CE and related methods are well-established techniques in the analysis of biomolecules, such as DNA and proteins. Even though CE is a rather good alternative to HPLC for the evaluation of the impurity profile and the enantiomeric purity of a drug, it is rarely applied. This might be due to the reservation of national licensing authorities and the pharmacopoeia commissions for several reasons. In this review containing some experimental data we report on several drug examples which demonstrate the superiority of CE over HPLC in special cases, i.e., in the analysis of antibiotics, amino acids and peptides, and the determination of enantiomeric purity. However, in order to make the CE techniques more suitable for pharmacopoeial purposes the general methods describing separation methods have to be complemented with the adjustment of the electrophoretic conditions being necessary to satisfy the system suitability criteria without fundamentally modifying the methods. In this paper, we report the results of molybdenum K-edge X-ray absorption studies performed on the oxidized and reduced active sites of the sulfite dehydrogenase from Starkeya novella. Our results provide the first direct structural information on the active site of the oxidized form of this enzyme and confirm the conclusions derived from protein crystallography that the molybdenum coordination is analogous to that of the sulfite oxidases. The molybdenum atom of the oxidized enzyme is bound by two Mo=O ligands at 1.73 A and three thiolate Mo-S ligands at 2.42 A, whereas the reduced enzyme has one oxo at 1.74 A, one long oxygen at 2.19 A (characteristic of Mo-OH2), and three Mo-S ligands at 2.40 A. 相似文献
Transition-metal-catalyzed trimethylenemethane (TMM) [3 + 2] cycloadditions provide direct routes to functionalized cyclopentanes. This reaction has been shown to be a highly chemo-, regio-, and diastereoselective process. We report a palladium-catalyzed asymmetric [3 + 2] trimethylenemethane (TMM) cycloaddition between 3-acetoxy-2-trimethylsilylmethyl-1-propene and various di- and trisubstituted olefins. Yields of exo-methylenecyclopentane products range from 59 to 99%, and enantiomeric excesses range from 58 to 92% ee. 相似文献
[RhCl(PR3)3] (R = Ph, Et) reacts with the potassium salt of 4-mercaptobenzoic acid to give a mixture of the monomeric and dimeric complexes, [Rh(SC6H4COOH)(PR3)3] and [{Rh(-SC6H4COOH)(PR3)2}2], respectively. With the labile PPh3 coligand, the dimer is the major product, while for the electron-richer coligand PEt3, the equilibrium is easily shifted to the monomer by the addition of excess PEt3. Phosphane dissociation and dimerization could be prevented by using the chelating coligand PPh(C2H4PPh2)2. [{Rh(-SC6H4COOH)(PPh3)2}2] (2b), [Rh(SC6H4COOH)(PEt3)3] (3a), and [Rh(SC6H4COOH){PPh(C2H4PPh2)2}] (4) were fully characterized by nuclear magnetic resonance and infrared spectroscopy, mass spectrometry, and elemental analysis. The molecular structures of 2b and 4 were determined by X-ray structure analysis. In solution, the lability of the phosphane ligands leads to the decomposition of 2b. One of the decomposition products, namely, the mixed-valent complex [{RhIRhIII(-SC6H4COO)(-SC6H4COOH)(SC6H4COOH)(PPh3)3}2] (5), was characterized by X-ray structural analysis. The dinuclear rhodium(III) complex [{Rh(-SC6H4COO)(SC6H4COOH)(PEt3)2}2] (6) was shown to be a byproduct in the synthesis of 3a, and this demonstrates the reactivity of the rhodium(I) complexes toward oxidative addition. The structurally characterized complexes 2b, 4, 5, and 6 show hydrogen bonding of the free carboxyl groups. 相似文献
A homogeneous catalyst system for the asymmetric cis-hydrogenation of 2,5-disubstituted furans leading to 2',3'-dideoxynucleoside analogues is described. Best enantioselectivities (ee values of up to 72%) were obtained with cationic rhodium complexes ligated by diphospholanes of the butiphane family. The selectivity of the hydrogenation was reversed by the addition of a base or a polar protic solvent in certain cases. Ferrocene- and proline-based systems gave significant, but lower, ee values. 相似文献
Nitrogen doping-induced changes in the electronic properties, defect formation, and surface structure of TiO2 rutile(110) and anatase(101) single crystals were investigated. No band gap narrowing is observed, but N doping induces localized N 2p states within the band gap just above the valence band. N is present in a N(III) valence state, which facilitates the formation of oxygen vacancies and Ti 3d band gap states at elevated temperatures. The increased O vacancy formation triggers the 1 x 2 reconstruction of the rutile (110) surface. This thermal instability may degrade the catalyst during applications. 相似文献
We use a predator–prey representation of an urban system to analyse how policy interventions can prevent the adverse effects of air pollution on people’s health. The number of residents is treated as prey variable, and particulate matter that consists of particles with a diameter of up to 10 micrometres (PM10) as predator variable. This representation allows integration of population trends and the effects of environmental interventions on the average level of PM10 concentration (which establishes a baseline for the potential health burden for residents). For the case of Graz, Austria, we illustrate the insights generated regarding the interdependency of market-based and technological pollution controls, and propose an indicator that assesses the cost of delayed interventions by counting additional premature deaths caused by polluted environments.
