Until now the study of pathogenic related proteins in grape juice and wine, performed by ESI-MS, LC/ESI-MS, and MALDI/MS, has been proposed for differentiation of varieties. In fact, chitinases and thaumatin-like proteins persist through the vinification process and cause hazes and sediments in bottled wines. An additional instrument, potentially suitable for the grape varieties differentiation, has been developed by MALDI/MS for the grape seed protein analysis. The hydrosoluble protein profiles of seeds extract from three different Vitis vinifera grape (red and white) varieties were analyzed and compared. In order to evaluate the environmental conditions and harvest effects, the seed protein profiles of one grape variety from different locations and harvests were studied. 相似文献

The complex-forming properties of monoethyl 8-quinolylmethylphosphonate (8-Hmqmp) towards palladium(II) ion have been investigated by reactions of the hydrochloride, 8-Hmqmp · HCl · H2O, and sodium salt, Na(8-mqmp) · 2H2O, of this monoester with palladium(II) halide compounds in aqueous solution over a wide pH range. Depending on pH and initial quinolinium and palladium salts, four types of complexes have been formed. Under acidic solution the ion-pair salt complexes [8-H2mqmp]2[PdX4] (1 and 2, pH < 3) and [8-H2mqmp]2[Pd2X6] (3 and 4, pH ∼ 3), with protonated quinoline ligand as cation and tetrahalopalladate or hexahalodipalladate complex as anion (X = Cl, Br), were isolated. By heating in methanol the chloro complexes 1 and 3 as well as bromo complexes 2 and 4 were converted into the quinolinium salt complexes, [8-H2mqmp][Pd(8-Hmqmp)X3], 5 and 6, respectively, containing as anion the quinoliniummethylphosphonatetrihalopalladate complex with palladium bonded at the phosphonic acid moiety. The chelate complex 7, [Pd(8-mqmp)2], with ligand bonded through the quinoline nitrogen and the deprotonated phosphonic acid oxygen and forming two seven-membered {N,O} chelate rings, was obtained in neutral and basic media. The complexes were identified and characterized by elemental analysis, magnetic and conductance measurements, spectroscopic studies (IR, 1H NMR, UV–Vis, positive/negative ion FAB MS) and thermal analysis (TG, DTA). As a preliminary screening for their biological activity, complexes were investigated for their ability to inhibit the cancer growth in vitro in the human KB and murine L1210 cell lines. The results obtained were compared with those obtained for the complexes of diethyl 8-quinolylmethylphosphonate (8-dqmp) and monoethyl 2-quinolylmethylphosphonate (2-Hmqmp), and structural factors that determine the complex activity were discussed. 相似文献

Amber is a fossil resin constituted of organic polymers derived through complex maturation processes of the original plant resin. A classification of eight samples of amber of different geological age (Miocene to Triassic) and geographical origin is here proposed using direct mass spectrometric techniques, i.e. laser desorption ionization (LDI), atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization (APPI), in order to obtain a fingerprint related to the amber origin. Differences and similarities were detected among the spectra with the four methods, showing quite complex spectra, full of ionic species in the mass range investigated (up to m/z 2000). The evaluation required statistical analysis involving multivariate techniques. Cluster analysis or principal component analysis (PCA) generally did not show a clear clustering with respect to the age of samples, except for the APPI method, which allowed a satisfying clustering. Using the total ion current (TIC) obtained by the different analytical approaches on equal quantities of the different amber samples and plotted against the age, the only significant correlation appeared to be that involving APPI. To validate the method, four amber samples from Cretaceous of Spain were analyzed. Also in this case a significant correlation with age was found only with APPI data. PCA obtained with TIC values from all the MS methods showed a fair grouping of samples, according to their age. Three main clusters could be detected, belonging to younger, intermediate and older fossil resins, respectively. This MS analysis on crude amber, either solid or extract, followed by appropriate multivariate statistical evaluation, can provide useful information on amber age. The best results are those obtained by APPI, indicating that the quantity of amber soluble components that can be photoionized decreases with increasing age, in agreement with the formation of highly stable, insoluble polymers. 相似文献

