CZE‐based assays for carbohydrate‐deficient transferrin (CDT) in which serum is mixed with an Fe(III) ion‐containing solution prior to analysis are effective approaches for the determination of CDT in patient samples. Sera of patients with progressed diseases, however, are prone to interferences comigrating with transferrin (Tf) that prevent the proper determination of CDT by CZE in these samples. The need of a simple and economic approach to immunoextract Tf from human serum prompted us to investigate the use of a laboratory‐made anti‐Tf spin column containing polyclonal rabbit anti‐human Tf antibodies linked to Sepharose 4 Fast Flow beads. This article reports extraction column manufacturing and column characterization with sera having normal and elevated CDT levels. The developed procedure was applied to a number of relevant hepatology and dialysis patient samples and could thereby be shown to represent an effective method for extraction and concentration of all Tf isoforms. Furthermore, lipemic sera were delipidated using a mixture of diisopropyl ether and butanol prior to immunoextraction. CDT could unambiguously be determined in all pretreated samples. 相似文献

The synthesis of two new 2,7-disubstituted phenanthrene-based bis oximes is described. The ability of these two compound for complexing heavy metal cations have been studied and complexation constants and complex stoichiometry for Cr3+ and Fe3+ complex have been determined. The fluorescent properties of ligand 2 make this compound able to act as a sensor able to discriminate between Cr3+ and Fe3+. Detection limits for these two cations have been evaluated. 相似文献

The gpdA-promoter-controlled exocellular production of glucose oxidase (GOD) by recombinant Aspergillus niger NRRL-3 (GOD3-18) during growth on glucose and nonglucose carbon sources was investigated. Screening of various carbon substrates
in shake-flask cultures revealed that exocellular GOD activities were not only obtained on glucose but also during growth
on mannose, fructose, and xylose. The performance of A. niger NRRL-3 (GOD3-18) using glucose, fructose, or xylose as carbon substrate was compared in more detail in bioreactor cultures.
These studies revealed that gpdA-promoter-controlled GOD synthesis was strictly coupled to cell growth. The gpdA-promoter was most active during growth on glucose. However, the unfavorable rapid GOD-catalyzed transformation of glucose
into gluconic acid, a carbon source not supporting further cell growth and GOD production, resulted in low biomass yields
and, therefore, reduced the advantageous properties of glucose. The total (endo- and exocellular) specific GOD activities
were lowest when growth occurred on fructose (only a third of the activity that was obtained on glucose), whereas utilization
of xylose resulted in total specific GOD activities nearly as high as reached during growth on glucose. Also, the portion
of GOD excreted into the culture fluid reached similar high levels (≅ 90%) by using either glucose or xylose as substrate,
whereas growth on fructose resulted in a more pelleted morphology with more than half the total GOD activity retained in the
fungal biomass. Finally, growth on xylose resulted in the highest biomass yield and, consequently, the highest total volumetric
GOD activity. These results show that xylose is the most favorable carbon substrate for gpdA-promoter-controlled production of exocellular GOD. 相似文献

Summary The selective binding of 3-indole acetic acid (IAA) alkali phosphatase (AP) to respective antibodies immobilized on pretreated titanium wires as electrodes (40×0.8 mm ø) is studied and the conditions are optimized with respect to selectivity and sensitivity, and the cross-reactivity of some related compounds is determined. Using these experiences, such wires are used as electrodes to detect selectively the methylester of IAA (IAA-Me). They are shown to behave best when the change of the electrode potential is measured against a similar reference electrode, coated with immobilized immuno globulin G (IgG) of the respective pre-immuno serum. A second coating with bovine serum albumin (BSA) is shown to prevent unspecific binding of IAA-Me. The sensitivity goes down to around 100 pmol/ml.

We perform quantum mechanics/molecular mechanics molecular dynamics simulations on the [Pt(2)(P(2)O(5)H(2))(4)](4-) (abbreviated PtPOP) complex; in water, dimethylformamide and ethanol. These are used to calculate the ground state X-ray absorption spectrum of the complex. The structural parameters from X-ray spectra are usually extracted using a fit of the experimental data. In such simulations the solvent is neglected meaning that any effect of the local environment will be compensated for by structural changes of the solute, leading to possible discrepancies in the extracted structural parameters. Our simulations show a significant solvent effect on the spectra, which has important implications for the structural analysis of molecules in solution. 相似文献

For the first time, the pressure and temperature dependence of a chemical reaction at the solid/solution interface is studied by scanning tunneling microscopy (STM), and thermodynamic data are derived. In particular, the STM is used to study the reversible binding of O(2) with cobalt(II) octaethylporphyrin (CoOEP) supported on highly oriented pyrolytic graphite (HOPG) at the phenyloctane/CoOEP/HOPG interface. The adsorption is shown to follow the Langmuir isotherm with P(1/2)(298K) = 3200 Torr. Over the temperature range of 10-40 °C, it was found that ΔH(P) = -68 ± 10 kJ/mol and ΔS(P) = -297 ± 30 J/(mol K). The enthalpy and entropy changes are slightly larger than expected based on solution-phase reactions, and possible origins of these differences are discussed. The big surprise here is the presence of any O(2) binding at room temperature, since CoOEP is not expected to bind O(2) in fluid solution. The stability of the bound oxygen is attributed to charge donation from the graphite substrate to the cobalt, thereby stabilizing the polarized Co-O(2) bonding. We report the surface unit cell for CoOEP on HOPG in phenyloctane at 25 °C to be A = (1.46 ± 0.1)n nm, B = (1.36 ± 0.1)m nm, and α = 54 ± 3°, where n and m are unknown nonzero non-negative integers. 相似文献
