A generic high-throughput liquid chromatography (HTLC) tandem mass spectrometry (MS/MS) assay for the determination of compound I in human urine and dialysate (hemodialysis) was developed and validated. By using the HTLC on-line extraction technique, sample pretreatment was not necessary. The sample was directly injected onto a narrow bore large particle size extraction column (50 x 1.0 mm, 60 microm) where the sample matrix was rapidly washed away using a high flow rate (5 mL/min) aqueous mobile phase while analytes were retained. The analytes were subsequently eluted from the extraction column onto an analytical column using an organic-enriched mobile phase prior to mass spectrometric detection. The analytes were then eluted from the analytical column to the mass spectrometer for the determination. The linear dynamic range was 2.0-6000 ng/mL for the urine assay and 0.1-300 ng/mL for the dialysate assay. Intraday accuracy and precision were evaluated by analyzing five replicates of calibration standards at all concentrations used to construct the standard curve. For the urine assay, the precision (RSD%, n=5) ranged from 1.9 to 8.0% and the accuracy ranged from 87.8 to 105.2% of nominal value. For the dialysate assay, the precision (RSD%, n=5) ranged from 1.1 to 10.0% and the accuracy from 94.5 to 105.2% of nominal value. In-source fragmentation of the acyl glucuronide metabolite (compound III) did not interfere with the determination of parent compound I. The developed HTLC/MS/MS methodology was specific for compound I in the presence of compound III. Column life-time is increased and sample analysis time is decreased over traditional reversed-phase methods when direct injection assays for urine and dialysate are coupled with the technology of HTLC.  相似文献

It is known that silicon fullerenes cannot maintain perfect cage structures like carbon fullerenes. Previous density-functional theory calculations have shown that even with encapsulated species, nearly all endohedral silicon fullerenes exhibit highly puckered cage structures in comparison with their carbon counterparts. In this work, we present theoretical evidences that the tetrahedral fullerene cage Si(28) can be fully stabilized by encapsulating a tetrahedral metallic cluster (Al(4) or Ga(4)). To our knowledge, this is the first predicted endohedral silicon fullerene that can retain perfectly the same cage structure (without puckering) as the carbon fullerene counterpart (T(d)-C(28) fullerene). Density-functional theory calculations also suggest that the two endohedral metallosilicon fullerenes T(d)-M(4)@Si(28) (M=Al and Ga) can be chemically stable because both clusters have a large highest occupied molecular orbital-lowest unoccupied molecular orbital energy gap ( approximately 0.9 eV), strong spherical aromaticity (nucleus-independent chemical shift value of -36 and -44), and large binding and embedding energies.  相似文献

The calix[4] open-chain crown ether, 5,11,17,23-tetra-tert-butyl-25,27-di(2-allyloxyethoxyl)-26,28-dihydroxycalix[4]arene was synthesized and used for preparation of solid-phase microextraction (SPME) fibers of enhanced extraction efficiency. The new SPME coating made from calix[4] open-chain crown ether and hydroxyl-terminated silicone oil was developed with the aid of vinyltriethoxylsilane as bridge using sol-gel method and cross-linking technology. The efficiency of the novel fiber in the extraction of polar aromatic and aliphatic compounds, such as phenols, alcohols, and volatile fatty acids, was also investigated. Due to the introduction of the polar open-chain crown ether in calix[4]arene molecules, the calix[4] open-chain crown ether fiber showed much better selectivity and sensitivity to these polar compounds in comparison with calix[4]arene fiber. It also had superior extraction efficiency when compared to commercial poly(dimethylsiloxane)-divinylbenzene and polyacrylate fibers. Parts per billion to parts per trillion level detection limits were achieved for most of the analytes through SPME in conjunction with GC and flame ionization detector. The linear ranges were two to four orders of magnitude, and the RSD values were below 7% for all analytes. The novel fiber was applied to determine volatile alcohols and fatty acids in wine samples. The volatile-free wine prepared in this work was used to assure similar chemical environment for analytes in both calibration solutions and in real wine samples, thus compensating for possible matrix interferences. The established internal standard method using 4-methyl-2-pentanol as internal standard showed satisfactory accuracy and precision.  相似文献

