An HPLC-UV method was established for the determination of metformin and buformin in biological fluids. Metformin was not retained on particles packed in conventional solid-phase extraction cartridges; in contrast, buformin was retained too firmly and not eluted with a solvent for recovery. However, both drugs were retained on particles that had been treated with an ion-pair reagent of heptanesulfonate or dodecylsulfate and recovered almost completely. The recovered fraction was subjected to HPLC on a pentafluorophenylpropyl column which was suitable for the determination of both biguanides in serum and in urine. Limits of quantitation were low enough for clinical use, and reproducibility was high with an RSD of 0.9-2.3%. HPLC on a conventional octadecyl column was suitable only for the determination of buformin in serum since interfering peaks appeared on the chromatograms of urine samples. The method was applied to analysis of some clinical specimens.  相似文献

Neonatal exposure to androgen induces developmental abnormalities in the male reproductive system. To investigate whether neonatal exposure affects spermatogenesis in juvenile and pubertal testis, Sprague-Dawley rat pups were given androgen or various androgenic endocrine disruptors by a single injection on the day of birth at concentrations ranging between 4 mm to 200 mm, and sacrificed on day 21 (juvenile) or 50 (puberty). The testes were weighed and examined histologically at each stage. Further, the metabolites of steroidogenesis were analyzed using normal-phase high-performance liquid chromatography. Neonatal exposure significantly reduced testis weights and steroidogenesis of juveniles. Neonatal exposure to testosterone and dihydrotestosterone still suppressed pubertal steroidogenesis, although testis weight was completely restored during puberty.  相似文献

A carbonyl ylide cycloaddition approach to the squalene synthase inhibitors zaragozic acids A and C is described. The carbonyl ylide precursor 8 was synthesized starting from di-tert-butyl D-tartrate (47) via an eleven-step sequence involving the regioselective reduction of the mono-MPM (MPM=4-methoxybenzyl) ether 48 with LiBH4 and the diastereoselective addition of sodium tert-butyl diazoacetate to alpha-keto ester 10. The reaction of alpha-diazo ester 8 with 3-butyn-2-one (40) in the presence of a catalytic amount of [Rh2(OAc)4] gave the desired cycloadduct 59 as a single diastereomer. The dihydroxylation of enone 59 followed by sequential transformations permitted the construction of the fully functionalized 2,8-dioxabicyclo[3.2.1]octane core 5. Alkene 79 derived from 5 serves as a common precursor to zaragozic acids A (1) and C (2), since the elongation of the C1 alkyl side chain can be attained by olefin cross-metathesis, especially under the influence of Blechert's catalyst (85).  相似文献

Photoinduced electron-transfer processes of alkyl-inserted ferrocene-trimethylene-oligothiophene-fullerene (Fc-tm-nT-C60) linked triads and directly linked ferrocene-oligothiophene-fullerene(Fc-nT-C60) triads were investigated using time-resolved fluorescence and transient absorption spectroscopic methods. In nonpolar solvent, the energy-transfer (EN) process occurred from 1nT* to C60 for both triads, without forming the charge-separated (CS) state. In polar solvent, the initial CS state, Fc-tm-nT(*+)-C60(*-), was formed via Fc-tm-nT-1C60 after the EN process from 1nT by photoexcitation of the nT moiety and after direct photoexcitation of the C60 moiety. For Fc-tm-nT(*+)-C60(*-), the positive charge shifted from the nT(*+) moiety to the Fc moiety, producing the final CS state, Fc(*+)-tm-nT-C60(*-), which lasted for 22-330 ns by changing nT from 4T to 12T. For Fc-nT-C60 in polar solvent, the CS state, in which the radical cation is delocalized on both Fc and nT moieties ((Fc-nT)(*+)-C60(*-)), was formed immediately after direct photoexcitation of the nT and C60 moieties. The lifetimes of (Fc-nT)(*+)-C60(*-) were estimated to be 0.1-50 ns by changing nT from 4T to 12T. The longer lifetimes of Fc(*+)-tm-nT-C60(*-) than those of (Fc-nT)(*+)-C60(*-) are caused by the insertion of the trimethylene chain to prevent the pi-conjugation between the Fc and nT moieties. The lifetimes for Fc(*+)-tm-nT-C60(*-) and (Fc-nT)(*+)-C60(*-) are prolonged by changing nT from 4T to 12T. For the charge-recombination process of Fc(*+)-tm-nT-C60(*-), the damping factor was evaluated to be 0.10 A(-1). For (Fc-nT)(*+)-C60(*-), the oxidation potentials of the nT moieties control the electron-transfer process with reflecting stabilization of the radical cations of the nT moieties.  相似文献

