A convergent synthesis of cationic amphiphilic compounds is reported here with the use of the phosphonodithioester–amine coupling (PAC) reaction. This versatile reaction occurs at room temperature without any catalyst, allowing binding of the lipid moiety to a polar head group. This strategy is illustrated with the use of two lipid units featuring either two oleyl chains or two-branched saturated lipid chains. The final cationic amphiphiles were evaluated as carriers for plasmid DNA delivery in four cell lines (A549, Calu3, CFBE and 16HBE) and were compared to standards (BSV36 and KLN47). These new amphiphilic derivatives, which were formulated with DOPE or DOPE-cholesterol as helper lipids, feature high transfection efficacies when associated with DOPE. The highest transfection efficacies were observed in the four cell lines at low charge ratios (CR = 0.7, 1 or 2). At these CRs, no toxic effects were detected. Altogether, this new synthesis scheme using the PAC reaction opens up new possibilities for investigating the effects of lipid or polar head groups on transfection efficacies. 相似文献

The epiperimetric inequality introduced by E. R. Reifenberg in [3] gives a rate of decay at a point for the decreasing k‐density of area of an area‐minimizing integral k‐cycle. While dilating the cycle at that point, this rate of decay holds once the configuration is close to a tangent cone configuration and above the limiting density corresponding to that configuration. This is why we propose to call the Reifenberg epiperimetric inequality an upper‐epiperimetric inequality. A direct consequence of this upper‐epiperimetric inequality is the statement that any point possesses a unique tangent cone. The upper‐epiperimetric inequality was proven by B. White in [5] for area‐minimizing 2‐cycles in ?n. In the present paper we introduce the notion of a lower‐epiperimetric inequality. This inequality gives this time a rate of decay for the decreasing k‐density of area of an area‐minimizing integral k‐cycle, while dilating the cycle at a point once the configuration is close to a tangent cone configuration and below the limiting density corresponding to that configuration. Our main result in the present paper is to prove the lower‐epiperimetric inequality for area‐minimizing 2‐cycles in ?n. As a consequence of this inequality we prove the “splitting before tilting” phenomenon for calibrated 2‐rectifiable cycles, which plays a crucial role in the proof of the regularity of 1‐1 integral currents in [4]. © 2004 Wiley Periodicals, Inc. 相似文献

Advances in nanoparticle technology have recently offered new tools to the bioanalytical field of research. In particular, new nanoparticle‐based sensors have appeared able to give quantitative information about different species (ions, metabolites, biomolecules) in biosamples through ratiometric measurements. This article describes the methodologies developed so far in the design of such nanosensors. In particular, the different approaches to immobilize fluorescent chemosensor dyes to nanoparticles are presented. Herein, we report the photoinitiated polymerization-induced self-assembly (photo-PISA) of spherical micelles consisting of proapoptotic peptide–polymer amphiphiles. The one-pot synthetic approach yielded micellar nanoparticles at high concentrations and at scale (150 mg mL−1) with tunable peptide loadings up to 48 wt. %. The size of the micellar nanoparticles was tuned by varying the lengths of hydrophobic and hydrophilic building blocks. Critically, the peptide-functionalized nanoparticles imbued the proapoptotic “KLA” peptides (amino acid sequence: KLAKLAKKLAKLAK) with two key properties otherwise not inherent to the sequence: 1) proteolytic resistance compared to the oligopeptide alone; 2) significantly enhanced cell uptake by multivalent display of KLA peptide brushes. The result was demonstrated improved apoptosis efficiency in HeLa cells. These results highlight the potential of photo-PISA in the large-scale synthesis of functional, proteolytically resistant peptide–polymer conjugates for intracellular delivery. 相似文献
