The bulk magnetic properties and 166Er Mössbauer spectra of Er2?xPrxCo17 pseudo-binaries are reported. Incorporation of Pr into Er2Co17 increases the anisotropy field (HA) and the saturation magnetization, while resulting in only a slight (5%) decrease in the Curie temperature. The quadrupole interaction deduced from the Mössbauer spectrum suggests that Pr preferentially substitutes for Er in Er2Co17 at the 2d site. The rise in HA is a consequence of the preferential substitution, which occurs because spatial constraints at the 2d site are less for the larger Pr3+ ion. Coupling between Er-Co and Pr-Co is found to be antiferromagnetic and ferromagnetic, respectively. 相似文献

Background
Progranulin is a secreted high molecular weight growth factor bearing seven and one half copies of the cysteine-rich granulin-epithelin motif. While inappropriate over-expression of the progranulin gene has been associated with many cancers, haploinsufficiency leads to atrophy of the frontotemporal lobes and development of a form of dementia (frontotemporal lobar degeneration with ubiquitin positive inclusions, FTLD-U) associated with the formation of ubiquitinated inclusions. Recent reports indicate that progranulin has neurotrophic effects, which, if confirmed would make progranulin the only neuroprotective growth factor that has been associated genetically with a neurological disease in humans. Preliminary studies indicated high progranulin gene expression in spinal cord motor neurons. However, it is uncertain what the role of Progranulin is in normal or diseased motor neuron function. We have investigated progranulin gene expression and subcellular localization in cultured mouse embryonic motor neurons and examined the effect of progranulin over-expression and knockdown in the NSC-34 immortalized motor neuron cell line upon proliferation and survival.Results
In situ hybridisation and immunohistochemical techniques revealed that the progranulin gene is highly expressed by motor neurons within the mouse spinal cord and in primary cultures of dissociated mouse embryonic spinal cord-dorsal root ganglia. Confocal microscopy coupled to immunocytochemistry together with the use of a progranulin-green fluorescent protein fusion construct revealed progranulin to be located within compartments of the secretory pathway including the Golgi apparatus. Stable transfection of the human progranulin gene into the NSC-34 motor neuron cell line stimulates the appearance of dendritic structures and provides sufficient trophic stimulus to survive serum deprivation for long periods (up to two months). This is mediated at least in part through an anti-apoptotic mechanism. Control cells, while expressing basal levels of progranulin do not survive in serum free conditions. Knockdown of progranulin expression using shRNA technology further reduced cell survival.Conclusion
Neurons are among the most long-lived cells in the body and are subject to low levels of toxic challenges throughout life. We have demonstrated that progranulin is abundantly expressed in motor neurons and is cytoprotective over prolonged periods when over-expressed in a neuronal cell line. This work highlights the importance of progranulin as neuroprotective growth factor and may represent a therapeutic target for neurodegenerative diseases including motor neuron disease. 相似文献23.

The assembly of nanoparticles into large, two-dimensional structures provides a route for the exploration of collective phenomena among mesoscopic building blocks. We characterize the structure of Langmuir monolayers of dodecanethiol-ligated gold nanoparticles with in situ optical microscopy and X-ray scattering. The interparticle spacing increases with thiol concentration and does not depend on surface pressure. The correlation lengths of the Langmuir monolayer crystalline domains are on the order of five to six particle diameters. Further compression of the monolayers causes wrinkling; however, we find that wrinkled monolayers with excess thiol can relax to an unwrinkled state following a reduction of surface pressure. A theoretical model based on van der Waals attraction and tunable steric repulsion is adopted to explain this reversibility. 相似文献

Three sharp absorption features in the energy range 2.36–2.55 eV have been detected in the transmission spectrum of Co-diffused ZnSe, and a number of luminescence transitions originating from the lowest of these states at 2.361 eV have been observed. Photoluminescence excitation spectra prove that these are high energy excited states of the Co2+Zn impurity, a conclusion confirmed by comparison of measured and predicted luminescence energies. This represents the first identification of luminescence branching from a higher excited state of a transition metal ion in any semiconductor. The sharp, weakly phonon-coupled transitions involve either intra-impurity excitation or transitions from the impurity to localised states split off from a minimum in the conduction band. The implications of these observations for the mechanism of host-impurity energy transfer and for the nature of the excited state wavefunctions are discussed. 相似文献
