A column‐switching liquid chromatography/electrospray ionization tandem mass spectrometry to determine paclitaxel and its metabolites, 6α‐hydroxypaclitaxel and p‐3′‐hydroxypaclitaxel, in human plasma was developed. The analytical system had a Shim‐Pack MAYI‐ODS (10 × 4.6 mm i.d.) trapping column with deproteinization ability that concentrates analytes and removes water‐soluble components. This method covered a linearity range of 5–5000 ng/mL of concentrations in plasma for paclitaxel, a range of 0.87–870 ng/mL for 6α‐hydroxypaclitaxel and a range of 0.87–435 ng/mL for p‐3′‐hydroxypaclitaxel. The intra‐day precision and inter‐day precision of analysis were less than 11.1%, and the accuracy was within ±14.4% at concentrations of 5, 50, 500 and 5000 ng/mL for paclitaxel, 0.87, 8.7, 87 and 870 ng/mL for 6α‐hydroxypaclitaxel, and 0.87, 8.7, 87 and 435 ng/mL for p‐3′‐hydroxypaclitaxel. A potent inhibitor for Vibrio cholerae neuraminidase (VCNA) was developed by using a novel two‐step strategy, a target amino acid validation using mechanism‐based labeling information, and a potent inhibitor search using a focused library. The labeling information suggested the hidden dynamics of a loop structure of VCNA, which can be a potential target of the novel inhibitor. A focused library composed of 187 compounds was prepared from a 9‐azide derivative of 2,3‐dehydro‐N‐acetylneuraminic acid (DANA) to interrupt the function of the loop of the labeled residues. Inhibitor 3c showed potent inhibition properties and was the strongest inhibitor with FANA, a N‐trifluoroacetyl derivative of DANA. Validation studies of the inhibitor with a detergent and a Lineweaver–Burk plot suggested that the 9‐substitution group would interact hydrophobically with the target loop moiety, adding a noncompetitive inhibition property to the DANA skeleton. This information enabled us to design compound 4 having the combined structure of 3c and FANA. Compound 4 showed the most potent inhibition (Ki=73 nM , mixed inhibition) of VCNA with high selectivity among the tested viral, bacterial, and mammal neuraminidases.  相似文献

The electrochemical and photochemical reduction of the fully methylated derivative of gold meso-tetrakis-(4-pyridyl)porphine (AuTMPyP) in homogeneous solution gives not only a π-radical anion but also its successive product, phlorin, by disproportionation. The electrostatic fixation of AuTMPyP in a Nafion matrix inhibits the latter undesired reaction against effective charge separation, which is explained by the diffusion constant lower by 8 order of magnitude during electrolytic reduction of AuTMPyP compared to that in homogeneous aqueous solution.  相似文献

We evaluated the intracellular NAD(P)H:quinone oxidoreductase (NQO) activity of single HeLa cells by using the menadione–ferrocyanide double-mediator system combined with scanning electrochemical microscopy (SECM). The double-mediator system was used to amplify the current response from the intracellular NQO activity and to reduce menadione-induced cell damage. The electron shuttle between the electrode and menadione was mediated by the ferrocyanide/ferricyanide redox couple. Generation of ferrocyanide was observed immediately after the addition of a lower concentration (10 μM) of menadione. The ferrocyanide generation rate was constant for 120 min. At a higher menadione concentration (100 μM), the ferrocyanide generation rate decreased within 30 min because of the cytotoxic effect of menadione. We also investigated the relationship between intracellular reactive oxygen species or glutathione levels and exposure to different menadione concentrations to determine the optimal condition for SECM with minimal invasiveness. The present study clearly demonstrates that SECM is useful for the analysis of intracellular enzymatic activities in single cells with a double-mediator system.  相似文献

The visible fluorescence spectrum of single flavoprotein at a temperature of 1.5 K has been measured by one-photon excitation. The flavoprotein studied was a photoswitchable enzyme, photoactivated adenylyl cyclase. The time course of the spectrum revealed a structural change occurring at a rate of 10(-3) s(-1) around hydrogen bonds at the flavin cofactor binding site.  相似文献

In this study, we introduce the double-barrel carbon probe (DBCP)—a simple, affordable microring electrode—which enables the collection and analysis of single cells independent of cellular positioning. The target cells were punctured by utilizing an electric pulse between the two electrodes in DBCP, and the cellular lysates were collected by manual aspiration using the DBCP. The mRNA in the collected lysate was evaluated quantitatively using real-time PCR. The histograms of single-cell relative gene expression normalized to GAPDH were fit to a theoretical lognormal distribution. In the tissue culture model, we focused on angiogenesis to prove that multiple gene expression analysis was available. Finally, we applied DBCP for the embryonic stem (ES) cell-derived cardiomyocytes to substantiate the capability of the probe to collect cells, even from high-volume samples such as spheroids. This method achieves high sensitivity for mRNA at the single-cell level and is applicable in the analysis of various biological samples independent of cellular positioning.

We study theΛ formation cross sections andΛ polarizations based on the Regge pole amplitudes in theπ ?+12C→Λ+X(inclusive) reactions and the exclusive kaon channel reactions atp π =4 GeV/c. We found that the experimental data of inclusive cross sections can be reproduced qualitatively well by our calculation. As for the polarization, we have the experimental data that were reported by two different groups, although they have opposite sign and different magnitudes. Our calculated results do not agree with either set of data. We show that the sign of polarization is sensitive to the outgoingΛ kinetic energy in the inclusive reactions and to the outgoing kaon angle in the exclusive kaon channel reactions.  相似文献
