Pioglitazone is a medicine of thiazolidinedione (TZD) class with hypoglycemic (antihyperglycemic, antidiabetic) action. Pioglitazone binding to human serum albumin (HSA) was investigated at different temperatures (290, 300 and 310 K) by fluorescence spectroscopic method. Molecular docking study was also carried out besides the experiments. Experimental results revealed that pioglitazone have an ability to quench the intrinsic fluorescence of HSA tryptophan through a static quenching procedure. The binding constant was determined using Stern-Volmer modified equation and energy transfer mechanisms of quenching were discussed. Thermodynamic parameters were also calculated according to enthalpy changes dependence on different temperatures. According to the theoretical and experimental results, hydrogen bonding was found to play a major role in the interaction of pioglitazone with HSA. 相似文献

Multistep phosphorelay (MSP) pathways mediate a wide spectrum of adaptive responses in plants, including hormonal and abiotic stress regulations. Recent genetic evidence suggests both partial redundancy and possible functional cross-talk on the one hand and a certain level of specificity on the other. Here, we discuss recent achievements improving our understanding of possible molecular mechanisms of specificity in MSP. We consider a certain evolutionary conservation of ancestral two-component signalling systems from bacteria in a process of molecular recognition that, as we have recently shown, could be applied also to a certain extent in the case of plant MSP. Furthermore, we discuss possible roles of kinase and phosphatase activities, kinetics of both these enzymatic reactions, and phosphorylation lifetime. We include also recent findings on the expression specificity of individual members of MSP pathways and, finally, based on our recent findings, we speculate about a possible role of magnesium in regulation of MSP pathways in plants. All these mechanisms could significantly influence specificity and signalling output of the MSP pathways. 相似文献

Conjugates consisting of stigmasterol and L-phenylalanine, interconnected through short-chained dicarboxylic acyls by ester and amide bonds, respectively, were synthesized as potential low molecular weight/mass organic gelators (LMWGs/LMMGs). Their physico-chemical properties were subjected to investigation, especially their ability to form gels reversibly based on changes of the environmental conditions. Other self-assembly properties detectable by UV-VIS traces were measured in systems consisting of two miscible solvents (water/acetonitrile) with varying solvent ratios and using constant concentrations of the studied compounds. Partition and diffusion coefficients and solubility in water were calculated for the target conjugates. The conjugate 3a was the only compound from this series capable of forming a gel in 1-octanol. All three conjugates 3a-3c displayed supramolecular characteristics in the UV-VIS spectra. 相似文献

The application of palladium nanoparticles as electron-dense markers for labeling in both transmission and scanning electron microscopy requires their conjugation to a specific protein. The conjugation protocol described here includes the dihydrolipoic acid (DHLA) capping of Pd nanoparticles (8 nm equivalent diameter) and their subsequent covalent attachment to functional protein molecules such as streptavidin, protein A, or avidin. The single-step reaction was mediated using the cross-linking agent ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC). The final Pd conjugates were fully functional, as demonstrated by labeling of ultrathin resin sections of either bovine serum albumin or secretory granules of the salivary gland isolated from the partially fed female Ixodes ricinus tick. The results of bovine serum labeling were quantified, statistically evaluated, and compared with results obtained using commercially available gold particle conjugates (10 nm diameter). The highest values of labeling density were achieved using both streptavidin-Pd (106 ± 7 particles/μm2) and protein A-Au conjugates (130 ± 18 particles/μm2) compared to a commercial streptavidin-Au (66 ± 16 particles/μm2) and protein A-Pd conjugates (70 ± 11 particles/μm2). The concentrations of both DHLA and EDC, pH during conjugation, and finally thorough washing away of unbound proteins crucially influenced conjugation. 相似文献

Deactivation of UV-excited cytosine is investigated by non-adiabatic dynamics simulations, optimization of conical intersections, and determination of reaction paths. Quantum chemical calculations are performed up to the MR-CISD level. Dynamics simulations were performed at multiconfigurational level with the surface hopping method including four electronic states. The results show the activation of four distinct reaction pathways at two different subpicosecond time scales and involving three different conical intersections. Most trajectories relax to a minimum of the S(1) state and deactivate with a time constant of 0.69 ps mainly through a semi-planar conical intersection along the n(O)π* surface. A minor fraction deactivate along ππ* regions of the S(1) surface. Sixteen percent of trajectories do not relax to the minimum and deactivate with a time constant of only 13 fs. 相似文献

