Rapid assembly of matrix metalloprotease inhibitors using click chemistry

[reaction: see text] A panel of 96 metalloprotease inhibitors was assembled using "click chemistry" by reacting eight zinc-binding hydroxamate warheads with 12 azide building blocks. Screens of the bidentate compounds against representative metalloproteases provided discerning inhibition fingerprints, revealing compounds with low micromolar potency against MMP-7. The relative ease and convenience of the strategy in constructing focused chemical libraries for rapid in situ screening of MMPs is thereby demonstrated.     

Microarrays of tagged combinatorial triazine libraries in the discovery of small-molecule ligands of human IgG

A novel and highly diverse tagged triazine library incorporating a triethylene glycol-based linker was synthesized using an orthogonal combinatorial approach on the solid phase and covalently immobilized on a glass substrate as a small molecule microarray (SMM). The SMM was screened with a fluorophore-conjugated human IgG, and 4 novel binders from a library of 2688 compounds were identified from the fully spatially addressable array without the need for compound decoding. Using surface plasmon resonance (SPR) analysis, binding seen on the array was confirmed, and a binding constant as low as Kd = 2.02 x10(-6) M was measured.     

Zirconium(IV) complexes ofSchiff bases

Zirconium(IV)Schiff base derivatives have been synthesised by reacting zirconium isopropoxide with monofunctional bidentateSchiff bases in different stoichiometric ratios. The resulting derivatives of the type Zr(O-Isopr)₃(SB) and Zr(O-Isopr)₂(SB)₂, whereSB ^– is the anion of the correspondingSchiff baseSBH, have been isolated in almost quantitative yields. Their molecular weights have been determined ebullioscopically and their ir spectra recorded.

Zirkonium(IV)-Komplexe von Schiff-Basen

Zusammenfassung Es wurden Zirkonium(IV)-Schiff-Basen-Derivate in verschiedenen stöchiometrischen Zusammensetzungen über die Reaktion von Zirkoniumisopropoxid mit monofunktionellen zweizähnigenSchiff-Basen synthetisiert. Die Komplexe vom Typ Zr(O-Isopr)₃(SB) und Zr(O-Isopr)₂(SB)₂ [SB ^– als Anion derSchiff-BaseSBH] wurden in fast quantitativer Ausbeute erhalten. Es werden Strukturen vorgeschlagen, die auf ebullioskopisch bestimmten Molekulargewichten und den IR-Spektren basieren.

     

N-Substituted Amido Derivatives of Lanthanides

The synthesis of lanthanide amido complexes of the type Ln(CH₃CONR)₃ (I), (OPrⁱ)Ln(C₆H₅CONC₆H₅)₂ (II), and (OBu^t)Ln(C₆H₅CONC₆H₅)₂ (III) [where Ln = La, Pr, Nd; R = C₆H₅, p-NO₂C₆H₄, p-BrC₆H₄] are described. Infrared spectra indicate deprotonation of the secondary amide (anilide). Anion coordination is proposed as chelating bidenate ligands.