The Schiff base‐containing pendant monoaza crown ether HL1, HL2, HL3 and HL4 have been synthesized by condensation of salicylaldehyde with N‐(4‐aminoaryl) monoaza crown ethers, which were prepared conveniently from 4‐nitro‐N, N‐di(hydroxyethyl) aniline or 4‐nitrobenzyl chloride via cyclization or condensation and reduction. The structures of HL1—HL4 were verified by 1H NMR, IR spectra, MS and elemental analysis. Moreover, the oxygenation constants (KO2) and thermodynamic parameters (δH0 and δS0) of their cobalt(II) complexes were determined in the range of ?5 °C to 25 °C, and the effect of crown ring bonded to a Schiff base on the dioxygen affinities of cobalt(II) complexes was also observed as compared to the uncrowned analogue (CoL).  相似文献

The photostimulated reaction of halonaphthalene with a series of carbanions derived from,propionic acid derivatives in liquid ammonia led to the naphthylation at a-carbon of the carbanions inan isolated yield ranging from 25% to 86%.In all cases,the dehalonaphthalenes were found to,be by-products and the reaction was inhibited by p-dinitrobenzere.An electron transfer from thecarbanion to the halonaphthalene followed by ejection of halogen led to naphtbyl radical,as predictedby comparing the LUMOs of the carbanions and the halonaphthalenes,was involved in the process.Absence of the alkylnaphthalene and 1,2-dinaphthylalkane in the products indicates the smoothelectron transfer between(Naph-Nu)~+ and Naph-X.C-vs.O-naphthylation is fully addressed interms of MNDO calculations and acid-base principle.  相似文献

A new rapid, selective and sensitive on-line microwave flow injection-kinetic method was developed for spectrophotometric determination of micro amounts of Ir(III), based on its catalytic effect on the m-acetylchlorophosphonazo (CPA-mA) and KIO(4) reaction in NaOH media. An on-line microwave oven was employed to accelerate the reaction. The reaction was followed spectrophotometrically by measuring the decrease of the absorbance of CPA-mA at 580 nm. The effect of five variables for the determination of Ir(III) was optimized by means of a multilayer artificial neural network using extended delta-bar-delta (EDBD) algorithms. Under the optimum experimental conditions, Ir(III) could be determined in the range 0.060-0.60 micro gZZZ;mL(-1) with detection limit of 0.02 micro gZZZ;mL(-1) and the sampling frequency of 34 h(-1). The proposed method was applied to the determination of micro amounts of Ir(III) in refined ore and secondary alloy with the recoveries from 91.4% to 109%.  相似文献

A two-tiered organizing scheme with multiple-length scales for construction of dandelion-like hollow CuO microspheres has been elucidated: (1) mesoscale formation of rhombic building units from smaller nanoribbons via oriented aggregation and (2) macroscopic organization of these units into the CuO microspheres. 系统地研究了Ce(IV), H2SO4, Ru(bipy)3^2^+和5种α-羟基羧酸对Ce(IV)氧化钌(II)-联吡啶化学发光反应速率的影响。结果表明,在选定的试验条件下, 化学发光反应速率分别与Ce(IV),Ru(bipy)3^2^+, 5种α-羟基羧酸的浓度和1/[H2SO4]^2成正比, 据此提出了偶合化学发光反应机理, 推导得到的反应速率方程与实验结果吻合。  相似文献

Five structurally related amino acid derivatives were enantioseparated by HPLC with a commercially available chiral stationary phase, Chiralcel OD-H. The chromatographic experiments were performed in the normal phase mode. n-Hexane/polar alcohol was used as mobile phase. Excellent baseline enantioseparations could be obtained for all these solutes. The effects of the concentration of polar alcohol and the column temperature on the retentions and enantioseparations were studied in detail. From the van't Hoff plots the corresponding apparent thermodynamic parameters were derived. Mechanism aspects of chiral recognition were discussed based on the relationship between the thermodynamic parameters and the structures of the solutes. It was found that the substituent of the phenyl group on the residual group of the amino acid derivatives was close relevant to thermodynamic origin of enantioseparation. Much better enthalpy–entropy compensation effect was obtained by plotting the differential, rather than the original, thermodynamic parameters.  相似文献
