Saponins in Platycodi Radix (platycosides) exhibit potent biological activities in mammalian systems, including several beneficial effects such as anti-inflammatory, immunomodulatory and anti-obesity activities. In this study, we developed a new HPLC separation coupled with evaporative light scattering detector (ELSD) for the simultaneous quantitative determination of ten major saponins in Platycodi Radix. Simultaneous separation of these saponins was achieved on a C18 analytical column. The mobile phase consisted of a gradient of aqueous acetonitrile. The method was validated for linearity, precision, accuracy, limit of detection and quantification. Electrospray ionization mass spectrometry (ESI-MS) and liquid chromatography coupled with on-line mass spectrometry (LC-ESI MS/MS) were applied to identify platycosides in the purified fractions and in the crude extract. Under ESI-MS/MS conditions, the fragmentation patterns of [M-H]- ions exclusively show signals corresponding to cleavage of the glycosidic bonds, thus allowing a rapid identification of saponins in the crude extract of Platycodi Radix. The validated HPLC method provides a new basis of overall assessment on quality of Platycodi Radix, and ESI-MS/MS and LC-ESI MS/MS approaches offers analytical tools for a rapid screening of platycosides in the crude extract.  相似文献

The steady-state and transient emission properties of unsubstituted terthiophene and a series of 3'-[E-2-(4-R-phenyl)ethenyl]-2,2':5',2' '-terthiophenes (where R = H, MeO, NH(2), CN, NMe(2), NO(2)) have been examined. The R = NO(2) compound is nonemissive at room temperature in all solvents but cyclohexane. All of the other compounds show measurable steady-state emission in a variety of solvents. The behavior of these spectra may be split into two groups. The first group, those substituted compounds with R = CN, NH(2) and NMe(2), show solvatochromic behavior, where their Lippert-Mataga plots suggest changes in dipole upon photoexcitation ranging from 12.5 to 16.0 D. For the second group, where R = H and MeO (and unsubstituted terthiophene as well), the Lippert-Mataga plots indicate dipole moment changes ranging from 0 to 7.9 D. The difference in behavior between the two groups of emissive compounds can be attributed to a charge-transfer character of the emitting state in the first group. This conclusion is supported by density functional theory calculations, which show that the frontier MOs in the group one compounds are spatially separated whereas those of group two have frontier MOs that are delocalized over both the styryl and terthiophene moieties. Picosecond time-resolved fluorescence spectroscopy reveals that unsubstituted terthiophene has the shortest emission lifetime of 140 ps in acetonitrile. For the styryl substituted terthiophenes, the lifetimes are much longer and range from 320 to 670 ps for R = CN and NMe(2) respectively, a result that can be explained in terms of a smaller rate of intersystem crossing in these compounds.  相似文献

Arylthio-substituted succinic anhydrides undergo cycloaddition reactions with imines to produce gamma-lactams in high yield and with high diastereoselectivity. The origin of the selectivity is proposed to result from anion-pi repulsion in the transition state. We demonstrate high-speed terahertz (THz) time-domain spectroscopy based on electronically controlled optical sampling (ECOPS). The ECOPS system utilizes two synchronized Ti:sapphire femtosecond lasers with a 100 MHz repetition frequency. The time delay between the two laser pulses is demonstrated to be rapidly swept at a scan rate of 1 kHz on a time delay window of 77 ps by using an external offset voltage applied to a locking electronics. It is shown that a THz pulse can be exactly measured by ECOPS, as is done by asynchronous optical sampling (ASOPS), and the measurement time is shortened by a factor of 50 by using ECOPS compared with ASOPS in the case of employing 100 MHz repetition-rate lasers.  相似文献
