A method was developed for semi-preparative scale enantioseparation of racemic homochlorcyclizine (HCZ) by high performance liquid chromatography (HPLC) on Chiralcel OD column. The best resolution was achieved using an eluent composed of n-hexane plus 0.2 M isopropylamine. By this method, about 5.0 mg of racemic HCZ could be resolved completely in one run. The optical purity of the enantiomers were both greater than 99.9%. The studies of antihistamine activity on guinea pig ileum demonstrated that l-HCZ is significantly more potent than d- and racemic HCZ. The pharmacokinetics of d- and l-HCZ after oral administration to rats also differed. The successful resolution of racemic HCZ permits comparison of the pharmacokinetics and antihistamine activity of the enantiomers.  相似文献

Dendrimer-Au nanocomposites are prepared in aqueous solutions using poly(amidoammine)dendrimers (PAMAM) (generation 2, 3, and 5) and poly(propyleneimine)dendrimers (PPI)(generation 2, 3, and 4) by wet chemical NaBH(4) method. The Au nanoparticles thus obtained are 2-4 nm in diameter for both dendrimers and no generation dependence on the particle size is observed, whereas the generations of the dendrimers are increased as stabilization of Au-nanoparticles is achieved with lower dendrimer concentrations. Studies of the reduction reaction of 4-nitrophenol using these nanocomposites show that the rate constants for the PAMAM dendrimers (generations 2 and 3) are higher than those for the PPI dendrimers (generations 2 and 3), while a distinct difference in the rate constants is not seen for the PAMAM dendrimer (generation 5) or the PPI dendrimer (generation 4). In addition, the rate constants for the reduction of 4-nitrophenol involving all the dendrimers decrease with increases in dendrimer concentrations.  相似文献

Electron-transfer reduction of molecular oxygen (O2) by the phenolate anion (1-) of a vitamin E model, 2,2,5,7,8-pentamethylchroman-6-ol (1H), occurred to produce superoxide anion, which could be directly detected by a low-temperature EPR measurement. The rate of electron transfer from 1- to O2 was relatively slow, since this process is energetically unfavourable. The one-electron oxidation potential of 1- determined by cyclic voltammetric measurements is sufficiently negative to reduce 2,2-bis(4-tert-octylphenyl)-1-picrylhydrazyl radical (DOPPH*) to the corresponding one-electron reduced anion, DOPPH-, suggesting that 1- can also act as an efficient radical scavenger.  相似文献

Aqueous titanate sols were prepared by reactions of titanium tetraisopropoxide (TIP) with -hydroxycarboxylic acids in water. IR and Raman spectra, and elemental analyses of the precipitates obtained from the sols revealed that the carboxylates were chelated to titanium but the Ti species were polymerized to form a cluster that had a colloidal nature. Spin-coating of titania (TiO2) thin films from these sols was also examined. Interestingly, it was found that (004) preferentially oriented anatase films with refractive index of 2.54 were obtained from TIP-lactic acid-NH3 (1:1:1, molar ratio) aqueous sol. This crystallographic orientation was characteristic of the TIP-lactic acid-NH3 (1:1:1) system, and was not affected by the kinds of substrates used and the heating rate of the film. TEM observation indicated that small anatase grains had already formed at 200°C. Therefore, the crystallographic orientation might depend strongly on the structure of the chemical species of the precursor solutions.  相似文献

The enzymatic activity of diaphorase (Dp) immobilized on a solid substrate was characterized using a scanning electrochemical microscope (SECM) with shear force feedback to control the substrate-probe distance. The shear force between the substrate and the probe was monitored with a tuning fork-type quartz crystal and used as the feedback control to set the microelectrode probe close to the substrate surface. The sensitivity and the contrast of the SECM image were improved in the constant distance mode (distance, 50 nm) with the shear force feedback compared to the image in the constant height mode without the feedback. By using this system, the SECM and topographic images of the immobilized diaphorase were simultaneously measured. The microelectrode tip used in this study was ground aslant like a syringe needle in order to obtain the shaper topographic images. This shape was also effective for avoiding the interference during the diffusion of the enzyme substrates.  相似文献

