Immobilization of urease and glutamate dehydrogenase enzymes in electrochemically prepared polypyrrole-polyvinyl sulfonate films (PPY-PVS) was carried out using physical adsorption and electrochemical entrapment techniques. Detailed studies on optimum pH, Fourier transform infrared spectroscopy, cyclic voltammetry, and scanning electron microscopy of the enzymes in the immobilized state were conducted. The value of the apparent Michaelis-Menten constant was experimentally determined to be 2.5 and 2.7 for physically adsorbed and electrochemically entrapped urease in PPY-PVS films, respectively.  相似文献

The replacement of semolina with potato flour (PF) and potato mash (PM) at different levels was assessed for its effects on pasta quality. The results showed that the addition of PF and PM increased the pasting viscosity of the blends; in addition, PF enhanced the functional properties, while PM reduced them. The minimum cooking time decreased with PF and PM, while the PF pasta exhibited a higher cooking loss (5.02 to 10.44%) than the PM pasta, which exhibited a lower cooking loss. The pasta with PF and PM showed an increase in the total phenolic and flavonoid content, with reduced in vitro digestibility as confirmed by Fourier transform infrared spectroscopy. The PF pasta exhibited lower lightness and higher yellowness than the PM pasta, and its firmness and toughness also modulated owing to the complex interaction between potato starches and the gluten protein matrix, as evident from scanning electron microscopy. Sensory data revealed that pasta containing 30% PF and 16% PM was highly acceptable.  相似文献

Two particle correlations of hadrons produced in 360 GeV/cpp interactions are investigated in the transverse plane and in rapidity. The data were obtained at the European hybrid spectrometer equipped with a rapid cycling bubble chamber. The observed transverse and rapidity correlations are compared with the one string LUND-and a two string dual parton-model. These models predict in general stronger correlations in the transverse plane and much weaker correlations in rapidity than found in the data. The LUND-FRITIOF-and multichain dual parton models provide a better reproduction of the data, although the agreement is not yet satisfactory. Only the UA5 cluster model GENCL shows agreement with the data.  相似文献

A nanostructured iron oxide (NanoFe3O4, particle size ca. 25 nm and roughness ca. 21 nm) film deposited onto a hydrolyzed indium‐tin‐oxide (ITO) coated glass plate has been used to immobilize cholesterol oxidase (ChOx) to fabricate an impedimetric cholesterol sensor. Electrochemical studies reveal that surface charged Fe3O4 nanoparticles provide better conformation for ChOx loading resulting in enhanced electron transfer between ChOx and the electrode. Impedimetric response studies of the ChOx/NanoFe3O4/ITO bioelectrode exhibit improved linearity (2.5–400 mg/dL), low detection limit (0.25 mg/dL), fast response time (25 s), high sensitivity (86 Ω/mg dL?1/cm?2) and a low value of the Michaelis‐Menten constant (Km, 0.8 mg/dL) with a regression coefficient of 0.997.  相似文献

Background

The histological extent of inflammatory bowel disease (IBD) is greater than that evident by colonoscopic evaluation. We hypothesized that metabolic profile in macroscopically un-involved colonic mucosa in IBD is similar to that of controls with healthy colon. We thus assessed the differences in metabolic profile in macroscopically involved and un-involved colonic mucosa of IBD patients to further substantiate the extent of disease.

Patients and Methods

Colonic mucosal biopsies were obtained and snap frozen from both the macroscopically un-involved and involved colonic mucosa of IBD patients and macroscopically normal colonic mucosa of controls and were subjected to in-vitro high-resolution proton (1H) magnetic resonance (MR) spectroscopy and the concentrations of metabolites were determined.

Results

Thirty-two metabolites were assigned in the proton MR spectrum of colonic mucosa of IBD patients. The concentrations of amino acids (isoleucine, leucine, valine, arginine, lysine, glutamine/glutamate, alanine), membrane metabolites (choline, glycerophosphorylcholine/phosphorylcholine), glycolytic product (lactate) and short chain fatty acid (formate) were significantly lower while significantly high level of glucose were observed in the macroscopically un-involved colonic mucosa of IBD patients compared to the macroscopically normal mucosa of controls. There was no significant difference in the concentrations of metabolites in macroscopically involved and un-involved colonic mucosa of IBD patients.

Conclusions

The metabolic profile in macroscopically un-involved colonic mucosa of IBD patients is similar to that of macroscopically involved mucosa but different from colonic mucosa of controls. This suggests that even macroscopically un-involved colonic mucosa is metabolically abnormal and may explain the increase in extent of disease with time.  相似文献

Flash pyrolysis of fentanyl and its analogues has been studied on pyrolysis-gas chromatograph-mass spectrometer (Py-GC-MS) system. Initial pyrolytic fragmentation of these compounds led to the formation of N-substituted-1,2,5,6-tetrahydropyridine and N-phenylpropanamide as the primary pyrolytic products. Moreover, depending up on the furnace temperature, these pyrolytic products can also undergo further fragmentation to give different compounds. We, herein, discuss the probable fragmentation routes of parent as well as pyrolytic products. This study will be useful while developing technologies for thermal aerosol generation of fentanyl and related compounds.  相似文献

Detection of these pathogens originating from different sources has led to increased interest of researchers. New bio-molecular techniques for food pathogen detection are being developed to improve the sensor characteristics such as sensitivity, reusability, simplicity and economic viability. In this paper, we address the following probabilistic version (PSC) of the set covering problem: where A is a 0-1 matrix, is a random 0-1 vector and is the threshold probability level. We introduce the concepts of p-inefficiency and polarity cuts. While the former is aimed at deriving an equivalent MIP reformulation of (PSC), the latter is used as a strengthening device to obtain a stronger formulation. Simplifications of the MIP model which result when one of the following conditions hold are briefly discussed: A is a balanced matrix, A has the circular ones property, the components of are pairwise independent, the distribution function of is a stationary distribution or has the disjunctive shattering property. We corroborate our theoretical findings by an extensive computational experiment on a test-bed consisting of almost 10,000 probabilistic instances. This test-bed was created using deterministic instances from the literature and consists of probabilistic variants of the set covering model and capacitated versions of facility location, warehouse location and k-median models. Our computational results show that our procedure is orders of magnitude faster than any of the existing approaches to solve (PSC), and in many cases can reduce hours of computing time to a fraction of a second.
