Influence of amino acid side chain packing on Fe-methionine coordination in thermostable cytochrome C

Paramagnetic NMR and optical studies of the oxidized forms of mesophile Pseudomonas aeruginosa cytochrome c(551) and its quintuple mutant (F7A/V13M/F34Y/E43Y/V78I), and thermophile Hydrogenobacter thermophilus cytochrome c(552) demonstrated that the amino acid side chain packings in the protein interior influence the coordination bond between the heme iron and the axial methionine in the proteins. The strength of heme axial coordinations was found to correlate with the overall protein thermostability.     

Palladium-Catalyzed cross-Benzannulation of Aminoenynes with Diynes. Highly Regioselective Synthesis of Polysubstituted Anilines

Polysubstituted anilines were prepared by the palladium-catalyzed cross-benzannulation of conjugated aminoenynes 1-4 with diynes 8. The reaction proceeded in a highly regioselective manner under mild conditions, and the anilines were obtained as single regioisomers. Our method complements the well-known precedures for the preparation of polysubstituted anilines which are widely used in organic synthesis.     

Electrodeposition of aluminum–hafnium alloy from the Lewis acidic aluminum chloride-1-ethyl-3-methylimidazolium chloride molten salt

The electrochemistry of Hf(IV) and the electrodeposition of Al–Hf alloys were examined in the Lewis acidic 66.7–33.3 mol% aluminum chloride-1-ethyl-3-methylimidazolium chloride molten salt containing HfCl₄. When cyclic staircase voltammetry was carried out at a platinum disk electrode in this melt, the deposition and stripping waves for Al shifted to negative and positive potentials, respectively, suggesting that aluminum stripping is more difficult due to the formation of Al–Hf alloys. Al–Hf alloy electrodeposits containing ~13 at.% Hf were obtained on Cu rotating wire and cylinder electrodes. The Hf content in the Al–Hf alloy deposits depended on the HfCl₄ concentration in the melt, the electrodeposition temperature, and the applied current density. The deposits were composed of dense crystals and were completely chloride-free. The chloride-induced pitting corrosion potential of the resulting Al–Hf alloys was approximately +0.30 V against pure aluminum when the Hf content was above 10 at.%.     

Isomerization of aldehyde-2,4-dinitrophenylhydrazone derivatives and validation of high-performance liquid chromatographic analysis

The most widely used method for qualitative and quantitative analysis of carbonyl compounds is the 2,4-dinitrophenylhydrazine method through the formation of 2,4-dinitrophenylhydrazone derivatives. However, this method may cause an analytical error because 2,4-dinitrophenylhydrazones have both E- and Z-stereoisomers. Purified aldehyde-2,4-dinitrophenylhydrazone demonstrated only the E-isomer. However under UV irradiation and the addition of acid, both E- and Z-isomers were seen. The spectral patterns of Z-isomers were different from those of E-isomers and the absorption maximum wavelengths were shifted towards shorter wavelengths by 5-8 nm. An equilibrium Z/E isomer ratio was observed in 0.02-0.2% (v/v) phosphoric acid solutions. In the case of acetaldehyde- and propanal-2,4-dinitrophenylhydrazones, the equilibrium Z/E isomer ratios were 0.32 and 0.14, respectively. However, when irradiated with ultraviolet light at 364 nm, the isomer ratios were increased beyond this constant ratio and reached 0.55 and 0.33, respectively. Zero-order rates for decreases of aldehyde derivatives were observed under UV irradiation (364 nm), however the decreases of concentration were not observed in phosphoric acid solutions. The best method for the determination of aldehyde-2,4-dinitrophenylhydrazones by HPLC is to add phosphoric acid to both the sample and the standard solution, to form a 0.02-1% acid solution.     

Monitoring nano-flow rate of water by atomic emission detection using helium radio-frequency plasma

Recently, high-performance nano-scale flow pumping systems have been developed for micro and miniaturized analysis systems. A novel device capable of measuring and monitoring nanoliter scale flow rates has been required for the further development of the pumping system. In this study, an atomic emission detector using helium radio-frequency plasma (RFP-AED) was used for the measurement of the nanoliter scale flow rate of water by quantitatively detecting the emission from hydrogen in the water molecules. Monitoring nano-flow rates of water in the range up to 1.0 microl min(-1), and the change in the flow rate by the indication of the ratio of the emissions of H (656.3 nm) and He (667.8 nm) were successful. At present, the lowest flow rate that could be determined reproducibly was 4 nl min(-1) calculated as five times the standard deviation of the background noise. Additionally, similar evaluations for the deviation of each flow rate by using the RFP-AED and a flow-injection system were produced.     

Synthesis and Odor Evaluation of Stereoisomers of Imine Derivatives in Roasted Spotted Shrimp

All possible stereoisomers of imine derivatives 1 – 4 , which have the characteristic roast odor of seafood, were synthesized. As a result of odor evaluation of all isomers, we found that each isomer has a different and characteristic odor of roasted seafood.     

Structural variety of membrane permeable peptides

Peptide-mediated protein delivery into living cells has been attracting our attention. Among the peptides that have been reported to have carrier activity, the one from the human immunodeficient virus (HIV)-1 Tat has been most often used for the introduction of exogenous macromolecules into cells. We have shown that not only the Tat peptide, but also various arginine-rich peptides showed very similar characteristics in translocation, and the possible presence of ubiquitous internalization mechanisms among the arginine-rich peptides has also been suggested. These arginine-rich peptides includes ones derived from HIV-1 Rev and flock house virus coat proteins. The linear- and branched-chain peptides containing approximately 8 residues of arginine also show a similar ability. In this review, we present the structural variety of membrane permeable peptides and provide a survey of the findings on the translocation of these peptides through the cell membranes.     

Synthetic studies directed toward the assembly of the C-glycoside fragment of the telomerase inhibitor D8646-2-6

[reaction: see text] Construction and characterization of the C-glycosidic moiety of telomerase inhibitor D8646-2-6 (1) are described. This is the first example of the C-glycosylation using electron-poor aromatics, 4-hydroxypyrone, as a glycosyl acceptor. The glycosylation reaction and base-promoted isomerization affords desired beta-C-glycoside in a 61% overall yield.