The radical reactions of N‐(2‐halogenoalkanoyl)‐substituted anilines (anilides) of type 1 have been investigated under various conditions. Treatment of compounds 1a – 1o with Bu3SnH in the presence of (2,2′‐azobis(isobutyronitrile) (AIBN) afforded a mixture of the indolones (oxindoles) 2a – 2o and the reduction products 5a – 5o (Table 1). In contrast, the N‐unsubstituted anilides 1p – 1s, 1u , and 1v gave the corresponding reduction products exclusively (Table 1). Similar results were obtained by treatment of 1 with Ni powder (Table 2) or wth Et3B (Table 3). Anilides with longer N‐(phenylalkyl) chains such as 6 and 7 were inert towards radical cyclization, with the exception of N‐benzyl‐2‐bromo‐N,2‐dimethylpropanamide ( 6b ), which, upon treatment with Ni powder in i‐PrOH, afforded the cyclized product 9b in low yield (Table 4). Upon irradiation, the extended anilides 6, 7, 10 , and 11 yielded the corresponding dehydrobromination products exclusively (Table 5).  相似文献

We have recently demonstrated a one-step process to fabricate silver-polypyrrole (PPy) coaxial nanocables (Chen, A.; Wang, H.; Li, X. Chem. Commun. 2005, 14, 1863). The formation process of silver-PPy coaxial nanocables is discussed in this article. It was found from the results of TEM and SEM images that large numbers of silver atoms were formed when AgNO3 was added to a pyrrole solution. Then silver atoms transform to silver-PPy nanosheets with regular morphology, which will connect together to be more stable. Silver-PPy nanocables will be able to grow at the expense of the silver-PPy nanosheets. Poly(vinylpyrrolidone) (PVP) plays crucial roles in this process: as a capping agent to form silver nanowires, and as a dispersant of pyrrole monomers, which can influence the site at which pyrrole monomer exists. On the basis of experimental analysis, the possible mechanism was proposed. Because of the effect of PVP, silver ions and pyrrole monomers are apt to be adsorbed at the [111] and [100] facets of silver nanosheets, respectively. Obvious polymerization will take place on the boundary of the [111] and [100] facets. The PPy layer stays stable on the [100] facets. Meanwhile, newly formed silver atoms and silver nanosheets will further ripen and grow on the [111] facets. In a word, the morphology of final products and the formation process are determined by the reaction site between AgNO3 and the pyrrole monomer, which is influenced by PVP.  相似文献

Morphological change of gold-dendrimer nanocomposites by laser irradiation   总被引:1,自引:0,他引:1  
Gold-dendrimer nanocomposites are prepared in aqueous solutions in the presence of poly(amidoamine)dendrimers (PAMAM) (generation 3 and 5) or poly(propyleneimine)dendrimers (PPI) (generation 3 and 4) by wet chemical NaBH(4) method. Thus prepared gold-dendrimer nanocomposites are irradiated by laser at 532 nm. UV-vis absorption spectroscopy and transmission electron microscopy reveal that the gold nanoparticles grow with the laser irradiation time as well as the fluence of the laser; in particular, the gold nanoparticles prepared at lower concentrations of PAMAM dendrimer as well as lower generations of PAMAM grow significantly. On the other hand, in the case of PPI dendrimers, the gold nanoparticles hardly grow by irradiation. In addition, dynamic light-scattering measurements show that the laser irradiation markedly promotes the association of the gold-PAMAM G3 dendrimer nanocomposites compared to that of the gold-PAMAM G5 dendrimer nanocomposites, while the sizes of association for the gold-PPI G3, G4 dendrimer nanocomposites hardly change by laser irradiation.  相似文献
