The potential of 1.7 μm ethylene bridged hybrid silica phase was investigated for the separation of twelve imidazolium-based ionic liquid cations. U-shaped retention profile was observed for all solutes with an increase in retention at both low and high acetonitrile content. Chromatographic behaviour of imidazolium cations in both hydrophilic interaction chromatography (HILIC) and per aqueous liquid chromatography (PALC) modes was studied by varying key parameters such as buffer concentration and pH, acid additive, organic modifier and column temperature. Experimental data provided some evidences that under PALC conditions cationic solutes are retained predominantly by mixed hydrophobic/ion-exchange interactions. In the HILIC mode, both partitioning and ion-exchange interactions are responsible for the retention of solutes. Compared to PALC, HILIC provided significantly higher efficiencies with less or even no peak tailing, better separation selectivity and greater resistance to overload. In PALC mode gradient elution was required to achieve adequate retentivity of all solutes but selectivity was not sufficient to distinguish between solutes with very similar hydrophobicity. In contrast, under HILIC conditions twelve solutes were almost completely resolved in less than 4 min by using isocratic elution. Summarizing, it could be concluded that ethylene bridged hybrid silica column providing a dual retention mechanism offers the possibility of selecting between the two retention modes with opposite separation selectivity, just by changing the composition of the mobile phase.  相似文献

The phloroglucides of the ferns of the Dryopteris filix-mas complex; i.e. D. abbreviata, D. filix-mas s. str. and D. borreri (diploid and triploid) as well as those of D. villarii, subspvillarii and subsp. pallida were reinvestigated with improved semiquantitative analytical methods. The phloroglucides of D. aitoniana and D. athamantica were examined for the first time. The results (table 1) are compatible with the hypothesis, without proving it, that the allotetraploid species D. filix-mas s. str. originated from a hybrid of D. abbreviata with D. villard (with subsequent doubling of its chromosomes) and the apogamous triploid D. remota from D. assimils with diploid D. borreri, and the equally apogamous triploid D. borreri from a hybrid of D. abbreviata (or a related diploid sexual taxon) with diploid D. borreri. D. aitoniana contains a large amount of trisflavaspidic acid but no filixic acid and differs in this respect from the three representatives of the D. filix-mas complex.  相似文献

Motivated by an open problem from graph drawing, we study several partitioning problems for line and hyperplane arrangements. We prove a ham-sandwich cut theorem: given two sets of n lines in ?2, there is a line ? such that in both line sets, for both halfplanes delimited by ?, there are $\sqrt{n}$ lines which pairwise intersect in that halfplane, and this bound is tight; a centerpoint theorem: for any set of n lines there is a point such that for any halfplane containing that point there are $\sqrt{n/3}$ of the lines which pairwise intersect in that halfplane. We generalize those results in higher dimension and obtain a center transversal theorem, a same-type lemma, and a positive portion Erd?s–Szekeres theorem for hyperplane arrangements. This is done by formulating a generalization of the center transversal theorem which applies to set functions that are much more general than measures. Back to graph drawing (and in the plane), we completely solve the open problem that motivated our search: there is no set of n labeled lines that are universal for all n-vertex labeled planar graphs. In contrast, the main result by Pach and Toth (J. Graph Theory 46(1):39–47, 2004), has, as an easy consequence, that every set of n (unlabeled) lines is universal for all n-vertex (unlabeled) planar graphs.  相似文献

Oncogenic RAS proteins, involved in ∼30% of human tumors, are molecular switches of various signal transduction pathways. Here we apply a new protocol for the NMR study of KRAS in its (inactive) GDP- and (activated) GTP-bound form, allowing a comprehensive analysis of the backbone dynamics of its WT-, G12C- and G12D variants. We found that Tyr32 shows opposite mobility with respect to the backbone of its surroundings: it is more flexible in the GDP-bound form while more rigid in GTP-complexes (especially in WT- and G12D-GTP). Using the G12C/Y32F double mutant, we showed that the presence of the hydroxyl group of Tyr32 has a marked effect on the G12C-KRAS-GTP system as well. Molecular dynamics simulations indicate that Tyr32 is linked to the γ-phosphate of GTP in the activated states – an arrangement shown, using QM/MM calculations, to support catalysis. Anchoring Tyr32 to the γ-phosphate contributes to the capture of the catalytic waters participating in the intrinsic hydrolysis of GTP and supports a simultaneous triple proton transfer step (catalytic water → assisting water → Tyr32 → O1G of the γ-phosphate) leading to straightforward product formation. The coupled flip of negatively charged residues of switch I toward the inside of the effector binding pocket potentiates ligand recognition, while positioning of Thr35 to enter the coordination sphere of the Mg2+ widens the pocket. Position 12 mutations do not disturb the capture of Tyr32 by the γ-phosphate, but (partially) displace Gln61, which opens up the catalytic pocket and destabilizes catalytic water molecules thus impairing intrinsic hydrolysis.

