Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.Subject terms: Diagnostic markers, Non-coding RNAs  相似文献

以85%的食品级磷酸为原料,三聚氰胺为缩合剂,合成了一种具有网状结构的结晶V型聚磷酸铵,对其水溶性、XRD、13C NMR、红外光谱、热重等进行了测试与表征,并与结晶I型、结晶Ⅱ型产品进行了对比。结果表明,本实验制备产品具有良好的耐水性, 其XRD图谱与结晶V型完全一致,其13C NMR及红外图谱表明该物质是以三嗪环为基本骨架的聚磷酸铵链交联网状结构。热重分析表明,700℃时该产品残重53.6%,大于结晶I型的40.29%及结晶II型的36.16%,初步显示该产品具有良好的高温热稳定性。  相似文献

Simultaneous bandwidth(BW) enhancement and time-delay signature(TDS) suppression of chaotic lasing over a wide range of parameters by mutually coupled semiconductor lasers(MCSLs) with random optical injection are proposed and numerically investigated. The influences of system parameters on TDS suppression(characterized by autocorrelation function(ACF) and permutation entropy(PE) around characteristic time) and chaos BW are investigated. The results show that, with the increasing bias current, the ranges of parameters(detuning and injection strength) for the larger BW(> 20 GHz) are broadened considerably, while the parameter range for optimized TDS(< 0.1) is not shrunk obviously.Under optimized parameters, the system can simultaneously achieve two chaos outputs with enhanced BW(> 20 GHz)and perfect TDS suppression. In addition, the system can generate two-channel high-speed truly physical random number sequences at 200 Gbits/s for each channel.  相似文献

We report on temperature-programmed growth of graphene islands on Ru(0001) at annealing temperatures of 700°C,800°C,and 900°C.The sizes of the islands each show a nonlinear increase with the annealing temperature.In 700°C and 800°C annealings,the islands have nearly the same sizes and their ascending edges are embedded in the upper steps of the ruthenium substrate,which is in accordance with the etching growth mode.In 900°C annealing,the islands are much larger and of lower quality,which represents the early stage of Smoluchowski ripening.A longer time annealing at 900°C brings the islands to final equilibrium with an ordered moir’e pattern.Our work provides new details about graphene early growth stages that could facilitate the better control of such a growth to obtain graphene with ideal size and high quality.  相似文献

Three new compounds (13) and seven known compounds (410) have been isolated from the ethanolic extract of Viburnum macrocephalum f. keteleeri using bioactivity-guided fractionation and identified as methyl (2-α-L-rhamnopyranosyloxy)acetate (1), methyl (2R-3-α-L-rhamnopyranosyloxy)glycerate (2), methyl (3R-4-α-L-rhamnopyranosyloxy-3-hydroxy)butanoate (3), bridelionoside B (4), (6S,7E,9R)-roseoside (5), linarionoside A (6), 3,7,11-trimethyl-1,6-dodecadien-3,10,11-triol (7), (+)-8-hydroxylinalool (8), β-sitosterol (9) and daucosterol (10). The structures of 13, including absolute configurations, were determined by spectroscopic data (1H and 13C NMR, HSQC, HMBC and ORD) and chemical methods. In addition, compounds 1–8 were assayed for their insecticidal and antimicrobial activities. Compounds 7 and 8 exhibited moderately insecticidal effects against Mythimna separata with LD50 values of 180 and 230 μg g?1, respectively. Compounds 2, 3, 7 and 8 showed varying antimicrobial activities with IC50 values ranging from 125 to 529 μM.  相似文献

完成了医药类院校物理化学实验“根皮苷B-Z振荡电化学指纹图谱”的设计,简述了医药类院校物理化学实验课程的设计思路。建立了教学实验标准操作方法及数据处理方法,实验所得B-Z振荡电化学指纹图谱稳定清晰,重现性好,特异性强;处理得到的特征参数可用于定性、定量分析。并探究了实验温度、根皮苷浓度对B-Z振荡实验的影响,结果表明不同温度下实验数据对Arrhenius方程拟合良好;根皮苷浓度低于0.4 mmol/L时可用本操作进行定量分析,高于0.4 mmol/L时可观察到停振现象。  相似文献

分别在B3LYP/6-31G*和MP2/6-31G*的理论水平下,计算得到硝酸正丙酯、硝酸异丙酯、硝酸异辛酯和二缩三乙二醇二硝酸酯4种硝酸酯的O-NO2键离解能(BDE)。 采用常压DSC和高压DSC实验方法,研究了4种硝酸酯的热分解过程,获得热分解反应动力学参数。 常压下4种物质的热分解反应发生在气相区,当压力增大至2 MPa时,4种物质直接发生液相分解反应。 4种硝酸酯的O-NO2键离解能在很大程度上符合由实验分析得到的活化能,表明4种硝酸酯的热分解反应只是单分子O-NO2键的均裂反应。  相似文献

在N2/H2O混合气流中将硅片上金覆盖的金属铟颗粒加热到800 ℃制备出了不同形貌的In2O3纳米结构, 在距铟源不同距离处依次得到In2O3的八面体、纳米带、锯齿状纳米线和纳米链. 采用拉曼光谱、扫描电镜、X射线衍射和透射电镜对产物进行了表征分析. 结果表明, 八面体、纳米带、锯齿状纳米线和纳米链均为立方相单晶结构的In2O3. 基于气-固和气-液-固生长机理详细分析了八面体、纳米带、锯齿状纳米线和纳米链的生长过程, 提出了不同形貌In2O3纳米结构的生长模式.  相似文献
