Capillary electrophoretic separation of 60 mer single-stranded DNA (ssDNA) and a single-base-substituted ssDNA was demonstrated using a size- and composition-controlled poly(ethylene glycol)-oligodeoxyribonucleotide block copolymer (PEG-b-ODN) as an affinity ligand. Under appropriate conditions, PEG-b-ODN and ssDNA with a complementary sequence formed a reversible complex via hybridization during the electrophoresis, while the copolymer did not interact with the single-base-substituted ssDNA. The copolymer's PEG length determined the electrophoretic mobility of the ssDNA; upon formation of the complex, the electrically neutral PEG added hydrodynamic friction to ssDNA. Simultaneously using two types of PEG-b-ODN copolymers whose PEG segments were of different lengths, we achieved the complete separation of the 60 mer ssDNA, its single-base-substituted ssDNA, and impurities. This method was sensitive enough to quantify a slight amount (approximately 1%) of the single-base-substituted ssDNA. The present results suggest that our approach is applicable to quantitative detection of minor genotypes.  相似文献

A biomimetic synthesis of naturally occurring lactams rubrobramide, flavipucine, and isoflavipucine is described. The key step is a regioselective Darzens reaction between isobutyl glyoxal and an α‐bromo‐β‐ketoamide. The construction of the core tricyclic ring system of rubrobramide was achieved by a cascade reaction in a single step from an α,β‐epoxy‐γ‐lactam. Furthermore, the absolute configuration of naturally occurring (+)‐rubrobramide was determined by vibrational circular dichroism. (±)‐Flavipucine and (±)‐isoflavipucine were synthesized from an epoxyimide, which was prepared by reaction of isobutyl glyoxal with a protected α‐bromo‐β‐ketoamide. Deprotection of the epoxyimide and formation of the pyridone ring gave (±)‐flavipucine, which was converted into (±)‐isoflavipucine by thermal isomerization.  相似文献

We study the global Cauchy problem for the mass critical nonlinear Schrödinger equations. We prove the global existence of analytic solutions in both space and time variables for sufficiently small and exponentially decaying Cauchy data. The method of proof depends on the Leibniz rule for the generator of pseudo-conformal transforms at the L 2 critical level.  相似文献

Based on an amphotericin B (AmB) ion-channel model where the close proximity of neighboring molecules is effected by interaction between carboxyl and amino groups, we prepared covalent dimers of AmB connected between these functionalities. While directly connected and short-tethered derivatives (2 and 3) lacked the activities, dimer 4 with a longer linker revealed K+ ion flux activity, suggesting that some distance and/or flexibility between the carboxyl and amino groups in adjacent molecules is required for the formation of ion-permeable complex in biomembranes.  相似文献

This paper reconsiders the capacity problemthrough a statistical approach to code acquisition. First, aDS/CDMA system model is reviewed. Second, properties of thecounting method for code acquisition are discussed. It is provedthat the method can acquire the target user's code under anynumber of interference users that simultaneously transmit theirsignals, and that the method can guarantee a considerable precisionin code acquisition. Third, an observation time necessary forcode acquisition is given from a statistical discussion, andthe original counting method is modified into a simpler one.It is concluded that the capacity of the DS/CDMA communicationsystem can be set by the ‘bit error-rate-based capacityor signal-to-noise ratio-based capacity’, i.e. ‘post-acquisition-basedcriterion’, rather than the ‘acquisition-based capacity’.  相似文献

2‐(3,4,5,6‐Tetrafluoro‐2‐hydroxyphenyl)imidazo[1,2‐a]pyridine ( 1 ) emits long wavelength light around 540 nm both in polar and in nonpolar solvents. Zn2+ perchlorate in acetonitrile causes the intermediate wavelength emission around 430 nm, which is ascribed to the species where the imidazole nitrogen atom and the phenolate oxygen atom bridge Zn2+. In the presence of Hg2+ and Al3+ perchlorates, short wavelength emission around 370 nm is strongly increased and this fluorescent enhancement is attributable not to the coordination of Hg2+ and Al3+ to 1 but to the formation of the salt of perchloric acid of 1 .  相似文献

The total synthesis of borrelidin has been achieved. The best feature of our synthetic route is macrocyclization at C11-C12 for the construction of an 18-membered ring after esterification between two segments. A detailed examination of the macrocyclization led us to the samarium(II) iodide-mediated intramolecular Reformatsky-type reaction as the most efficient synthetic approach. The two key segments were synthesized through regioselective methylation, directed hydrogenation, stereoselective Reformatsky-type reaction, and MgBr2.Et2O-mediated chelation-controlled allylation.  相似文献

Aqueous solutions of three kinds of surface active ionic liquids composed of the 1-alkyl-3-methylimidazolium cation have been investigated by means of surface tension and electrical conductivity measurements at room temperature (298 K). The surface tension measurements provided a series of parameters, including critical micelle concentration (cmc), surface tension at the cmc (gammacmc), adsorption efficiency (pC20), and effectiveness of surface tension reduction (Picmc). In addition, with application of the Gibbs adsorption isotherm, maximum surface excess concentration (Gammamax) and minimum surface area/molecule (Amin) at the air-water interface were estimated. The effect of sodium halides, NaCl, NaBr, and NaI, on the surface activity was also investigated. It was found that both the pC20 and the Picmc were rather larger than those reported for traditional ionic surfactants and the cmc values were somewhat lower than those for typical cationic surfactants, alkyltrimethylammonium bromides, and comparable to typical anionic surfactants, sodium alkyl sulfates. These results demonstrate that the surface activity of long-chained imidazolium IL is somewhat superior to that of conventional ionic surfactants.  相似文献

Abstract— Using dark-grown 3 day-old Sorghum bicolor seedlings, photorepair of cyclobutane pyrimidine dimers (CPD) by CPD-photoIyase and pyrimidine-(6–4)pyrimidinone photoproducts ([6–4]PP) by [6–4]PP-photolyase was studied in vivo and in vitro. The two types of DNA photoproducts were measured with specific monoclonal antibodies. Sorghum seedlings have different photorepair enzymes for CPD and [6–4]PP. The CPD-photolyase was found to be increased in response to light with maximal efficiency in the UVA/blue spectral range, whereas [6–4]PP-photolyase was not influenced by light.  相似文献