We present a periodic density-functional study of hydrogen adsorption and diffusion on the Si(110)-(1×1) and (2×1) surfaces, and identify a local reconstruction that stabilizes the clean Si(110)-(1×1) by 0.51 eV. Hydrogen saturates the dangling bonds of surface Si atoms on both reconstructions and the different structures can be identified from their simulated scanning tunneling microscopy/current image tunneling spectroscopy (STM/CITS) images. Hydrogen diffusion on both reconstructions will proceed preferentially along zigzag rows, in between two adjacent rows. The mobility of the hydrogen atom is higher on the (2×1) reconstruction. Diffusion of a hydrogen vacancy on a monohydride Si(110) surface will proceed along one zigzag row and is slightly more difficult (0.2 eV and 0.6 eV on (1×1) and (2×1), respectively) than hydrogen atom diffusion on the clean surface. 相似文献

Siliceous SBA-15 mesoporous molecular sieves were functionalized with different amounts of 3-aminopropyl-trimethoxysilane. To obtain a more detailed insight into the material properties of the prepared samples, their textural parameters were combined with results of thermal analysis. Adsorption isotherms of carbon dioxide on parent and functionalized SBA-15 were measured in the temperature range from 273 to 333 K. From the temperature dependence of CO(2) isotherms the isosteric adsorption heats of CO(2) were determined and discussed. Information about the surface energetic heterogeneity caused by tethered 3-aminopropyl groups were obtained from CO(2) adsorption energy distributions calculated using the theoretical CO(2) adsorption isotherms derived from the non-local density functional theory. The values of isosteric heats and the energy distributions of CO(2) adsorption detect highly energetic sites and enabled quantification of their concentrations. 相似文献

This paper shows the results of combined experimental and theoretical work that have unravelled the mechanism of ultrafast ejection of a methyl group from a cluster, the methyl iodide dimer (CH(3)I)(2). Ab initio calculations have produced optimized geometries for the dimer and energy values and oscillator strengths for the excited states of the A band of (CH(3)I)(2). These calculations have allowed us to describe the blue shift that had been observed in the past in this band. This blue shift has been experimentally determined with higher precision than in all previously reported experiments, since it has been measured through its effect upon the kinetic energy release of the fragments using femtosecond velocity map imaging. Observations of the reaction branching ratio and of the angular nature of the fragment distribution indicate that two main changes occur in A-band absorption in the dimer with respect to the monomer: a substantial change in the relative absorption to different states of the band, and, more importantly, a more efficient non-adiabatic crossing between two of those states. Additionally, time resolved experiments have been performed on the system, obtaining snapshots of the dissociation process. The apparent retardation of more than 100 fs in the dissociation process of the dimer relative to the monomer has been assigned to a delay in the opening of the optical detection window associated with the resonant multiphoton ionization detection of the methyl fragment. 相似文献

The adsorption of CO on low index copper single crystals in electrochemical environments has been investigated. The results, analysed through a combination of in situ infrared spectroscopy, DFT and cyclic voltammetry, reveal a unique adsorption behaviour when compared to previous studies on copper and the more widely studied noble metal surfaces. By employing small, weakly specifically adsorbed electrolytes, it is shown that carbon monoxide is adsorbed over a much wider electrode potential range than previously reported. The electrochemical Stark shift (δν/δE) observed is similar for the three Cu(hkl) surfaces examined despite different surface coverages. Most notably, however, is an electrochemical feature observed at ca. -1.0 V (vs. Ag/AgCl) on the (110) surface. It is proposed that this voltammetric feature arises from the reduction/oxidation of Cu(δ+) surface sites involved in the binding of carbon monoxide with the participation of the electrolyte anion. This provides additional specific sites for CO adsorption. DFT calculations support the proposed presence of low-coordination copper sites stabilised by electrolyte anions. An experimental electron transfer rate constant of 4.2 s(-1) to the Cu(δ+) surface sites formed was found. These new observations concerning the surface electrochemistry of CO on Cu indicate that the electrocatalytic behaviour of Cu electrodes in processes such as CO(2) reduction need to be re-evaluated to take account of the rich adsorption behaviour of CO, including the co-adsorption of the electrolyte anion to these sites. 相似文献
