The reaction of [Ni(aet)2] with [CoCl2(R,R-chxn)2]+ (aet = 2-aminoethanethiolate, R,R-chxn = 1R,2R-cyclohexanediamine) in water gave a CoIIINiIICoIII trinuclear complex, DeltaRRDeltaRR-[Ni(Co(aet)(2-)(R,R-chxn))2]4+ ([1a]4+), in which two cis(S)-[Co(aet)2(R,R-chxn)]+ units are linked by a central NiII ion through sulfur bridges. The two CoIII units in [1a]4+ uniformly adopt the Delta configuration, which is induced by the chirality of the terminal R,R-chxn ligands. The central NiII ion in [1a]4+ was replaced by a PdII ion to produce an analogous CoIIIPdIICoIII trinuclear complex, DeltaRRDeltaRR-[Pd(Co(aet)2(R,R-chxn))2]4+ ([2a]4+), with retention of the Delta configuration. When racemic R,R/S,S-chxn was employed instead of R,R-chxn, not only the chirality about two CoIII centers but also the chirality about two chxn ligands was unified in the S-bridged trinuclear structure, leading to the selective formation of a pair of enantiomers, DeltaRRDeltaRR/LambdaSSLambdaSS-[M(Co(aet)2(chxn))2]4+ (M = NiII ([1b]4+) and PdII ([2b]4+)). The stereochemical and spectroscopic features of these complexes are discussed on the basis of the electronic absorption, CD, and NMR spectroscopies, along with the crystal structures of [1a]4+ and [2a]4+.  相似文献

A method has been developed for highly sensitive determination of haloperidol in human serum involving a simple extraction procedure followed by gas chromatographic separation. Target components were separated from the extracting solvents with a Van den Berg type solventless sample injector before introduction Into a DB-1 capillary separation column. A surface ionization detector (SID), which has highly selective sensitivity for Substituted amines, was employed for quantitation using bromperidol as an internal standard. Chloroform proved to be the best extracting solvent, yielding a quantitative detection limit of 5 ng/ml (S/N = 2). Comparison of the response to target compounds obtained by the SID, FTD (flame thermionic detector), and FID (flame ionization detector) showed the SID to be superior.  相似文献

The partial molar volumes (Va) of 1-alkanols (carbon number, m=5, 6, 7) in - and -cyclodextrin (CD) solutions at 5.00 mmol kg–1 have been determined as a function of alkanol concentration (Ca) between 293.2 and 308.2 K by using a dilatometer. It has been observed that with an increase in Ca, Va increased in -CD solution but decreased in -CD solution, asymptotically to a value of Va in CD-free water. The dependence of Va on Ca provided the binding constant (K) of 1:1 complex, the volume change in complex formation, and the partial molar volume of complex itself. The complex formation mechanism has been discussed on the basis of these values and their carbon number dependences in the respect of geometric behavior, hydrophobic interaction, and van der Waals interaction. It is concluded that the CD cavity in water is not rigid but flexible for fitting in nicely with guest molecule.  相似文献

[Reaction: see text]. The synthesis of neuropeptide Y antagonist 1, currently under clinical investigation for the treatment of obesity, is described. The convergent synthesis from trans-spirolactone carboxylic acid intermediate 2a and aminopyrazole 3 is predicated on a stereoselective route to the former. The coupling reaction of ethyl 4-oxocyclohexanecarboxylate (10a) with lithiated isonicotinamide 11 was investigated in detail, but even optimized conditions only provided a 45:55 ratio of trans:cis isomers (12a:12b). While selective crystallization schemes were developed to isolate the thermodynamically less stable trans isomer 2a, improved stereocontrol was subsequentially achieved by the application of ketene chemistry. The ketene formation and quench was investigated under a variety of conditions aimed at maximizing the trans:cis ratio. Reacting a mixture of carboxylic acids 2a and 2b with POCl3 in THF, followed by concomitant addition of tert-butyl alcohol in the presence of TMEDA at 35 degrees C provided a 4:1 ratio of trans:cis tert-butyl esters (18a:18b) via in situ ketene formation. Ester hydrolysis, followed by selective crystallization of undesired 2b as the HCl salt, led to isolation of 2a in 47% overall yield. Aminopyrazole intermediate 3 was synthesized via the condensation reaction of 2-fluorophenylhydrazine hydrochloride (4a) with acrylonitrile derivative 5 in 65-70% yield. Coupling of advanced intermediates 2a and 3b via activation with thionyl chloride gave a 92% yield of 1.  相似文献

Concentration determination of meropenem, a carbapenem antibiotic, using a capillary electrophoresis method by direct injection of serum samples without any pretreatment is described herein. Sodium tetraborate (25 mM)-sodium hydroxide (0.1 M) containing sodium dodecyl sulfate (90 mM) is used as a run buffer (pH 10.0). Meropenem is detected at its absorption maximum at 297 nm. Migration time of meropenem is approximately 7.2 min, and the detection limit of the assay is 2.0 mg/L at a signal-to-noise ratio of 3.0. The relative standard deviations of intra- and interassay accuracies are 3.43-8.87% and 4.28-8.54%, respectively, at a nominal concentration of 6.3-100.0 mg/L, and their recovery rates are 94-111% and 92-105%, respectively.  相似文献

Glass bead/x‐ray fluorescence spectrometry of the sludge incineration ashes generated in sewage processing was developed for the determination of ten major components (Na2O, MgO, Al2O3, SiO2, P2O5, K2O, CaO, TiO2, MnO, Fe2O3) and five minor elements (Zn, Cu, Cr, As, Pb). Sewage sludge ashes consisted of rock‐forming minerals and phosphate crystals that had been used for phosphorus removal. Ash samples were melted and molded with lithium tetraborate to 35 mm diameter glass disks in a Pt–Au crucible. Analytical results of ten major components and five minor elements agreed well with the recommended values of a phosphate rock standard reference material (NIST SRM 694). Elemental compositions of sewage sludge ash from seven sewage‐processing plants in Japan were determined using this method. Concentrations of Fe2O3, SiO2, and CaO, along with loss of ignition in sewage sludge ash mutually differed among the sewage‐processing plant products. Seasonal variations in concentrations of ten major components and five minor components of ash samples produced from October 2001 to September 2002 were determined using the proposed method. Concentrations of SiO2increased with the inflow of gravel by rainfall, thereby decreasing concentrations of P2O5 originating from excreta and microorganisms. The techniques of chemical ligation have attracted great attention as an alternative to enzymatic joining of DNA ends. Here we introduce the photoligation of anthracene-modified ODN conjugates through anthracene cyclodimer formation. The effect of the positions and the kinds of single base mismatch on the template was evaluated using eight templates with one-base displacements. We found out that the yield of the ligation was affected by mispairing in a position-dependent manner. Such results would be attributed to the disruption of the local structure at the ligation site.  相似文献

During the process development for multistep synthesis of Rosuvastatin calcium several impurities were obtained along with the final Rosuvastatin calcium. Out of this; synthesis of impurity A (acetone adduct) a minor impurity of Rosuvastatin calcium (3R,5S,6E)-7-[4-(4-fluorophenyl)-2-[[(2-hydroxy-2-methylpropyl)sulfonyl(methyl)amino]-6-(1-methylethyl)-pyrimidin-5-yl]-3,5-dihydroxyheptenoicacid hemicalcium salt, is described. The synthesis of impurity A has been accomplished in 6 steps; starting from formation of β-hydroxy sulfonamide as the key intermediate and followed by using convenient routes with overall yield of 13.5%. The target compound can be used as the reference substance of impurity of the Rosuvastatin calcium.  相似文献
