Reactive collisions of low energy (<100-eV) mass-selected ions are used to chemically modify fluorinated self-assembled monolayer surfaces comprised of alkanethiolate chains CF3(CF2)11(CH2)2S— bound to Au. Typical experiments were done by using 1-nA/cm2 beams and submonolayer doses of reactant ions. Characterization of the modified surface was achieved by in situ chemical sputtering (60-eV Xe) and by independent high mass resolution time-of-flight-secondary ionization mass spectrometry (TOF-SIMS) (15–25-keV, Ga+) experiments. Treatment with Si35C1 4 produced a surface from which Xe+ sputtering liberated CF2 35C1+ ions, which suggested Cl-for-F halogen exchange at the surface. Isotopic labeling studies that used Si35Cl2 37Cl 2 ; and experiments with bromine-containing and iodine-containing projectiles, confirmed this reaction. High mass resolution TOF-SIMS spectra, as well as high spatial resolution images, provided further evidence as to the existence of halogen-exchanged species at the bombarded surface. Analogous Cl-for-F halogen substitution was observed in a model gas-phase reaction. The ion-surface reaction is suggested to proceed through an intermediate fluoronium ion in which the projectile is bonded to the target molecule. The most significant conclusion of the study is that selective chemical modification of monolayer surfaces can be achieved by using reactive ion beams, which lead to new covalent bonds at the surface and in the scattered ions.  相似文献

Analysis of DNA sequences isolated directly from the environment, known as metagenomics, produces a large quantity of genome fragments that need to be classified into specific taxa. Most composition-based classification methods use all features instead of a subset of features that may maximize classifier accuracy. We show that feature selection methods can boost performance of taxonomic classifiers. This work proposes three different filter-based feature selection methods that stem from information theory: (1) a technique that combines Kullback-Leibler, Mutual Information, and distance information, (2) a text mining technique, TF-IDF, and (3) minimum redundancy-maximum-relevance (mRMR). The feature selection methods are compared by how well they improve support vector machine classification of genomic reads. Overall, the 6mer mRMR method performs well, especially on the phyla-level. If the number of total features is very large, feature selection becomes difficult because a small subset of features that captures a majority of the data variance is less likely to exist. Therefore, we conclude that there is a trade-off between feature set size and feature selection method to optimize classification performance. For larger feature set sizes, TF-IDF works better for finer-resolutions while mRMR performs the best out of any method for N=6 for all taxonomic levels.  相似文献

Helically folded aromatic oligoamide foldamers have a size and geometrical parameters very distinct from those of α-helices and are not obvious candidates for α-helix mimicry. Nevertheless, they offer multiple sites for attaching side chains. It was found that some arrays of side chains at the surface of an aromatic helix make it possible to mimic extended α-helical surfaces. Synthetic methods were developed to produce quinoline monomers suitably functionalized for solid phase synthesis. A dodecamer was prepared. Its crystal structure validated the initial design and showed helix bundling involving the α-helix-like interface. These results open up new uses of aromatic helices to recognize protein surfaces and to program helix bundling in water.  相似文献

ABSTRACT

The rapid electrochemical determination of Aceclofenac (ACF) has been employed by cyclic voltammetry (CV), differential pulse voltammetry (DPV) using developed OH-functionalised multiwalled carbon nanotube carbon paste electrode (OH-MWCNT/CPE). Modified electrode was characterised by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDAX), X-ray diffraction spectroscopy (XRD), electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The ACF exhibits two oxidation peaks at +0.4 V, +0.66 V and one reduction peak at +0.3 V. The active surface area of the bare carbon paste electrode (BCPE) and modified electrode have been characterised by using K3[Fe(CN)6] solution containing 0.1 M KCl. In DPV mode, variation of ACF gave the limit of detection (LOD = 3s/m) 0.246 μM over the concentration range 1.0 to 190.0 μM (R2 = 0.9994). The developed electrode has good stability, reproducibility and could be successfully validated for the detection of ACF in pharmaceutical samples and biological fluids.  相似文献

The degree of monoclonal antibody galactosylation is known to affect complement-dependent cytotoxicity (CDC) activity by affecting C1q binding, suggesting that galactose is associated with CDC bioactivity. However, whether this association also exists under temperature stress conditions is not known. This study highlights the impact of variations in the terminal galactose content of an anti-CD20 monoclonal antibody on CDC bioactivity under high-temperature stress conditions compared with storage conditions at 2–8?°C. Drug product samples with a total galactose content of >38% showed stable CDC bioactivity at higher temperatures (45?°C), while those with 16% galactose content showed reduced CDC activity.  相似文献

The PdFe nanorods (PdFe-NRs) with tunable length were synthesized by an organic phase reaction of [Pd(acac)2] and thermal decomposition of [Fe(CO)5] in a mixture of oleyamine and octadecene at 160 °C. They show a better proton exchange membrane fuel cell (PEMFC) performance than commercial Pt/C in working voltage region of 0.80–0.65 V, due to their high intrinsic activity to oxygen reduction reaction (ORR), reduced cell inner resistance, and improved mass transport.  相似文献
