Cananodine is a guaipyridine alkaloid with activity against liver cancer. Cananodine was synthesized using a remarkable intramolecular opening of a trisubstituted epoxide as the key step in construction of the seven-membered carbocycle of the target. Large (10 × 10 cm) sheets of surface-enhanced Raman spectroscopy (SERS) active polymer have been prepared by stabilising metal nanoparticle aggregates within dry hydroxyethylcellulose (HEC) films. In these films the aggregates are protected by the polymer matrix during storage but in use they are released when aqueous analyte droplets cause the films to swell to their gel form. The fact that these “Poly-SERS” films can be prepared in bulk but then cut to size and stored in air before use means that they provide a cost effective and convenient method for routine SERS analysis. Here we have tested both Ag and Au Poly-SERS films for use in point-of-care monitoring of therapeutic drugs, using phenytoin as the test compound. Phenytoin in water could readily be detected using Ag Poly-SERS films but dissolving the compound in phosphate buffered saline (PBS) to mimic body fluid samples caused loss of the drug signal due to competition for metal surface sites from Cl ions in the buffer solution. However, with Au Poly-SERS films there was no detectable interference from Cl and these materials allowed phenytoin to be detected at 1.8 mg L−1, even in PBS. The target range of detection of phenytoin in therapeutic drug monitoring is 10–20 mg L−1. With the Au Poly-SERS films, the absolute signal generated by a given concentration of phenytoin was lower for the films than for the parent colloid but the SERS signals were still high enough to be used for therapeutic monitoring, so the cost in sensitivity for moving from simple aqueous colloids to films is not so large that it outweighs the advantages which the films bring for practical applications, in particular their ease of use and long shelf life.  相似文献

The infrared spectra (4,000–30 cm?1) of the gas and solid and the Raman spectrum of liquid 2,2-difluoroethanol as well as variable temperature infrared spectra of krypton/xenon solutions have been recorded. From all these data, two (Gg and Tg) out of the five possible stable conformers have been confidently identified. The order of the stabilities has been predicted to be Gg > Tg > Gt > Gg′ > Tt by utilizing ab initio MP2 (full) and DFT (B3LYP method) calculations, where the first indicator (capital letter) is in reference to rotation around the C–C bond (G = gauche or T = trans) and the second one (small letter) refers to the orientation of the hydroxyl group. The percentage of the minor conformer Tg, at ambient temperature, is estimated to be (16 ± 3%). The optimized geometries, fundamental frequencies, infrared intensities, Raman activities, and depolarization values as well as centrifugal distortion constants have been obtained from ab initio and density functional theory calculations by utilizing a variety of basis sets as well as those with diffuse functions. By utilizing the previously reported microwave rotational constants for two isotopomers of the Gg conformer combined with ab initio MP2(full)/6-311+G(d,p) predicted structural values, adjusted r 0 parameters have been obtained. The determined heavy atom distances (Å) for the Gg conformer are: C1–C2 = 1.510(3), C2–F4 = 1.371(3), C2–F5 = 1.362(3), C1–O3 = 1.412(3) Å and angles ∠O3C1C2 = 111.0(5), ∠F4C2C1 = 108.8(5), ∠F5C2C1 = 109.8(5), τF4C2C1O3 = 63.5(5), τF5C2C1O3 = 179.1(5)°. Barriers of internal rotation have been obtained and vibrational assignments for the Gg and Tg conformers are given. The five predicted centrifugal distortion constants compared to the experimental values are in reasonable agreement except for ?K, which appears to be in error. The results are discussed and the structural parameters compared to the corresponding ones for 2-fluoroethanol and 2,2,2-trifluoroethanol where those for the latter molecule have been redetermined. The currently determined heavy atom parameters are quite different from the earlier assumed values, which led to poor values of the six adjusted parameters.  相似文献

The covalent functionalization of exfoliated hexagonal boron nitride (h-BN) nanosheets by nitrene addition is described. Integration of functionalized h-BN nanosheets within a polycarbonate matrix is demonstrated and was found to afford significant increases in mechanical properties. This integration methodology was further extended by the covalent modification of the h-BN nanosheets with polymer chains of a polycarbonate analogue, and the integration of the polymer modified h-BN within the polymer matrix.  相似文献

