Application of the 19F NMR technique to observe binding of the general anesthetic halothane to human serum albumin.

19F NMR techniques were employed to characterize the binding property of the widely used general anesthetic halothane with human serum albumin (HSA). It was found that 19F(1H) NOE and 2D 1H-19F HOESY experiments detected intermolecular NOEs between halothane 19F and HSA protons. Measurements of the diffusion coefficients for halothane were also carried out by 1H and 19F NMR, indicating the interaction of halothane with HSA. The present results indicate that these techniques are very suitable to identify a fluorine-containing ligand binding with a protein receptor in the drug-discovery process.     

Stabilization of enhanced chirality from pyrene-containing l-glutamide lipid in methyl methacrylate by photo-induced polymerization

Enhanced CD spectra based on chirally-oriented structures of pyrene-containing L-glutamic acid-derived lipid were observed in polymerizable monomers and CD strength was maintained after photo-induced polymerization of the monomer solvents.     

Bis(pentafluorophenyl) disulfide as a hydrogen abstractor and an electron acceptor from the resulting radical intermediate

The pentafluorobenzenethiyl radical is an efficient hydrogen abstractor from an activated methylene or methine group and bis(pentafluorophenyl) disulfide is an efficient electron acceptor from the resulting radical intermediate. Thus benzyl-OTBS ether was easily converted into the corresponding pinacol, and 2-phenyl-1,3-dioxanes are converted into the monobenzoates of diols.     

Study on Segmental Motion and Ion Binding in Polyelectrolyte Solutions by Ultrasonic Spectroscopy

The ultrasonic absorption spectra of aqueous solutions of polyacrylate (PA), polyphosphate (PP), and polystyrenesulfonate (PSS), neutralized by tetramethylammonium hydroxide (TMAOH), were measured. The effects of addition of tetramethylammonium chloride (TMACl) and sodium chloride (NaCl) to the polyelectrolyte solutions were investigated in the frequency range from 500 kHz to 100 MHz. Two ultrasonic relaxation processes due to the local segmental motions were observed. The relaxation frequency for TMAPP solution decreased as the ionic strength was increased by the addition of TMACl. For the other two polymer solutions, the ionic strength did not affect the relaxation spectra. The addition of NaCl led to an increase of the ultrasonic absorption, which was ascribed to ion binding. The ultrasonic absorption due to the ion binding was estimated by subtracting the contribution of the segmental motion from the measuring ultrasonic spectra. The volume changes accompanying the ion binding for polyacrylate and polyphosphate salts were estimated to be 5 and 8 cm³-mol^?1, respectively.     

Injectable Hydrogel for Sustained Protein Release by Salt‐Induced Association of Hyaluronic Acid Nanogel

A hyaluronic acid‐based anionic nanogel formed by self‐assembly of cholesteryl‐group‐bearing HA is designed for protein delivery. The HA nanogel spontaneously binds various types of proteins without denaturation, such as recombinant human growth hormone, erythropoietin, exendin‐4, and lysozyme. The HA nanogel shows unique colloidal properties, in particular that an injectable hydrogel is formed by salt‐induced association of the HA nanogel. A pharmacokinetic study in rats shows that an in situ gel formulation, prepared by simply mixing rhGH and HA nanogel in phosphate buffer, maintains plasma rhGH levels within a narrow range over one week. Therefore, HA nanogels offer a simple method for easy formulation of therapeutic proteins and are effective for sustained protein release systems.

     

Reductive coupling of N-methoxycarbonyl lactams with benzophenone and 9-fluorenone by low-valent titanium

The reductive coupling of N-methoxycarbonyl lactams with benzophenone by Zn-TiCl₄ in THF gave cross-coupled products as cyclic α-diphenylidene-N-methoxycarbonylamines and ring-opening α,α-diphenyl-α-hydroxy-ω-(N-methoxycarbonyl)amino ketones selectively depending on the reduction conditions. The reductive coupling of N-methoxycarbonyl lactams with 9-fluorenone by Zn-TiCl₄ gave cyclic α-(9H-fluoren-9-ylidene)-N-methoxycarbonylamines preferentially irrespective to the conditions.     

Absolute configuration of pterocidin,a potent inhibitor of tumor cell invasion from a marine-derived Streptomyces

During the course of screening natural products for the inhibitors of tumor cell invasion, pterocidin, a linear polyketide with a δ-lactone terminus, was rediscovered from a Streptomyces strain of a marine sediment-origin. A series of J-based configuration analyses and NOESY analysis, coupled with chemical derivatization and chiral anisotropy analysis, established the absolute stereochemistry of five asymmetric centers in this compound.     

Dielectrically controlled resolution (DCR) of 3-aminopiperidine via diastereomeric salt formation with N-tosyl-(S)-phenylalanine

A useful key intermediate for the dipeptidyl peptidase-4 (DPP-4) inhibitor, 3-aminopiperidine 1, was successfully resolved with an enantiomerically pure resolving agent, N-tosyl-(S)-phenylalanine 2, to give both stereoisomers (R)-1 and (S)-1 as a less-soluble diastereomeric salt with (S)-2, via a dielectrically controlled resolution (DCR) phenomenon.
