A simple route toward the synthesis of symmetrical and unsymmetrical aromatic azo compounds through oxidative coupling of anilines using widely available NaOCl is presented. This metal catalyst-free protocol is performed in water and affords the desired products in high yields.  相似文献

Although several investigations have been reported on the effect of pH or ionic strength on protein adsorption, most of them have been carried out with protein monolayers and not with single molecules. We have used atomic force microscopy to image, in phosphate buffer, single fibrinogen molecules adsorbed on mica and compare the surface coverage at variable pH (7.4, 5.8, 3.5) or ionic strength (15, 150, 500 mM) conditions. The images obtained and the statistical analysis of the surface coverage indicate adsorption enhancement at the IEP of fibrinogen (pH 5.8) and minimum adsorption at pH 3.5. On the other hand, more protein was adsorbed when the salt concentration of the buffer at pH 7.4 was increased from 15 to 150 mM. However, further increase of salt concentration up to 500 mM resulted in decreased adsorption. To confirm the aforementioned results an approaching bare Si(3)N(4) tip was used as an electrostatic analogue to a protein molecule and interaction force curves between it and the substrate were recorded. The results were in consistence with the double layer theory which justifies the screening of electrostatic repulsion as the salt concentration increases.  相似文献

A series of protected beta2-dehydroamino acids has been prepared in three steps from commercially available starting materials in good yields. These were used as substrates in rhodium-catalyzed asymmetric hydrogenation applying a mixed ligand system of monodentate phosphoramidites and phosphines. Optimization of the catalyst structure was achieved by high throughput experimentation. High enantioselectivities were obtained (up to 91%) with full conversion for a number of beta-amino acids.  相似文献

Parallel Screening has been introduced as an in silico method to predict the potential biological activities of compounds by screening them with a multitude of pharmacophore models. This study presents an early application example employing a Pipeline Pilot-based screening platform for automatic large-scale virtual activity profiling. An extensive set of HIV protease inhibitor pharmacophore models was used to screen a selection of active and inactive compounds. Furthermore, we aimed to address the usually critically eyed point, whether it is possible in a parallel screening system to differentiate between similar molecules/molecules acting on closely related proteins, and therefore we incorporated a collection of other protease inhibitors including aspartic protease inhibitors. The results of the screening experiments show a clear trend toward most extensive retrieval of known active ligands, followed by the general protease inhibitors and lowest recovery of inactive compounds.  相似文献

The phenolic components of Aronia melanocarpa were quantitatively recovered by three successive extractions with methanol. They comprise anthocyanins (mainly cyanidin glycosides) phenolic acids (chlorogenic and neochlorogenic acids) and flavonols (quercetin glycosides). Approximately 30% of the total phenolic compounds are located in the peel and the rest in the flesh and seeds. Peels contain the major part of anthocyanins (73%), while the flesh contains the major part of phenolic acids (78%). Aronia juice, rich in polyphenols, was obtained by mashing and centrifugation, while the pomace residue was dried and subjected to acidified water extraction in a fixed bed column for the recovery of residual phenolics. A yield of 22.5 mg gallic acid equivalents/g dry pomace was obtained; however, drying caused anthocyanins losses. Thus, their recovery could be increased by applying extraction on the wet pomace. The extract was encapsulated in maltodextrin and gum arabic by spray drying, with a high (>88%) encapsulation yield and efficiency for both total phenols and anthocyanins. Overall, fresh aronia fruits are a good source for the production of polyphenol-rich juice, while the residual pomace can be exploited, through water extraction and spray drying encapsulation for the production of a powder containing anthocyanins that can be used as a food or cosmetics additive.  相似文献

The development of a faradic impedimetric immunosensor based on electropolymerized polytyramine (Ptyr) films for the detection of S. typhimurium in milk is described for the first time. Polyclonal anti-Salmonella was cross-linked, in the presence of glutaraldehyde vapors, on Ptyr-modified gold electrodes. The dielectric behaviour of Ptyr films was evaluated with capacitance measurements, while their stability in neutral aqueous solutions was examined with impedimetric measurements. The effect of the concentration of tyramine in the forming solution on both the sensitivity and the dynamic range of the resulted immunosensors was also investigated. The alteration of the interfacial features of the electrodes due to different modification or recognition steps, was measured by faradic electrochemical impedance spectroscopy in the presence of a hexacyanoferrate(II)/(III) redox couple. At samples containing a low initial concentration of 10 cfu mL−1S. typhimurium, that actually defines the LOD of the immunosensors, signal changes of 33% and 88% were achieved after 3 and 10 h incubation, respectively. To achieve the working simplicity expected by a biosensor, immunoreaction was performed directly in cultures. This resulted in the elimination of various centrifugation and washing steps, which are used for the isolation of bacteria cells from the culture, thus making the proposed immunosensors promising candidates for on-site applications. Finally, the proposed immunosensors were successfully used for the detection of S. typhimurium in experimentally inoculated milk samples.  相似文献
