A new copper hydroxydiphosphate Cu3(P2O6OH)2 was synthesized, by soft chemistry. The crystal structure was solved ab initio from X-ray powder diffraction data in the triclinic space group P. The structure is built up from [Cu3O10]infinity zigzag chains linked by P2O6(OH) groups to form a tridimensional framework. The [Cu3O10]infinity chains consist of edge-sharing polyhedra. The structure contains two sorts of copper polyhedra: one CuO6 octahedron and two CuO5 pyramids. Magnetization measurements confirm the presence of divalent copper and suggest antiferromagnetic interactions at low temperature.  相似文献

Amyloid β oligomers (Aβo) are the main toxic species in Alzheimer''s disease, which have been targeted for single drug treatment with very little success. In this work we report a new approach for identifying functional Aβo binding compounds. A tailored library of 971 fluorine containing compounds was selected by a computational method, developed to generate molecular diversity. These compounds were screened for Aβo binding by a combined 19F and STD NMR technique. Six hits were evaluated in three parallel biochemical and functional assays. Two compounds disrupted Aβo binding to its receptor PrPC in HEK293 cells. They reduced the pFyn levels triggered by Aβo treatment in neuroprogenitor cells derived from human induced pluripotent stem cells (hiPSC). Inhibitory effects on pTau production in cortical neurons derived from hiPSC were also observed. These drug-like compounds connect three of the pillars in Alzheimer''s disease pathology, i.e. prion, Aβ and Tau, affecting three different pathways through specific binding to Aβo and are, indeed, promising candidates for further development.

A new approach combining virtual screening, 19F and STD NMR, and biochemical assays using hiPSC and targetting multiple pathways involving Aβ, PrPC and Tau provides a more effective strategy for Alzheimer''s disease drug discovery than Aβ only approach.  相似文献

It is now recognized that self-assembly is a powerful synthetic approach to the fabrication of nanostructures with feature sizes smaller than achievable with state of the art lithography and with a complexity approaching that of biological systems. For example, recent research has shown that silica/surfactant self-assembly combined with evaporation (so-called evaporation induced self-assembly EISA) can direct the formation of porous and composite thin-film mesostructures characterized by precise periodic arrangements of inorganic and organic constituents on the 1-50-nm scale. Despite the potential utility of these films for a diverse range of applications such as sensors, membranes, catalysts, waveguides, lasers, nano-fluidic systems, and low dielectric constant (so-called low k) insulators, the mechanism of EISA is not yet completely understood. Here, using time-resolved grazing incidence small-angle X-ray scattering (GISAXS) combined with gravimetric analysis and infrared spectroscopy, we structurally and compositionally characterize in situ the evaporation induced self-assembly of a homogeneous silica/surfactant/solvent solution into a highly ordered surfactant-templated mesostructure. Using CTAB (cetyltrimethylammonium bromide) as the structure-directing surfactant, a two-dimensional (2-D) hexagonal thin-film mesophase (p6mm) with cylinder axes oriented parallel to the substrate surface forms from an incipient lamellar mesophase through a correlated micellar intermediate. Comparison with the corresponding CTAB/water/alcohol system (prepared without silica) shows that, for acidic conditions in which the siloxane condensation rate is minimized, the hydrophilic and nonvolatile silicic acid components replace water maintaining a fluidlike state that avoids kinetic barriers to self-assembly.  相似文献

The development of methodologies to control on demand and reversibly supramolecular transformations from self-assembled metalla-structures requires the rational design of architectures able to answer to an applied stimulus. While solvent or concentration changes, light exposure or addition of a chemical have been largely explored to provide these transformations, the case of pH sensitive materials is less described. Herein, we report the first example of a pH-triggered dissociation of a coordination-driven self-assembled interlocked molecular link. It incorporates a pH sensitive benzobisimidazole-based ligand that can be selectively protonated on its bisimidazole moieties. This generates intermolecular electrostatic repulsions that reduces drastically the stability of the interlocked structure, leading to its dissociation without any sign of protonation of the pyridine moieties involved in the coordination bonds. Importantly, the dissociation process is reversible through addition of a base.  相似文献

