Chromatographic determination of glutathione disulfide (GSSG) without any preliminary reduction has been presented using GSSG derivatization by p-hydroxymercuribenzoate (pHMB) in strong alkaline medium followed by the determination of GS-pHMB complex by reversed phase chromatography coupled to chemical vapour generation and atomic fluorescence detector (RPC-CVGAFS). A detection limit of 35 nM for GSSG (corresponding to 1.8 pmol) detected as GS-pHMB species was achieved based on a signal-to-noise ratio of 3 in buffer and in blood. The proposed method was applied to the determination of GSSG in whole blood and validated by the classical determination of GSSG by derivatization after reduction with dithiothreitol (DTT).  相似文献

A new CE system based on the use of polymeric-mixed micelles (cholic acid, SDS and the poloxamine Tetronic(?) 1107) was developed for the simultaneous determination of nine steroids in human urine. This method allows the baseline separation and quantitation of cortisol, androstenedione, estriol, dehydroepiandrosterone sulfate, testosterone, dehydroepiandrosterone, estrone, progesterone and estradiol in less than 25 min showing to be sensitive enough to detect low concentrations of these steroids in urine samples (5-45 ng/mL). The optimized electrophoretic conditions were performed using a 50 cm × 75 μm capillary, 18 kV, 25°C, with 44 mM cholic acid, 10 mM SDS, 0.05% w/v tetronic(?) 1107, 2.5% v/v methanol, 2.5% v/v tetrahydrofuran in 5 mM borate - 5 mM phosphate buffer (pH=8.0) as a background electrolyte and a dual 210/254 UV-detection. The method can simultaneously determine 0.1-120 μg/mL, which corresponds to 5-6000 ng/mL of steroids in 2 mL urine. The recoveries ranged between 82.4 and 101.5%. Due to its simplicity, speed, accuracy and reliability, the proposed method could be a potential alternative to the traditional methodologies used with clinical purposes.  相似文献

The lipid transfer protein (LTP), Pru p 3, has been identified as the major allergen present in peach, and its sequence obtained by direct amino acid sequencing has been previously reported. However, several sequences, obtained from c‐DNA and available in databases, show differences among them and from the originally proposed structure. In this paper, we report the fast and unambiguous determination of the structure of Pru p 3 protein, extracted from three different varieties of peach, by electrospray ionization mass spectrometry (ESI‐MS), both coupled to single stage (quadrupole) or advanced (FT‐HRMS) analyzers. The structure was identical to one of the cDNA‐derived sequences and different in two positions from the previously reported structure obtained by amino acid sequencing. Moreover, the exclusive localization of the protein in the outer part of the fruits was assessed by Matrix‐Assisted Laser Desorption Ionization Mass Spectrometry Imaging (MALDI MSI). The results reported here demonstrate the full potential of mass spectrometry for rapidly obtaining high quality structural data of relevant food proteins. The photodegradation of the herbicide clomazone in the presence of S2O82? or of humic substances of different origin was investigated. A value of (9.4 ± 0.4) × 108 m ?1 s?1 was measured for the bimolecular rate constant for the reaction of sulfate radicals with clomazone in flash‐photolysis experiments. Steady state photolysis of peroxydisulfate, leading to the formation of the sulfate radicals, in the presence of clomazone was shown to be an efficient photodegradation method of the herbicide. This is a relevant result regarding the in situ chemical oxidation procedures involving peroxydisulfate as the oxidant. The main reaction products are 2‐chlorobenzylalcohol and 2‐chlorobenzaldehyde. The degradation kinetics of clomazone was also studied under steady state conditions induced by photolysis of Aldrich humic acid or a vermicompost extract (VCE). The results indicate that singlet oxygen is the main species responsible for clomazone degradation. The quantum yield of O2(a1Δg) generation (λ = 400 nm) for the VCE in D2O, ΦΔ = (1.3 ± 0.1) × 10?3, was determined by measuring the O2(a1Δg) phosphorescence at 1270 nm. The value of the overall quenching constant of O2(a1Δg) by clomazone was found to be (5.7 ± 0.3) × 107 m ?1 s?1 in D2O. The bimolecular rate constant for the reaction of clomazone with singlet oxygen was kr = (5.4 ± 0.1) × 107 m ?1 s?1, which means that the quenching process is mainly reactive.  相似文献

The complex formation equilibria of aspergillic acid with iron(III) were studied by potentiometric and spectrophotometric methods. The ligand was prepared by a biosynthetic route and its purity checked by MS and 1H NMR spectroscopies. Some structural features of the ligand are discussed on the basis of NMR results. The iron(III) coordination model is compared with those of two other cyclic hydroxamates, 1-hydroxy-2-pyridinone and 1,4-dihydroxy-2-pyridinone, and its features are discussed on the basis of quantum chemical calculations.  相似文献

New tricks for an old dog : Calixpyrroles bind anions efficiently and can be transformed into transition‐metal complexes only under forcing conditions. Reducing the macrocycle creates a ligand that easily forms classical Werner complexes with copper, nickel, and palladium ions. The metal complexes present an array of four directed hydrogen bonds, which specifically bind the counterions (see picture; blue N, white H, green Cl, red Cu, Ni, or Pd).

An addition to the family : The introduction of β‐amino acid residues into a modified amyloid β peptide fragment resulted in well‐defined helical nanoribbons (see cryo‐TEM image) comprising β strands mainly oriented perpendicular to the ribbon axis. The nanoribbons order into a flow‐aligning nematic phase at higher concentration. The β‐strand nanoribbon structure is an addition to the known set of secondary structures adopted by β‐peptides.

By using a tridentate N^C^N-coordinating ligand, the luminescence of a cyclometallated Pt(II) complex can be shifted into the blue region, without the problematic drop-off in quantum yield observed for bidentate analogues. [PtL(6)X] {X = Cl or NCS and L(6) = 5-mesityl-1,3-di(2-pyridyl)-benzene} display similar luminescence in solution but, in the solid state, the packing of the molecules is different, with short PtPt interactions for X = NCS, leading to a red-shifted emission band. CEP‐18770, [(1R)‐1‐{[(2S,3R)‐3‐hydroxy‐2‐{[(6‐phenyl‐2‐pyridinyl)carbonyl]amino}butanoyl]amino}‐3‐methylbutyl]boronic acid, is a novel proteasome inhibitor, now under early clinical evaluation as an anticancer agent. To investigate its clinical pharmacokinetics, a high‐performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS) method was developed and validated to measure the drug in human plasma, based on simple protein precipitation with acetonitrile after the addition of irbesartan as internal standard. The method requires a small volume of sample (100 µl) and is rapid and selective, allowing good resolution of peaks in 5 min. It is sensitive, precise and accurate, with overall precision, expressed as coefficient of variation (CV%), always < 10.0%, accuracy in the range 93.8–107.7% and high recovery, close to 100%. The limit of detection is 0.01 ng/ml and the lower limit of quantitation (LLOQ) is 0.20 ng/ml. The assay was validated in the range from the LLOQ up to 50.00 ng/ml. This is the first method developed and validated for analyzing a proteasome inhibitor with a boronic‐acid‐based structure in human plasma. The method was successfully applied to study the pharmacokinetics of CEP‐18770 in cancer patients with solid tumors or multiple myeloma who had received the drug as a short intravenous bolus during the initial Phase I trial.
