Prostate-specific antigen (PSA) is a routinely used marker of prostate cancer; however, the cut-off values for unambiguous positive/negative prostate cancer diagnoses are not defined. Therefore, despite the best effort, certain percentage of misdiagnosed cases is being recorded every year. For this reason, search for more specific diagnostic markers is of great interest. In this study, systematic comparison of PSA and metallothionein (MT) levels in blood serum of 46 prostate cancer-diagnosed patients is presented. It is clearly demonstrated that PSA levels vary significantly and despite normal total PSA values in the range of 0 - 4?ng/mL were obtained in over 36.9% of cases, positive prostate cancer was diagnosed by biopsy. In contrary, MT levels were considerably elevated in all tested samples and no significant variations were observed. These results are indicating the potential of MT as an additional prostate cancer marker reducing, in combination with PSA, the probability of false positive/negative diagnosis. To increase the throughput of the screening, chip-based capillary electrophoresis was suggested as a rapid and effective method for the fingerprinting analysis of prostate cancer from diseased blood sera.  相似文献

The expression of genes responsible for the biosynthesis of stress proteins corresponds to the exposition of an organism to abiotic and/or biotic stress. We utilize two types of paramagnetic particles for isolation of total mRNA from early somatic embryos of Norway Spruce (Picea abies /L./ Karst.) and maize plants (Zea mays L.) treated with cadmium(II) ions. The paramagnetic particles were evaluated for analysis of real samples, and poly-adenine was used as a model mRNA. Various approaches (from non-automatic to fully automatic) were tested in terms of handling the particles.

The cyclic alumosiloxane [{LAl(μ-O)(Ph(2)Si)(μ-O)}(2)] (3) and alumosilicate [{LAl(μ-O){((t)BuO)(2)Si}(μ-O)}(2)] (4) were obtained by reaction of the appropriate R(2)Si(OH)(2) precursor (R = Ph, O(t)Bu) with [{LAl(H)}(2)(μ-O)] (1), providing a nice illustration of the Loewenstein rule at work at the molecular level.  相似文献

The chiroptical properties (CD. and ORD.) of four stereoisomeric yohimbanes ( 1–4 ) and the corresponding 17-keto derivatives 5–8 have been studied. The aromatic and the ketonic Cotton effects have been considered in terms of the appropriate Sector and Octant Rules, respectively. Positively charged nitrogen makes an ‘anti-octant’ contribution to the ketonic Cotton effect. The chiroptical properties, the NMR. spectra and dehydrogenation reactions with mercuric acetate together indicate the presence of a C/D-trans-quinolizidine system in all the compounds studied, except those of the pseudo series ( 3 and 7 ), in which a C/D-cis-quinolizidine system is present. An explanation of the unexpectedly slow dehydrogenation of the 3-epi-allo compounds 4 and 8 is suggested. Conformational analysis of the results of equilibration experiments on pairs of 3-epimeric compounds (in both glacial acetic acid at 145° and in K-t-butylate at 211°) leads independently to conclusions which agree with those drawn from the physical methods. The great differences between the equilibrium positions of the free bases and the protonated forms are due to the presence in the latter cases of oriented ion pairs R3N+H…??O2CCH3.  相似文献

[reaction: see text] Two-photon induced Wolff rearrangement of a terphenyl diazoketone 1 was achieved by using focused laser pulses of 532 nm from a Q-switched Nd:YAG laser. The nonfluorescent terphenyl diazoketone 1 was transformed into a fluorescent ester derivative 4, which can be detected in situ using the focused laser pulses at 532 nm. Laser power dependence studies show that the Wolff rearrangement is induced by two-photon absorption of the terphenyl diazoketone 1, but suggests that more than two photons of 532 nm are involved (a multiphoton process) in excitation of the ester derivative 4.  相似文献

