Cyclic dinucleotides(CDNs) are known to activate stimulator of interferon genes(STING) and induce type I interferon responses, therefor possess great potentials to be of immunotherapeutic value for cancers and infectious diseases. However, the existence of different single nucleotide polymorphism(SNP) of human STING(hSTING) gene poses an obstacle to achieve broad-spectrum activation by CDNs. We reported here the design and synthesis of a total of 36 CDNs, representing all structural variations, that contain four bases(A, G, C, U) and two linkage directions(2′-5′-linked and 3′-5′-linked phosphodiester).Through systematic evaluation of IFN-β induction with a dual-luciferase reporter assay, we discovered that wild type hSTING and two isoforms(HAQ and AQ) showed strong response while hSTING-R232 H and R293 Q exhibited the relatively weak response to CDNs stimulation. For the first time, we found that the c[G(2′,5′)U(2′,5′)] showed excellent activity against all five hSTING variants even equivalent to the endogenous ligand c[G(2′,5′)A(3′,5′)]. Furthermore, we have also demonstrated that 3′-3′CDNs with two 3′-5′ phosphodiesters showed higher serum and hydrolase stability than 2′-2′ CDNs with two 2′-5′ phosphodiesters and 2′-3′ CDNs with one 2′-5′ and one 3′-5′ phosphodiester. It is very interesting to note that 2′-2′ CDNs has been found for the first time to show strong activity. These findings will stimulate our exploration for the new functional role of CDNs, and provide guidelines to design CDNs based hSTING targeted drugs.  相似文献

稀土Eu掺杂PtRu/C催化剂及其对甲醇电氧化的性能   总被引:1,自引:0,他引:1  
采用化学还原和热处理方法对商业PtRu/C催化剂进行稀土Eu掺杂,制备了不同Eu含量的PtRuEux/C催化剂.透射电子显微镜(TEM)、X射线能量色散光谱(EDX)、X射线衍射(XRD)和X射线光电子能谱(XPS)等方法表征催化剂的结果表明,Eu的掺杂未改变PtRu/C催化剂的平均粒径(约为3nm),并且Eu以金属和氧化物两种形态修饰PtRu表面.循环伏安和计时电流法测试显示,PtRuEux/C催化剂较商业PtRu/C对甲醇氧化具有更高的活性,其中PtRuEu0.3/C的活性最高.运用原位傅里叶变换红外(FTIR)光谱从分子水平研究了该催化剂对甲醇电催化氧化的反应过程,检测到甲醇在催化剂上解离吸附的吸附态产物是线型吸附态CO(COL),Eu的掺杂使COL的氧化电位降低,明显提高了催化剂的活性和抗CO毒化的能力.  相似文献

We investigate quantum well intermixing of a double-quantum-well structure caused by phosphorus ion implantation by means of photoluminescence (PL). The ion implantation is performed at the energy of 120keV with the dose ranging from 1 × 10^11 to 1 × 10^14/cm^2. The rapid thermal annealing is performed at the temperature of 700℃ for 30s under pure nitrogen protection. The PL measurement shows that the band gap blueshift is influenced by the depth of ion implantation. The blueshift of the upper well which is closer to the implanted wcancies is enhanced with the ion dose faster than that from a lower well under the lower dose implantation (〈 5 × 10^11/cm^2). When the ion dose is over 10^12/cm^2, the band gap blueshift from both the wells increases with the ion dose and finally the two peaks combine together as one peak, indicating that the ion implantation results in a total intermixing of both the quantum wells.  相似文献

