Six reducing monosaccharides (mannose, galactose, fucose, glucose, xylose, and arabinose) were derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP) and chiral resolution of these racemic PMP-monosaccharides was studied by ligand-exchange CE using borate anion as a central ion of the chiral selector and (S)-3-amino-1,2-propanediol (SAP) as a chiral selector ligand. PMP-mannose, PMP-galactose and PMP-fucose were successfully enantioseparated. Lowering the capillary temperature increased the resolution of PMP-mannose system, but decreased that of PMP-galactose and PMP-fucose systems. Whereas the maximum resolution was obtained at pH 8.9 in the PMP-mannose system, resolution increased gradually with pH in the PMP-galactose and PMP-fucose systems. Expecting the formation of the ternary borate complexes with SAP and PMP-monosaccharide in the CE experiments, the optimized structures of the borate diastereomers were obtained by semiempirical molecular orbital calculations to discuss the structural difference of the diastereomers in connection with the enantioseparation behaviors.  相似文献

We have developed a method to analyze the substrate transport of ATP-binding cassette (ABC) transporters, which are associated with drug resistance in tumor cells. Our microfluidic method is well suited to the single-vesicle estimation of substrate transport and the rapid drug screening of ABC transporters. Using this method, we have demonstrated, for the first time, the analysis of substrate transport by a single transporter and performed drug-inhibition experiments in less than 3 h.  相似文献

Isoprenoids are a diverse group of molecules found in all organisms, where they perform such important biological functions as hormone signaling (e.g., steroids) in mammals, antioxidation (e.g., carotenoids) in plants, electron transport (e.g., ubiquinone), and cell wall biosynthesis intermediates in bacteria. All isoprenoids are synthesized by the consecutive condensation of the five-carbon monomer isopentenyl diphosphate (IPP) to its isomer, dimethylallyl diphosphate (DMAPP). The biosynthetic pathway for the formation of IPP from acetyl-CoA (i.e., the mevalonate pathway) had been established mainly in mice and the budding yeast Saccharomyces cerevisiae. Curiously, most prokaryotic microorganisms lack homologs of the genes in the mevalonate pathway, even though IPP and DMAPP are essential for isoprenoid biosynthesis in bacteria. This observation provided an impetus to search for an alternative pathway to synthesize IPP and DMAPP, ultimately leading to the discovery of the mevalonate-independent 2-C-methyl-D-erythritol 4-phosphate pathway. This review article focuses on our significant contributions to a comprehensive understanding of the biosynthesis of IPP and DMAPP.  相似文献

We overcome the difficulties in pulling long draglines from spiders, twist bundles of dragline filaments, and succeed in preparing violin strings. The twisting is found to change the cross section shapes of filaments from circular to polygonal and to optimize the packing structure with no openings among filaments providing mechanically strong and elastic strings. The spider string signal peaks of overtones for the violin are relatively large at high frequencies, generating a soft and profound timbre. Such a preferable timbre is considered to be due to the unique polygonal packing structure which provides valuable knowledge for developing new types of materials.  相似文献

Compared with a nano-sized particle, dynamics of a micron-sized particle in a liquid is often associated with sedimentation (or floating) due to its relatively large mass. The motion of more than two particles is dominated by the hydrodynamic interactions, which are known to persist over a fairly long range, e.g., several millimeters, in suspensions. The particle size may be obtained from the dynamic ultrasound scattering (DSS) technique by the analysis of velocity fluctuations, whose origin is believed to take root in the particle-number fluctuations among temporally formed domains involving collective motion of particles with a certain cut-off length. In this study, such collective particle motion in highly turbid solutions was visualized by means of the phase-mode DSS technique with a single element transducer. Quantitative agreement between the velocity fluctuations obtained by the phase- and conventional amplitude-mode analyses was confirmed, followed by examination of the concentration and the particle size dependences on the dynamic structures induced by the long-ranged interactions. It was found that the phase mode-DSS was a promising method to evaluate the time-dependent structures of the micro-particles in highly turbid suspensions.  相似文献

The software release game developed in Ref. 1 is reconsidered in the framework of a two-person nonzero-sum game of timing. More precisely, noisy-type software release strategies are derived in closed form under two different criteria as well as an alternative silent-type software release strategy. Our method overcomes the fatal problem in Ref. 1 and has an advantage on computational tractability. Also, the method can be extended directly to obtain noisy-type strategies.  相似文献

We calculate the elastic scattering of a long chain in a polymer melt during the process of relaxation after a sudden deformation, i.e., the return to equilibrium with time, while the deformed shape is maintained. The scattering thus depends on the duration t of the relaxation, as well as on the scattering vector, and is a physical quantity characteristic of the dynamics of long chains. The reptation model of de Gennes is used as developed for deformed melts by Doi and Edwards: the chain is confined by other chains in a tube, from which disengagement by the ends is the only way of renewing its configuration. The tube diameter is taken as a parameter. We give both an analytic form and numerical evaluations. On comparison with calculations based on the Rouse model, in which the chain is assumed to be free in a viscous medium, it is seen that the experimental data (given elsewhere) should enable one to distinguish between the two models.  相似文献

In this paper, we discuss the 2-stage output procedure of a finite dam under the condition that water must be released by a fixed time. From this standpoint, the reservoir model we consider is subject to a sample path constraint and has a more general cost function than the earlier contributions. We analytically derive explicit formulas for the long-run average and the expected total discounted costs for an infinite time span and numerically calculate the optimal control policy. Finally, the optimal policy is compared with one by Zuckerman [1] and the effect of the fixed release time is discussed further.  相似文献

alpha-Ester-substituted 1,3-dienylphosphonates 7 and 8, prepared by the Knoevenagel condensation, underwent intramolecular [2 + 2] cycloaddition in the presence of Lewis acid to form bicyclo[4.2.0] (26-57% yield) and bicyclo[3.2.0]skeleton (14-38% yield), respectively. Similar treatment of homologous 1,3-dienylphosphonate 11 and 1,3,5-trienylphosphonate 12 resulted in the formation of ionone derivatives (30-94% yield). The intramolecular cycloaddition reaction was applicable to several conjugated dienes bearing an ester group.  相似文献