The Lanczos homogeneous filter diagonalization method has been employed to compute the HOCl ro-vibrational states for a range of total angular momenta (J = 0, 1, 5, 10, 11, 20, 30) on a newly developed ab initio potential energy surface by Nanbu et al. (J. Theor. Comput. Chem. 2002, 1, 263). For such computationally challenging calculations, a parallel computing strategy has been incorporated into our method to perform the matrix-vector multiplications. For the computed low bound states, a spectroscopic assignment has been made and the widely used approximate adiabatic rotation method has been tested for the broad range of total angular momenta for this deep-well system. Comparison of experimental results with exact quantum mechanical calculations for the selected far-infrared transitions involving the range of total angular momenta has been made possible for the first time.  相似文献

In order to develop a chiral stationary phase (CSP), which has even higher separation ability than the corresponding commercially available crown ether based CSP (OA-8000 having a pseudo-18-crown-6 ether with an OMe group as a selector), chemically bonded type CSP having a phenolic OH group on a crown ring was developed. Normal mobile phases with or without acid additive can be used with this OH type CSP in contrast to the conventional OMe type CSP which has a neutral chiral selector. Enantiomers of 25 out of 27 amino compounds, including 20 amino acids, 5 amino alcohols, and 2 lipophilic amines, were efficiently separated on a column with this CSP. Nine amino compounds out of 27 were separated with better separation factors than the corresponding OMe type CSP. It is noteworthy that the chromatography on this CSP exhibited excellent enantiomer-separations for amines and amino alcohols when triethyl amine was used as an additive in the mobile phase. Comparison of enantiomer separation ability on this OH type of CSP and on the OMe type of CSP and correlation between the enantioselectivity in chiral chromatography and that of the corresponding model compounds in solution imply that the chiral separation arose from chiral recognition in host guest interactions.  相似文献

The rheological behavior and microstructure of shear-thinning suspensions of core-shell structured carboxylated latex particles were examined. The steady shear viscosity of the suspension increased with increasing dissociation of the carboxyl groups or increasing particle concentration, however the critical shear stress sigma(c) and inter-particle distance xi of the microstructure did not change. With increasing particle diameter, sigma(c) increased and xi decreased. These results were consistent with a Brownian hard sphere model, in which competition exists between the bulk mass transfer due to the applied field and diffusion of the particles. We confirmed that sigma(c) depends on xi, as expressed by sigma(c) = 3kT/4pi xi3. This relationship is consistent with the dynamics of a Brownian hard sphere model with particle diameter xi. Thus the dynamics of shear-thinning suspensions of core-shell particles can be explained by a Brownian thermodynamic model.  相似文献

A phosphine-free catalytic system [Pd(OAc)2-Cu(OAc)2-air] induced a substrate-specific carbonylation of amines in boiling toluene under CO gas (1 atm). Symmetrical N,N'-dialkylureas were obtained by the carbonylation of primary amines. N,N,N'-Trialkylureas were selectively formed by addition of a secondary amine to the above reaction vessel. Secondary amines did not give tetraalkylureas. However, dialkylamines with a phenyl group on their alkyl chains, such as N-monoalkylated benzylic amine or phenethylamine derivatives, underwent a direct aromatic carbonylation to afford five- or six-membered benzolactams. In the carbonylation, the chelation effect or steric repulsion between Pd(II) and the meta-substituent in the ortho-palladation and the ring sizes of cyclopalladation products that were formed prior to carbonylation were found to generate good site selectivity and increase the reaction rate. In contrast, carbonylation of omega-arylalkylamines with a hydroxyl group gave neither ureas nor benzolactams but instead produced 1,3-oxazolidinones smoothly. Hydrochlorides of amines also underwent carbonylation to afford the corresponding amides under the conditions used. This procedure made it possible to prepare ureas of amino acid esters and N-alkylcarbamates in practical yields.  相似文献
