonstrate that PμSs can be applied as an active microfluidic component for large-scale cell patterning and manipulation, which could be useful in many cell-based tissue organization, immunosensor, and high-throughput imaging and screening.  相似文献

The fatty acyl residues composition and molecular species of phosphatidylcholines (PCs) in soy lecithin powder were studied and characterized using ultra-performance liquid chromatography-electrospray tandem mass spectrometry (UPLC–ESI-MS/MS) via a two-step procedure. ESI-MS/MS by precursor ion scan (PIS) m/z 184.1 in positive ion mode first detected isobaric PCs, and further identified positional isomers and fatty acyl residue composition in negative ion mode. The results showed that the fatty acyl residue composition was in strong agreement with that detected by the classical analysis method. Likewise, ten peaks of isobaric PCs were obtained by ESI-MS/MS analysis in positive ion mode, and 20 positional isomers of PCs were characterized in negative ion mode. Thus, this study presents a simple and powerful way to analyze fatty acyl residue composition and molecular species of PCs without destroying chemical structure.

To discuss the protection mechanism of DNA from radiation as well as assess the performance of PM6-DH2 on noncovalent interactions, the interaction of four nucleic acid bases (NABs) such as adenine (A), cytosine (C), guanine (G), and thymine (T), with Li@C(60) was extensively investigated with the-state-of-art theoretical methods describing noncovalent systems, like M06-2x, PBE-D, and PM6-DH2 methods. In the gas phase, the binding strength of NABs to Li@C(60) from M06-2x decreases in the sequence, G>C>A>T. As dispersion was explicitly included, PBE-D relatively enhances the binding of A and T and corrects the sequence to, G>A>C~T. PM6-DH2 predicted similar binding energies to those from PBE-D within 0.5 kcal/mol and the same binding sequence, suggesting that the PM6-DH2 method is promising for nano-scale systems. In the aqueous solution, binding of NABs-Li@C(60) is considerably decreased, and the M06-2X and PM6-D methods yield a different sequence from the gas phase, G>A>T>C. The encapsulation of Li atom results in a lower IP for Li@C(60) than those of NABs, and the dominant localization of single-occupied molecular orbital on Li@C(60) moiety of the complexes NABs-Li@C(60) further indicates that an electron would be ejected from Li@C(60) upon radiation and Li@C(60) is therefore able to protect DNA bases from radiation. In addition, it was revealed that Li prefers coordination with the hexagonal ring at Li@C(60) , which clarifies the existing controversy in this respect. Finally, Yang's reduced density gradient approach clearly shows that the weak and strong noncovalent interaction regions in the complexes, NABs-Li@C(60) and (NABs-Li@C(60) )(+).  相似文献

从10L连续搅拌式罐式反应器(CSTR)中分离得到1株嗜温高效产氢菌株08-1.根据菌株的形态特征和16S rDNA序列结果分析,初步鉴定菌株08-1属于Clostridium sp..同时还进一步研究了温度、pH值控制、底物浓度和种类对菌株08-1产氢的影响.结果表明.该菌株更适合利用蔗糖或成分复杂的生物质木薯粉以及废弃物厨余垃圾生长及产氢,最适产氢温度为40℃,产氢系统pH值控制在5.5时获得最大产氢量.在间歇发酵中,蔗糖浓度为20 g/L,控制温度40℃,pH值5.5,搅拌速度100 r/min时实现最大产氢速率为245 mL·(L·h)~(-1),最大产氢量达到3.06 mol.该菌株在生物制氢中具有潜在的应用价值.  相似文献

As one of the most abundant renewable resources, rice straw is an attractive lignocellulosic material for animal feeding or for the production of biochemical. An appropriate pre-treatment technique is essential for converting rice straw to rich fodder or biofuel. Based on previous work, Coprinopsis cinerea can grow on rice straw medium and therefore it is useful for the treatment of rice straw. However, little is known regarding its degradation systems and nutrition values. In this study, we firstly found that C. cinerea could grow rapidly on rice straw without any additives by the production of a series of enzymes (laccase, cellulase, and xylanase) and that the microstructure and contents of rice straw changed significantly after being treated by C. cinerea. We propose that a possible underlying mechanism exists in the degradation. Moreover, C. cinerea has a high nutrition value (23.5% crude protein and 22.2% total amino acids). Hence, fermented rice straw with mycelium could be a good animal feedstuff resource instead of expensive forage. The direct usage of C. cinerea treatment is expected to be a practical, cost-effective, and environmental-friendly approach for enhancing the nutritive value and digestibility of rice straw.  相似文献
