Purpose

To investigate the correlation between perfusion-related parameters obtained with intravoxel incoherent motion (IVIM) and classical perfusion parameters obtained with dynamic contrast-enhanced (DCE) magnetic resonance imaging in patients with head and neck squamous cell carcinoma (HNSCC), and to compare direct and asymptotic fitting, the pixel-by-pixel approach, and a region of interest (ROI)-based approach respectively for IVIM parameter calculation.

Materials and methods

Seventeen patients with HNSCC were included in this retrospective study. All magnetic resonance (MR) scanning was performed using a 3 T MR unit. Acquisition of IVIM was performed using single-shot spin-echo echo-planar imaging with three orthogonal gradients with 12 b-values (0, 10, 20, 30, 50, 80, 100, 200, 400, 800, 1000, and 2000). Perfusion-related parameters of perfusion fraction ‘f’ and the pseudo-diffusion coefficient ‘D*’ were calculated from IVIM data by using least square fitting with the two fitting methods of direct and asymptotic fitting, respectively. DCE perfusion was performed in a total of 64 dynamic phases with a 3.2-s phase interval. The two-compartment exchange model was used for the quantification of tumor blood volume (TBV) and tumor blood flow (TBF). Each tumor was delineated with a polygonal ROI for the calculation of f, f ? D* performed using both the pixel-by-pixel approach and the ROI-based approach. In the pixel-by-pixel approach, after fitting each pixel to obtain f, f ? D* maps, the mean value in the delineated ROI on these maps was calculated. In the ROI-based approach, the mean value of signal intensity was calculated within the ROI for each b-value in IVIM images, and then fitting was performed using these values. Correlations between f in a total of four combinations (direct or asymptotic fitting and pixel-by-pixel or ROI-based approach) and TBV were respectively analyzed using Pearson's correlation coefficients. Correlations between f ? D* and TBF were also similarly analyzed.

Results

In all combinations of f and TBV, f ? D* and TBF, there was a significant correlation. In the comparison of f and TBV, a moderate correlation was observed only between f obtained by direct fitting with the pixel-by-pixel approach, whereas a good correlation was observed in the comparisons using the other three combinations. In the comparison of f ? D* and TBF, a good correlation was observed only with f ? D* obtained by asymptotic fitting with the ROI-based approach. In contrast, moderate correlations were observed in the comparisons using the other three combinations.

Conclusion

IVIM was found to be feasible for the analysis of perfusion-related parameters in patients with HNSCC. Especially, the combination of asymptotic fitting with the ROI-based approach was better correlated with DCE perfusion.  相似文献

The family Ariidae comprises approximately 130 catfish species on both warm-temperate and tropical continental shelves around the world. The systematics of the group is problematic, with several misidentification problems. In order to better understand the evolutionary relationships in the family, the present study used a cytogenetic approach to characterize two populations of Genidens genidens and two populations of Aspistor luniscutis from the southern coast of Brazil using conventional techniques and fluorescent in situ hybridization with 18S rDNA probes. The two species had the same diploid number (2n = 56), high fundamental numbers and similar banding patterns, thereby corroborating the karyotypic homogeneity proposed for the group. Single nucleolus organizer regions (NORs) were found in the genus Genidens and multiple NORs were found in Aspistor, which are considered an important cytotaxonomic marker for this genus. Karyotypic evolution trends were hypothesized, providing a better understanding of the karyotype diversity and chromosome evolution processes.  相似文献

Monodisperse homogeneous and core–shell latex particles of various sized between 200 and 600 nm were synthesized by emulsion copolymerization. Some of the core–shell particles were functionalized with epoxy groups at their peripheries upon introduction of glycidyl methacrylate (GMA) during the synthesis. The core consisted of crosslinked polybutylacrylate and the shell polymethylmethacrylate. Synthesis conditions at high and low temperatures were optimized to obtain coreshell particles with a well-defined morphology. The particles were characterized by quasi-elastic light scattering, scanning electron microscopy and transmission electron microscopy. The latex particles functionalized with GMA were then dispersed into a reactive matrix (styrene and maleic anhydride copolymer) using a batch mixer to obtain blends with well-defined and stabilized morphology. 4 Dimethylaminopyridine was used as a catalyst. The reaction between the epoxy groups at the particle surface and the maleic anhydride or diacid groups of the matrix was evaluated by torque and extraction techniques. A small amount of conversion generates sufficient amounts of grafted species at the matrix and particle interfaces to ensure a good interfacial adhesion.  相似文献

Abiraterone acetate efficacy against prostate cancer is dependent on the circulating levels of abiraterone and its active metabolites, which present significant pharmacokinetic variability among patients. Thus, therapeutic drug monitoring can be performed to improve treatment outcomes. To support such studies, there are only a limited number of bioanalytical methods in current literature. This work presents a fast method to quantify abiraterone and D4A in plasma in 4 min by UPLC–MS/MS. Bioanalytical method validation was performed according to the recommendations of the US Food and Drug Administration. The method was linear within the range of 1–400 ng/ml for abiraterone and 0.2–20 ng/ml for D4A (r2 > 0.99). Based on the analysis of quality control samples at the lower limit of quantification, low, medium and high concentrations, the method was precise (CVabiraterone ≤ 9.72%; CVD4A ≤ 14.64%) and accurate (CVabiraterone 95.51–107.59%; CVD4A 98.04–99.89%). Application of the method to the quantification of abiraterone and D4A in 10 clinical samples revealed important variability in the conversion ratio of abiraterone to D4A (CV 90.85%). Considering the current literature, this is the fastest method to quantify abiraterone and D4A in plasma, allowing for optimization of the analytical routine.  相似文献

Alcohol use disorder (AUD) causes about 3.3 million deaths around the world each year. It is the primary risk factor for the global burden of diseases in American countries. Long-term abuse of alcohol induces numerous molecular and biochemical changes in tissues exposed to alcohol. The toxic effects of alcohol are mediated by DNA damage through various mechanisms, such as induction of oxidative damage, DNA adducts, crosslinks, and DNA strand breaks. The main aim of the current study was to compare the frequency of SNP polymorphisms in XRCC1 (rs7997782) and GSTP1 (rs1695) genes involved in DNA repair of single strand breaks (SSB) and xenobiotic detoxification between alcohol addicts and a control group comprised of non-drinkers. Genetic polymorphisms were identified following allelic specific PCR designed to generate the amplicons containing the variants. Then amplicons were sequenced, and sequences were aligned against the human genome reference deposited in GenBank using the CLC Sequence Viewer software (version 7.6.1). The GG homozygotes in rs1695 (GSTP1) were significantly (p = 0.023) 3.8x more frequent among those with AUD when compared to the control group. No SNP variation was observed in rs7997782 (XRCC1). rs1695 variant has been associated with susceptibility to various diseases, including those related to alcohol consumption.  相似文献

Four compounds were isolated from roots and aerial parts of Pluchea sagittalis (Asteraceae), 3, 5-dihydroxy-6, 7, 3′, 4′-tetramethoxiflavunol (1), 5-hydroxymethylfurfural (2), 3, 4-dimethoxybenzaldehyde (3) and 2, 3, 4-trihydroxybenzaldeyde (4). Their herbicidal potential was detected by polarographic techniques. All of them inhibited the non-cyclic electron transport on basal, phosphorylating and uncoupled conditions from H2O to methylviologen (MV); thus, they act as Hill reaction inhibitors. Studies on fluorescence of chlorophyll a (ChL a) indicated they have different modes of interaction and inhibition sites on the photosystem II electron transport chain; 13 have interacted with the acceptor side while 4 has interacted at the donor side.  相似文献