 In this paper, the thixotropy of the aqueous suspension consisting of aluminum magnesium hydroxide (AMH) possessing permanent positive charges and Na-montmorillonite possessing permanent negative charges was studied. Besides positive thixotropy and negative thixotropy, a novel thixotropic phenomenon was found, i.e., a given system studied may display, early and late, positive M28.8nthixotropic character and negative thixotropic character, we describe it as “complex thixotropy”. Finite tight frames are widely used for many applications. An important problem is to construct finite frames with prescribed norm for each vector in the tight frame. In this paper we provide a fast and simple algorithm for such a purpose. Our algorithm employs the Householder transformations. For a finite tight frame consisting of m vectors in ?n or ?n only O(nm) operations are needed. In addition, we also study the following question: Given a set of vectors in ?n or ?n, how many additional vectors, possibly with constraints, does one need to add in order to obtain a tight frame?  相似文献

采用动态光散射法研究了电解质对Mg-Al类水滑石(HTlc)溶胶聚集动力学行为的影响,发现随惰性电解质(NaCl,NaNO3)浓度的增大,Mg-Al HTlc溶胶经历稳定、慢速聚集和快速聚集三个区域,而特性吸附电解质(NaOH)在所研究的浓度范围(0~4mmol/L)内未观察到快速聚集.NaCl和NaNO3的临界聚沉浓度分别为127和180mmol/L,慢速聚集区稳定比与NaCl,NaNO3和NaOH浓度间的关系式分别为:logW=10.18-4.4410gC,logW=11.98-4.7910gC,losW=1.92-0.7410gC.NaOH诱导体系聚集的能力大大高于NaCl和NaNO3.  相似文献

Oxytropis falcata Bunge is a plant used in traditional Tibetan medicine, with reported anti-inflammatory and antioxidants effects and alleviation of myocardial ischemia reperfusion injury (MIRI). However, the underlying mechanism against MIRI and the phytochemical composition of O. falcata are vague. One fraction named OFF1 with anti-MIRI activity was obtained from O. falcata, and the chemical constituents were identified by ultra-high-performance liquid chromatography coupled with tandem mass spectrometry (UHPLC–MS). The potential targets and signaling pathways involved in the action of O. falcata against MIRI were predicted by network pharmacology analysis, and its molecular mechanism on MIRI was determined by in vitro assays. The results revealed that flavonoids are the dominant constituents of OFF1. A total of 92 flavonoids reported in O. falcata targeted 213 potential MIRI-associated factors, including tumor necrosis factor (TNF), prostaglandin-endoperoxide synthase 2 (PTGS2), and the NF-κB signaling pathway. The in vitro assay on H9c2 cardiomyocytes subjected to hypoxia/reoxygenation injury confirmed that the flavonoids in OFF1 reduced myocardial marker levels, apoptotic rate, and the inflammatory response triggered by oxidative stress. Moreover, OFF1 attenuated MIRI by downregulating the ROS-mediated JNK/p38MAPK/NF-κB pathway. Collectively, these findings provide novel insights into the molecular mechanism of O. falcata in alleviating MIRI, being a potential therapeutic candidate.  相似文献

液态固体电解质材料的离子电导率低,安全性问题在一定程度上限制了其发展与应用,而固体电解质材料在室温下具有很好的稳定性和高的离子电导率值,具有较好的应用前景.本文采用机械化学球磨法制备固体电解质Rb4Cu16I7Cl13粉末,探索制备工艺和球磨参数,对其晶体结构进行解析、观察粉体微观结构、通过交流阻抗谱及等效电路分析得到了离子电导率与活化能、并详细探讨其离子传导性能与晶体结构的关系以及化学成分稳定性进行研究.实验结果表明,在480 rpm转速下球磨6 h时可得到纯的固体电解质Rb4Cu16I7Cl13物相.粉体晶粒尺寸分布均匀,均在20 nm-400 nm之间,室温下固体电解质Rb4Cu16I7Cl13离子电导率可达到0.213 S/cm且活化能为0.087(9)eV.在真空干燥条件下存放5天和12天后观察了微观形貌和化学稳定性...  相似文献

The mode dispersion of trapezoidal cross-section narrow ridge dielectric optical waveguides is calculated by the Weighted Residual Method (WRM). In the sloping region, the field is expressed in terms of Airy functions, thus having no arbitrariness at the boundaries.