A weak ion-exchange grafted methacrylate monolith was prepared by grafting a methacrylate monolith with glycidyl methacrylate and subsequently modifying the epoxy groups with diethylamine. The thickness of the grafted layer was determined by measuring permeability and found to be approximately 90nm. The effects of different buffer solutions on the pressure drop were examined and indicated the influence of pH on the permeability of the grafted monolith. Protein separation and binding capacity (BC) were found to be flow-unaffected up to a linear velocity of 280cm/h. A comparison of the BC for the non-grafted and grafted monolith was performed using beta-lactoglobulin, bovine serum albumin (BSA), thyroglobulin, and plasmid DNA (pDNA). It was found that the grafted monolith exhibited 2- to 3.5-fold higher capacities (as compared to non-grafted monoliths) in all cases reaching values of 105, 80, 71, and 17mg/ml, respectively. It was determined that the maximum pDNA capacity was reached using 0.1M NaCl in the loading buffer. Recovery was comparable and no degradation of the supercoiled pDNA form was detected. Protein z-factors were equal for the non-grafted and grafted monolith indicating that the same number of binding sites are available although elution from the grafted monolith occurred at higher ionic strengths. The grafted monolith exhibited lower efficiency than the non-grafted ones. However, the baseline separation of pDNA from RNA and other impurities was achieved from a real sample.  相似文献

Marine macroalgae are well known to release a wide spectrum of volatile organic components, the release of which is affected by environmental factors. This paper aimed to identify the essential oil (EO) compounds of the brown algae Cystoseira compressa collected in the Adriatic Sea monthly, from May until August. EOs were isolated by hydrodistillation using a Clavenger-type apparatus and analyzed by gas chromatography coupled with mass spectrometry (GC–MS). One hundred four compounds were identified in the volatile fraction of C. compressa, accounting for 84.37–89.43% of the total oil. Samples from May, June, and July were characterized by a high share of fatty acids (56, 69, and 34% respectively) with palmitic acid being the dominant one, while in the August sample, a high content of alcohols (mainly phytol and oleyl alcohol) was found. Changes in the other minor components, which could be important for the overall aroma and biological activities of the algal samples, have also been noted during the vegetation periods. The results of this paper contribute to studies of algal EOs and present the first report on C. compressa EOs.  相似文献

n‐Butyl acrylate (BA), 2‐ethylhexyl acrylate (EHA), and methyl methacrylate (MMA) are commonly used monomers in pressure‐sensitive adhesive formulations. The bulk free‐radical copolymerizations of BA/EHA, MMA/EHA, and BA/MMA are studied at 60 °C to demonstrate the use of copolymer reactivity ratios for the prediction of BA/MMA/EHA terpolymer composition. The reactivity ratios for BA/EHA and MMA/EHA copolymer systems are determined using low conversion experiments; BA/MMA reactivity ratios are already known from the literature. The reactivity ratio estimates for the BA/EHA system are r BA = 0.994 and r EHA = 1.621 and the estimates for MMA/EHA are r MMA = 1.496 and r EHA = 0.315. High conversion experiments are conducted to validate the reactivity ratios. The copolymer reactivity ratios are shown to predict terpolymer composition of high conversion BA/MMA/EHA experiments.  相似文献

Novel 2‐(1‐substituted‐1H‐1,2,3‐triazol‐4‐yl)pyridine (pytl) ligands have been prepared by “click chemistry” and used in the preparation of heteroleptic complexes of Ru and Ir with bipyridine (bpy) and phenylpyridine (ppy) ligands, respectively, resulting in [Ru(bpy)2(pytl‐R)]Cl2 and [Ir(ppy)2(pytl‐R)]Cl (R=methyl, adamantane (ada), β‐cyclodextrin (βCD)). The two diastereoisomers of the Ir complex with the appended β‐cyclodextrin, [Ir(ppy)2(pytl‐βCD)]Cl, were separated. The [Ru(bpy)2(pytl‐R)]Cl2 (R=Me, ada or βCD) complexes have lower lifetimes and quantum yields than other polypyridine complexes. In contrast, the cyclometalated Ir complexes display rather long lifetimes and very high emission quantum yields. The emission quantum yield and lifetime (Φ=0.23, τ=1000 ns) of [Ir(ppy)2(pytl‐ada)]Cl are surprisingly enhanced in [Ir(ppy)2(pytl‐βCD)]Cl (Φ=0.54, τ=2800 ns). This behavior is unprecedented for a metal complex and is most likely due to its increased rigidity and protection from water molecules as well as from dioxygen quenching, because of the hydrophobic cavity of the βCD covalently attached to pytl. The emissive excited state is localized on these cyclometalating ligands, as underlined by the shift to the blue (450 nm) upon substitution with two electron‐withdrawing fluorine substituents on the phenyl unit. The significant differences between the quantum yields of the two separate diastereoisomers of [Ir(ppy)2(pytl‐βCD)]Cl (0.49 vs. 0.70) are attributed to different interactions of the chiral cyclodextrin substituent with the Δ and Λ isomers of the metal complex.  相似文献
