Solid-Phase Synthesis of a Monofunctional trans-a(2)Pt(II) Complex Tethered to a Single-Stranded Oligonucleotide

Cross-linking ability is possible with the oligonucleotide-tethered, monofunctional trans-Pt(II) complex shown. It was synthesized by a novel solid-phase approach comprising conjugation of immobilized tetrathymidylic acid with a trans-a(2)Pt(II) building unit, ammonolysis, and transformation of the resulting complex (R=1-N-cyclohexylmethylthyminate) into the chloro derivative (R=Cl). a=NH(2)CH(3), T=thymine.     

Surface-strain measurements on a hemispherical shell using holographic interferometry

A method is presented for using real-time fringe counting and double-exposure single-hologram interferometry to measure surface strain on pressurized hemispherical shells. An aluminum shell is employed for experimental verification of these techniques. Surface-strain variations caused by a defect in the aluminum wall are detected and measured. Strain-gage arrays are used for comparison of experimental results. Bendix Kansas City Division is a prime contractor to the Atomic Energy Commission.     

Thermodynamic study of a liquid crystal as a liquid phase in gas-liquid chromatography. II. A cholesteric liquid crystal.

Gas-liquid chromatography is utilized for the determination of thermodynamic solution parameters for various organic solutes at infinite dilution in the meso- and isotropic phases of cholesteryl palmitate. The thermodynamic data and trends in values of the activity coefficients for the solutes are discussed in relation to their structure and to the orientations of the liquid crystal.     

Predicting turbulent flow in a staggered tube bundle

This paper presents the results of calculations performed for the turbulent, incompressible flow around a staggered array of tubes for which carefully obtained experimental results are available as part of an established ERCOFTAC-IAHR test case. The Reynolds-averaged Navier–Stokes equations are solved using a pressure-based finite volume algorithm, using collocated cell vertex store on an unstructured and adaptive mesh of tetrahedra. Turbulence closure is obtained with a truncated form of a low-Reynolds number k– model developed by Yang and Shih. The computational domain covers all seven rows of tubes used in the experimental study and periodic flow is allowed to develop naturally. The results of the computations are surprisingly good and compare favourably with results obtained by others using a wide range of alternative k– models for a single cylinder with periodic inflow and outflow boundaries on structured meshes.     

Stachyonic Acid: A Dengue Virus Inhibitor from Basilicum polystachyon

Stachyonic acid A, arising from the first in-depth phytochemical investigation of the herb Basilicum polystachyon, was found to display potent inhibitory activity against dengue virus, with limited cytotoxicity. Andrographolide, a known dengue virus inhibitor and closely related labdane-type diterpene, is structurally more complex but displayed poor antiviral activity in the PRNT assay, and increased cytotoxicity in comparison. Furthermore, a Diels–Alder reaction with PTAD identified the active pharmacophore of stachyonic acid to be the conjugated diene.     

Thermally reversible,covalently crosslinked polyphosphazenes

The concept of thermally reversible crosslinking was investigated by using poly[bis(trifluoroethoxy)phosphazenes] with different percentages of cyclopentadienylethoxy and dicyclopentadienyldiethoxy substituent groups. The resultant structures of these polymers are discussed and the phenomenon of reversible crosslinking is demonstrated by means of a DSC study and solubility behavior.     

Radical cyclization of sugar-derived beta-(alkynyloxy)acrylates: synthesis of novel fused ethers

[formula: see text] Tributyltin radical mediated cyclization of carbohydrate-derived beta-(alkynyloxy)acrylates leading to highly functionalized cis- and trans-fused bicyclic ethers of various ring sizes is described. The efficacy of the radical cyclization is nicely illustrated in the iterative construction of a trans-fused tricyclic tetrahydropyran.     

Influences of non-selective interactions of nucleic acids on response rates of nucleic acid fiber optic biosensors

The immobilization of oligonucleotides to solid surfaces can provide a platform of chemistry that is suitable for the development of biosensor and microarray technologies. Experiments were performed using a fiber optic nucleic acid biosensor based on total internal reflection fluorescence to examine the effects of the presence of non-complementary DNA on the detection of hybridization of complementary target DNA. The work has focused on the rates and extent of hybridization in the presence and absence of non-selective adsorption using fluorescein-labeled DNA. A stop-flow system of 137 microL volume permitted rapid introduction and mixing of each sample. Response times measured were on the order of seconds to minutes. Non-selective adsorption of non-complementary oligonucleotides (ncDNA) was found to occur at a significantly faster rate than hybridization of complementary oligomers (cDNA) in all cases. The presence of ncDNA oligonucleotides did not inhibit selective interactions between immobilized DNA and cDNA in solution. The presence of high concentrations of non-complementary genomic DNA had little effect on the extent of hybridization of complementary oligonucleotides, but actually reduced the response times of sensors to cDNA oligonucleotides.     

Polymerization of Ion-Pair Comonomers of Related Structures

Two ion-pair comonomers of 2-methacryloyloxyethyltrimethyl-ammonium 2-methacryloyloxyethanesulfonate (METMAMES) and 3-acrylamido-3-methylbutyltrimethylammonium 2-acrylamido-2-methylpropanesulfonate have been synthesized. The spontaneous and radical homocopolymerizations of these monomer pairs, wherein no nonpolymerizable counterions are present, have also been studied. Moreover, the intrinsic viscosity of the homocopolymer derived from METMA MES was found to increase with increasing KCl concentration in aqueous solutions.
