Capillary electrochromatography (CEC) was employed as a rapid and high-efficiency method for the isocratic separation of all 20 important phenylthiohydantoin (PTH) amino acids, the end products of Edman degradation during N-terminal protein sequencing. For this purpose, 75 microm ID fused-silica capillaries were packed with standard 3 microm Hypersil octadecyl silica (ODS) particles using a two-step column fabrication process, which represents a fast, reliable and efficient means of producing long-term stable columns. The influence of solvent composition, pH, type of buffer cation, buffer concentration, and temperature on retention behavior of PTH amino acids was investigated. Same-day and day-to-day reproducibility of the retention times (over a period of two months) were found to be better than 3%. When comparing this new technique with traditional reversed phase-high performance liquid chromatography (RP-HPLC) methods applied in automated protein sequenators, CEC shows essentially shorter separation times and superior resolution.  相似文献

Military training commonly results in land degradation, but protocols for assessing and predicting long-term environmental impact are lacking. An ability to assess the impact of repeated disturbance and subsequent recovery is needed to balance training requirements against environmental quality. To develop methodology for assessing soil quality, a study evaluating disturbance resulting from tank maneuvers was initiated on Fort Riley Military Installation, Kansas. The objectives were to identify and quantify soil-quality indicators on two soil types exposed to controlled tank traffic. We examined physical, chemical, and biotic indicators after treatments were applied during wet and dry soil conditions. A randomized complete-block design, with three blocks per soil type and three treatments per block, was used. Treatments consisted of disturbance created by a 63-ton M1A1 tank making five passes in a figure-8 pattern during either dry or wet soil conditions. The M1A1 was operated at a speed of approximately 8 km/h. Control plots received no tank traffic. Soil-quality indicators evaluated were soil compaction, soil penetration resistance, rut depth, soil bulk density, soil texture, soil chemical composition, plant biomass, soil microbial diversity, and nematode and earthworm taxa. Soil-quality indicators were sampled within one week after tank disturbance. Preliminary data indicate soil-texture-dependent treatment effects (p  0.05) for bulk density and porosity. Bulk density increased and porosity decreased on trafficked areas, in the silt loam soil, but showed no change in the silty clay loam soil. Disturbance during wet soil conditions raised penetrometer resistance and gravimetric water content more than disturbance during dry soil conditions (p  0.05). A significant difference in disturbance was measured between the outside and inside portion of the same track (p  0.01 and 0.001, respectively). The outside track caused the greatest amount of disturbance, as measured by the height of the disturbed soil ridge above the track bed. Tank disturbance significantly reduced total vegetative biomass (p  0.05) compared with that of un-trafficked areas. Disturbance under wet soil conditions significantly reduced grass biomass (p = 0.040), whereas disturbance under dry soil conditions significantly reduced forb biomass (p = 0.0247) compared to un-trafficked areas. Total earthworm abundance (p = 0.011) was reduced by 82% when disturbance occurred during wet soil conditions regardless of soil type.  相似文献

We investigated the surface tension and surface composition of various mixtures of the two ionic liquids (ILs) 1-methyl-3-octyl-imidazolium hexafluorophosphate [C8C1Im][PF6] and 1,3-bis(polyethylene glycol)imidazolium iodide [(mPEG2)2Im]I in the temperature range from 230 to 370 K under ultraclean vacuum conditions. The surface tension was measured using a newly developed apparatus, and the surface composition was determined by angle-resolved X-ray photoelectron spectroscopy (ARXPS). In the pure ILs, the alkyl chains of [C8C1Im][PF6] and the PEG chains of [(mPEG2)2Im]I are enriched at the IL/vacuum interface. In the mixtures, a strong selective surface enrichment of the alkyl chains occurs, which is most pronounced at low [C8C1Im][PF6] contents. For the surface tension, strong deviations from an ideal mixing behaviour take place. By applying a simple approach based on the surface composition of the mixtures as deduced from ARXPS, we are able to predict and reproduce the experimentally measured temperature-dependent surface tension values with astonishingly high accuracy.  相似文献

Dielectric measurements on samples consisting of banana-shaped molecules with a polymorphism I/SmCPA/B7 have been performed for the first time. The dielectric increment for the reorientation of the molecules about the long axis, and the corresponding relaxation time, surprisingly decrease at the SmCPA/B7 transition. This effect is discussed in terms of the undulated structure which partially destroys the ferroelectric short range order. Decreasing temperature gives an even stronger hindrance for this reorientation and could explain controversial results obtained in the B7 phases of different samples. At lower temperatures a further mesophase was detected. This was additionally observed by calorimetric and atomic force microscopy methods.  相似文献

Summary The optical absorption and fluorescence of scattering samples is investigated theoretically and experimentally under the condition of steady-state and pulsed irradiation, taking into account inhomogeneous distribution of the absorbing species. Based on the Kubelka-Munk model, numerical iterative expressions for the steady-state and time-resolved light flues of excitation and emission are derived. The intensities of reflected, transmitted and forward or backward emitted radiation in layered samples with vertical concentration gradients are discussed. Modification of fluorescence spectra and decay curves due to self absorption, background absorption, secondary emission and time of flight dispersion are calculated and compared to experimental data of hydrocarbons adsorbed on polycrystalline silica gel.  相似文献