Several different approaches have traditionally been used for detection of X‐ray powder diffraction patterns, including area detectors, point detectors and position‐sensitive detectors. Each has advantages. This paper discusses use of a low‐cost CCD detector attached to a diffractometer arm, where line‐by‐line readout of the CCD is coupled to continuous motion of the arm. When this type of detector is used and where X‐ray optics are employed to focus the source image onto the detector plane both high‐resolution and rapid measurements can be performed, with data collection over a complete 2θ range. This is particularly advantageous for synchrotron applications but valuable also for Guinier diffractometer laboratory instruments. Peak resolutions are shown to be moderately better than what can be obtained with a position‐sensitive detector and significantly better than with an area detector. Many samples have intrinsically broadened peak shapes for which little improvement in data quality could be obtained with an analyzer‐crystal detector. With comparable numbers of modules, these CCD data collection speeds can be close to those with position‐sensitive detectors, but without the low‐angle asymmetry seen in the latter.  相似文献

A reduced order model is developed for low frequency, undamped, fully coupled structural–acoustic analysis of interior cavities backed by flexible structural systems. The reduced order model is obtained by applying a projection of the coupled system matrices, from a higher dimensional to a lower dimensional subspace, whilst preserving essential properties of the coupled system. The basis vectors for projection are computed efficiently using the Arnoldi algorithm, which generates an orthogonal basis for the Krylov Subspace containing moments of the original system. The key idea of constructing a reduced order model via Krylov Subspaces is to remove the uncontrollable, unobservable and weakly controllable, observable parts without affecting the transfer function of the coupled system. Three computational test cases are analyzed, and the computational gains and the accuracy compared with the direct inversion method in ANSYS.  相似文献

The immunological synapse is a specialized intercellular junction between a T cell and a target cell that orchestrates the engagement of receptors and ligands in space and time as a means of regulating function. Here we introduce a reagent for controlling the spatial and temporal presentation of natural antigen to T cells. Moth cytochrome c (88-103) peptide (MCC), an agonist to the murine T cell receptor AND when presented in the context of H2 IEk major histocompatibility complex (IEk), was synthesized with the side-chain amine of Lys99 conjugated to a photosensitive protecting group, 6-nitroveratryloxycarbonyl (NVOC). Cells plated on supported bilayers displaying mobile intercellular adhesion molecule-1 (ICAM-1) and NVOC-MCC loaded IEk did not form immunological synapses and exhibited low intracellular calcium levels, similar to cells presented with self-peptide. Irradiation with UV light was sufficient to restore agonist activity in situ.  相似文献

Molecular dynamics simulations were performed to understand further the concentration dependence of the self-diffusion of methanol in the faujasite zeolite systems. The evolution of the self-diffusivity was investigated as a function of coverage for DAY and NaY systems to study the effect of both the pore confinement and the presence of the extraframework cations within the supercage. It was found that the self-diffusivity decreases with loading for DAY, whereas for NaY it passes through a maximum at intermediate coverage, in agreement with pulse-field gradient NMR and quasi elastic neutron scattering data reported in similar systems. The activation energies of the methanol diffusion corresponding to a combination of both intra- and intercage motions were evaluated as a function of the coverage. The simulated trends are interpreted on the basis of the predominant interactions which take place in both systems. Finally, the preferential arrangement of the adsorbate molecules are provided and compared with those simulated in the liquid phase. For the fully loaded materials, it was seen that the methanol molecules form a one-dimensional hydrogen-bonded chain along the channels in DAY whereas only dimers are present in NaY.  相似文献

The SER spectrum is reported for N-methylpyridinium iodide, an organic cation with no unshared electron pair available for interaction with the electrode. Both Raman- and infrared-active bands are observed with small frequency shifts. No SERS is observed for N-methylpyridinium chloride until trace amounts of iodide are added to the solution.  相似文献

DNA sequences attached to Au nanoparticles via thiol linkers stand up from the surface, giving preferential enhancement of the adenine ring breathing SERS band. Non-specific binding via the nucleobases reorients the DNA, reducing this effect. This change in intensity on reorientation was utilised for label-free detection of hybridization of a molecular beacon.  相似文献