Stepwise addition of one equivalent of n-butyllithium and trimethylsilyl chloride to 2-tert-butylmercaptoaniline affords the new ligand 1-(Me3SiNH)-2-(t-BuS)C6H4 (LH), that reacts with one equivalent of butyllithium to its lithium salt LLi. Dioxodichloromolybdenum [MoO2Cl2] and dioxodichlorotungsten dimethoxyethane [WO2Cl2(dme)] react in tetrahydrofuran solution at low temperature with two equivalents LLi to monomeric dioxomolybdenum(VI) [MoO2L2] (1) and dioxotungsten(VI) complex [WO2L2] (2) employing two bidentate amido thioether ligands. The crystallographic determination of the molecular structures of 1 and 2 show evidence for M...S contacts. The reaction of [MoO2Cl2] with LLi in tetrahydrofuran solution at room temperature leads next to 1 to two compounds where silyl group migration from nitrogen to oxygen atoms occurs forming [Mo(=NL')2(OSiMe)2] (3) and [Mo(=NL')2(OSiMe3)L] (4, L' = N-2-t-BuSC6H4) as determined by NMR spectroscopy. Compound 4 was isolated in low yield and its molecular structure determined by X-ray crystallography. Higher yields of a bisimido complex can be obtained by the direct reaction of one equivalent of LLi with [Mo(NAr)2Cl2(dme)] (Ar = 2,6-Me2C6H4) forming [Mo(NAr)2LCl] (5).  相似文献

Respiratory infections are a real threat for humans, and therefore the pig model is of interest for studies. As one of a case for studies, Actinobacillus pleuropneumoniae (APP) caused infections and still worries many pig breeders around the world. To better understand the influence of pathogenic effect of APP on a respiratory system—lungs and tracheobronchial lymph nodes (TBLN), we aimed to employ matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI). In this study, six pigs were intranasally infected by APP and two were used as non-infected control, and 48 cryosections have been obtained. MALDI-TOF MSI and immunohistochemistry (IHC) were used to study spatial distribution of infectious markers, especially interleukins, in cryosections of porcine tissues of lungs (necrotic area, marginal zone) and tracheobronchial lymph nodes (TBLN) from pigs infected by APP. CD163, interleukin 1β (IL-1β) and a protegrin-4 precursor were successfully detected based on their tryptic fragments. CD163 and IL-1β were confirmed also by IHC. The protegrin-4 precursor was identified by MALDI-TOF/TOF directly on the tissue cryosections. CD163, IL-1β and protegrin-4 precursor were all significantly (p < 0.001) more expressed in necrotic areas of lungs infected by APP than in marginal zone, TBLN and in control lungs.  相似文献

Creatinine is an important diagnostic marker and is also used as a standardization tool for the quantitative evaluation of exogenous/endogenous substances in urine. This study aimed at evaluating and comparing three analytical approaches, based on hyphenations of different separation [two-dimensional capillary isotachophoresis (CITP–CITP), capillary zone electrophoresis (CZE), ultra-high-performance liquid chromatography (UHPLC)] and detection [conductivity (CD), ultraviolet (UV), tandem mass spectrometry (MS/MS)] techniques, for their ability to provide reliable clinical data along with their suitability for the routine clinical use (cost, simplicity, sample throughput). The developed UHPLC–MS/MS, CITP–CITP–CD, and CZE–UV methods were characterized by favorable performance parameters, such as linearity (r ˃ 0.99), precision (relative standard deviation, 0.22–2.97% for the creatinine position in analytical profiles), and recovery (87.1–115.1%). Clinical data, obtained from the analysis of 24 human urine samples by a reference enzymatic method, were comparable with those obtained by the tested methods (Passing–Bablok regression and Bland–Altman analysis), approving their usefulness for the routine clinical use. In this context, the UHPLC–MS/MS method provides benefits of enhanced orthogonality/accuracy and high sample throughput (threefold shorter total analysis times than the CE methods), whereas advantages of the CE methods for routine labs are simplicity and low cost of both the instrumentation and measurements.  相似文献

Correlated ab initio calculations on large systems, such as the popular MP2 (or RI-MP2) method, suffer from the intramolecular basis set superposition error (BSSE). This error is typically manifested in molecules with folded structures, characterized by intramolecular dispersion interactions. It can dramatically affect the energy differences between various conformers as well as intramolecular stabilities, and it can even impair the accuracy of the predictions of the equilibrium molecular structures. In this study, we will present two extreme cases of intramolecular BSSE, the internal stability of [n]helicene molecules and the relative energies of various conformers of phenylalanyl-glycyl-phenylalanine tripeptide (Phe-Gly-Phe), and compare the calculated data with benchmark values (experimental or high-level theoretical data). As a practical and cheap solution to the accurate treatment of the systems with large anticipated value of intramolecular BSSE, the recently developed density functional method augmented with an empirical dispersion term (DFT-D) is proposed and shown to provide very good results in both of the above described representative cases.  相似文献
