Using a highly sensitive flow‐type 27 MHz quartz crystal microbalance, we could detect a small mass change during stepwise and alternating one‐sugar transfer of glucuronic acid (GlcA) and N‐acetylgalactosamine (GalNAc) to an acceptor, catalyzed by chondroitin polymerase from Escherichia coli strain K4 (K4CP), and analyze the elongation mechanism of K4CP. K4CP was found to bind strongly to a chondroitin acceptor (Kd=0.97 μM ). Although the binding affinity and the catalytic rate constant for each monomer were considerably different, the apparent catalytic efficiency (kcat/Km) was similar (6.3×104 M ?1 s?1 for GlcA transfer and 3.4×104 M ?1 s?1 for the GalNAc transfer). This is reasonable for the smooth alternating elongation of GlcA and GalNAc on the acceptor. The title compound (2) has been synthesized, the structure and the physical properties of which are investigated by X-ray, spectroscopic analyses, and MO calculations. There exists charge transfer (CT) interaction between the tropylium ion and the facing phenyl ring. From the pKR+ value of 2, it can be clear that the cation 2 is stabilized by the CT interaction. We have developed the transparent photoactive TiO2 thin film coated on soda lime glass (SLG) by sol-gel process. Titanium dioxide thin films coated on SLG exhibit lower photocatalytic activity due to the thermal diffusion of Na ion from the SLG substrate. Thin SiO2 film precoating is very effective to prevent the thermal diffusion of Na ion. We have evaluated the photocatalytic decomposition of gaseous acetaldehyde and the photo-induced surface wettability of TiO2 films with and without SiO2 precoating layer. As expected, the TiO2 film on SiO2/SLG is more photoactive to decompose acetaldehyde than that on SLG. However, as for wettability conversion, there was little difference in the conversion rate between TiO2 film without SiO2, and TiO2 film with SiO2. Different dependence of Na ion diffusion on two kinds of photo-induced reaction on TiO2 is discussed based on the difference of the photo-induced reaction mechanism. 相似文献

To study the thrombolytic effect of tissue plasminogen activator (t-PA) on cerebral emboli, we characterized cerebral embolization in stroke-prone spontaneously hypertensive rats (SHRSPs) and Wistar Kyoto rats (WKYs). [125I]Fibrin clot particles (20-100 microns diameter) were injected twice at an interval of 90 min into the left internal carotid artery of WKYs and SHRSPs. After each injection, spontaneous embolus dissolution was monitored with a gamma-ray detector placed on the head of the embolic rats. Embolus dissolution was spontaneously generated in 15 min after the injection of fibrin clots. In WKYs, 21% and 42% of the clots were dissolved 30 and 90 min after the second embolization, respectively. On the other hand, the spontaneous embolus dissolution in SHRSPs was significantly lower than that of WKYs, indicating that the endogenous fibrinolytic ability of SHRSPs is less potent than that of normotensive rats. The intravenous administration of t-PA at doses of 75, 250 and 750 micrograms/kg caused a dose-dependent embolus dissolution in SHRSPs. Furthermore, systematically applied t-PA produced embolus dissolution without causing systemic plasminogen activation, fibrinogen breakdown or bleeding. In conclusion, the intravenous administration of t-PA produces selective embolus dissolution without systemic fibrino(geno)lysis in a cerebral embolic SHRSP. 相似文献

Selectivity of 15 stationary phases was examined, either commercially available or synthesized in-house. The highest selectivity factors were observed for solute molecules having different polarizability on the 3-(pentabromobenzyloxy)propyl phase (PBB), followed by the 2-(1-pyrenyl)ethyl phase (PYE). Selectivity of fluoroalkane 4,4-di(trifluoromethyl)-5,5,6,6,7,7,7-heptafluoroheptyl (F13C9) phase is lowest among all phases for all compounds except for fluorinated ones. Aliphatic octyl (C8) and octadecyl (C18) phases demonstrated considerable selectivity, especially for alkyl compounds. While PBB showed much greater preference for compounds with high polarizability containing heavy atoms than C18 phase, F13C9 phase showed the exactly opposite tendency. These three stationary phases can offer widely different selectivity that can be utilized when one stationary phase fails to provide separation for certain mixtures. The retention and selectivity of solutes in reversed-phase liquid chromatography is related to the mobile phase and the stationary phase effects. The mobile phase effect, related to the hydrophobic cavity formation around non-polar solutes, is assumed to have a dominant effect on retention upon aliphatic stationary phases such as C8, C18. In a common mobile phase significant stationary phase effect can be attributed to dispersion interaction. Highly dispersive stationary phases such as PBB and PYE retain solutes to a significant extent by (attractive) dispersion interaction with the stationary phase ligands, especially for highly dispersive solutes containing aromatic functionality and/or heavy atoms. The contribution of dispersion interaction is shown to be much less on C18 or C8 phases and was even disadvantageous on F13C9 phase. Structural properties of stationary phases are analyzed and confirmed by means of quantitative structure-chromatographic retention (QSRR) study. 相似文献

The copolymerization of ethylene and 5‐hexen‐1‐ol pretreated with trimethylaluminium was performed using [ethylene(1‐indenyl)(9‐fluorenyl)]zirconium dichloride/methylaluminoxane as the catalyst. The 5‐hexen‐1‐ol unit in the copolymer could be increased to about 50 mol‐% with increasing [5‐hexen‐1‐ol/ethylene[ ratio. 13C NMR analysis proved that the poly(ethylene‐co‐(5‐hexen‐1‐ol)) containing 50 mol‐% of 5‐hexen‐1‐ol units is an almost alternating copolymer. 相似文献

An experiment on the investigation of optical diffraction radiation (ODR) from a slit target as a possible tool for noninvasive electron beam-size diagnostics has been performed at the KEK accelerator test facility. The experimental setup has been installed at the diagnostics section of the extraction line. We have performed the first incoherent ODR observation from a slit target. The measured angular distributions are in reasonable agreement with the theoretical expectation. The beam-size effect onto the ODR angular pattern has been observed. Moreover, the sensitivity to the beam size as small as 14 microm has been achieved. 相似文献