The Claisen rearrangement is a well-known process occurring in condensed phase. In the gas-phase protonated allyl phenyl ethers, propargyl phenyl ethers, and N-allyl aniline produced by positive ion chemical ionization undergo Claisen rearrangement. This reaction has been observed even in the case of odd-electron molecular ions. Phenyl allenyl ether molecular ions actually undergo Claisen rearrangement, producing intense [M - CO](+*) ions. In this investigation, the behavior of protonated benzyloxy indole and some of its derivatives, obtained in electrospray conditions, is described. Low-energy MS/MS experiments carried out on [M + H](+) species show CO loss and an unexpected water loss: both can be justified only by the occurrence of Claisen rearrangement. Deuterium labeling experiments confirm this mechanism. The influence of different substituents in the indole moiety is discussed. 相似文献

In an investigation devoted to the search for plasma markers for colorectal cancer (CRC), carried out by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry, a series of overexpressed peptides were identified in the plasma of patients. Among them the peptide with molecular weight 903 Da was the most abundant one, with a mean +/- (SD) relative abundance of 37 +/- 17% and a frequency over 60%. Interestingly, also in plasma samples of ten subjects affected by familial adenomatous polyposis (FAP), the peptide with molecular weight 903 was overexpressed. In this investigation, MALDI/MS/MS experiments were carried out on the ion at m/z 904 detected in the MALDI mass spectra of CRC and FAP patients. The data analysis by SwissProt.2007.01.09 indicates that this peptide is due to the sequence RPPGFSPF, found in the kininogen-1 precursor, which is an alpha-2-thiol proteinase inhibitor. In the case of subjects affected by a particular FAP syndrome, the MALDI/MS/MS spectra were quite different from those obtained from CRC and FAP patients. In fact, two sequences have been evidenced: RPPGFSPF belonging to kininogen-1 precursor, and PRKSSSSR belonging to Forkhead box protein 01A. 相似文献

Silica nanoparticles (about 15 nm diameters), which contain a derivative of 6-methoxy-8-(p-toluensulfonamido)-quinoline (TSQ) as a Zn(II) fluorescent probe covalently linked to the silica network, were prepared and studied as Zn(II) fluorescent chemosensors. The systems selectively detect Zn(II) ions in water rich solutions with a submicromolar sensitivity: 0.13 microM concentrations of Zn(II) can be measured with the only interference of Cu(II) and Cd(II) ions. Compared with free TSQ, the nanoparticles based systems have the advantage that they can be employed in aqueous solutions without aggregation problems while at the same time, they maintain a similar Zn(II) affinity and sensing ability. Addition of a second, substrate insensitive, fluorophore to the particles leads to the realization of a ratiometric sensor. 相似文献

The measurement of trace-element concentration in soil, sediment and waste, is generally a combination of a digestion procedure
for dissolution of elements and a subsequent measurement of the dissolved elements. “Partial” and “total” digestion methods
can be used in environmental monitoring activities. To compare measurement results obtained by different methods, it is crucial
to determine and to maintain control of the bias of the results obtained by these methods. In this paper, ICP-MS results obtained
after matrix digestion with modified aqua regia (HCl+HNO3+H2O2) method and two “total” digestion methods (microwave aqua regia+HF and HNO3+HF) are compared with those obtained by instrumental neutron activation analysis, a non-destructive analytical method for
the determination of the total mass concentrations of inorganic components in environmental matrices. The comparison was carried
out on eight agricultural soil samples collected in one test area and measured by k0-INAA and ICP-MS to determine As, Co, Cr, Sb and Zn mass concentration. The bias of results for As, Cd, Co, Cr, Cu, Ni, Pb,
Sb and Zn of the three digestion methods were assessed using selected measurement standards. This paper highlights that the

The aim of this study was a non-destructive evaluation of composites through their behaviour under thermal stimulation. Such behaviour was monitored by infrared thermography. Several specimens were fabricated involving: glass/epoxy with inclusion of foreign materials; carbon/epoxy with backdrilled holes; carbon/epoxy with impact damage; Glare® failed in bearing way. The obtained results prove that infrared thermography is capable of detecting the materials inhomogeneities and/or damage listed above. In particular, lock-in thermography is capable of supplying useful information about: the distribution of the adhesive thickness in composite structures; the distribution of the paint thickness; the behaviour under load of aluminium layers and glass fibres in Glare®. 相似文献