The π coordination of arene and anionic heteroarene ligands is a ubiquitous bonding motif in the organometallic chemistry of d‐block and f‐block elements. By contrast, related π interactions of neutral heteroarenes including neutral bora‐π‐aromatics are less prevalent particularly for the f‐block, due to less effective metal‐to‐ligand backbonding. In fact, π complexes with neutral heteroarene ligands are essentially unknown for the actinides. We have now overcome these limitations by exploiting the exceptionally strong π donor capabilities of a neutral 1,4‐diborabenzene. A series of remarkably robust, π‐coordinated thorium(IV) and uranium(IV) half‐sandwich complexes were synthesized by simply combining the bora‐π‐aromatic with ThCl4(dme)2 or UCl4, representing the first examples of actinide complexes with a neutral boracycle as sandwich‐type ligand. Experimental and computational studies showed that the strong actinide–heteroarene interactions are predominately electrostatic in nature with distinct ligand‐to‐metal π donation and without significant π/δ backbonding contributions.  相似文献

The authors present a first-principles prediction of the energies of the eight lowest-lying anharmonic vibrational states of CO(2), including the fundamental symmetric stretching mode and the first overtone of the fundamental bending mode, which undergo a strong coupling known as Fermi resonance. They employ coupled-cluster singles, doubles, and (perturbative) triples [CCSD(T) and CCSDT] in conjunction with a range of Gaussian basis sets (up to cc-pV5Z, aug-cc-pVQZ, and aug-cc-pCVTZ) to calculate the potential energy surfaces (PESs) of the molecule, with the errors arising from the finite basis-set sizes eliminated by extrapolation. The resulting vibrational many-body problem is solved by the vibrational self-consistent-field and vibrational configuration-interaction (VCI) methods with the PESs represented by a fourth-order Taylor expansion or by numerical values on a Gauss-Hermite quadrature grid. With the VCI, the best theoretical estimates of the anharmonic energy levels agree excellently with experimental values within 3.5 cm(-1) (the mean absolute deviation). The theoretical (experimental) anharmonic frequencies of the Fermi doublet are 1288.9 (1285.4) and 1389.3 (1388.2) cm(-1).  相似文献

In an effort to reduce organ replacement and enhance tissue repair, there has been a tremendous effort to create biomechanically optimized scaffolds for tissue engineering applications. In contrast, the development and characterization of electroactive scaffolds has attracted little attention. Consequently, the creation and characterization of a carbon nanotube based poly(lactic acid) nanofiber scaffold is described herein. After 28 d in physiological solution at 37 °C, a change in the mass, chemical properties and polymer morphology is seen, while the mechanical properties and physical integrity are unaltered. No adverse cytotoxic affects are seen when mesenchymal stem cells are cultured in the presence of the scaffold. Taken together, these data auger well for electroactive tissue engineering.

This review article summarizes the findings of recent typing studies conducted on Vibrio cholerae, V. parahaemolyticus, and V. vulnificus. The DNA-based methods used to type the Vibrio spp. include whole genome approaches, such as pulsed field gel electrophoresis (PFGE), ribotyping, and repetitive extragenic palindromic (REP)-PCR, single gene targets, and multiple gene targets (multilocus approaches). The goals of these studies include establishing the relatedness of isolates from disease epidemics, discriminating among strains with more or less potential to cause disease or epidemics, and exploring the population biology of these waterborne pathogens. PFGE was consistently among the more discriminatory of the typing methods for all three Vibrio spp., and was useful for tracing the temporal and geographic relatedness of epidemic strains of V. cholerae and V. parahaemolyticus. However, PFGE did not group V. vulnificus strains according to the genotypes that have been proposed as markers of virulence potential. Typing methods that target repetitive elements distributed throughout the genome, such as BOX-PCR and REP-PCR, and DNA sequence-based methods, such as multilocus sequence typing, were also highly discriminatory and, in some cases, superior to PFGE for phylogenetic analysis and identification of strains with high epidemic or virulence potential. As typing methods and strategies are refined and used, the epidemiology, virulence potential, and ecology of these pathogenic Vibrio spp. will become better understood.  相似文献
