IspG is a 4Fe-4S protein that carries out an essential reduction step in isoprenoid biosynthesis. Using electron-nuclear double resonance (ENDOR) and hyperfine sublevel correlation (HYSCORE) spectroscopies on labeled samples, we have specifically assigned the hyperfine interactions in a reaction intermediate. These results help clarify the nature of the reaction intermediate, supporting a direct interaction between the unique fourth Fe in the cluster and C2 and O3 of the ligand. 相似文献

C(60) was reacted in the ionization chamber of a mass spectrometer under electron impact (EI) with aldehydes, RCHO (R = Ph, p-FC(6)H(4), F(5)C(6), p-MeOC(6)H(4), α-thienyl, o-HOC(6)H(4), o-BrC(6)H(4), m-BrC(6)H(4) and t-Bu), with the transfer of R? radicals and with Me?-transfer from i-PrCHO and t-BuCHO. Paramagnetic fullerene derivatives were stabilized by the addition of the next R? radical or a hydrogen atom, or hydrogen or bromine atom loss. A detailed study showed that the reaction between C(60) and PhCHO occurred via a homolytic mechanism that matches one reported earlier for the reaction with acetone. This suggests the generality of the mechanism for the reactions of fullerenes with other species in ionization chambers under EI at ca 300°C. All aldehydes, except one, had radicals at the carbonyl group which were different from those in the ketones examined earlier in the reactions. This expanded the variety of radicals which can be transferred to fullerenes during reactions in ionization chambers under EI. Due to this and the hydrogen atom at the CO group of aldehydes, some reactions occurred that were not found for the ketones: the formation of cyclic products C(60)COC(6)H(4) and C(60)OC(6)H(4) for PhCHO, o-BrC(6)H(4)CHO and o-HOC(6)H(4)CHO, respectively, and HC(60)Ph for o- and m-BrC(6)H(4)CHO. The reaction with α- formylthiophen gives the first example of transferring an aromatic heterocyclic radical to C(60) in an ionization chamber under EI. C(70) reacted with PhCHO, p-FC(6)H(4)CHO and i- PrCHO similarly to C(60). The results for the reactions of C(60) with PhCHO and with i- PrCHO were compared with those in solution under UV irradiation. Incomplete but reasonable coincidence was found; in both modes, the addition of Ph?, PhCO? and Me? radicals to C(60) occurred, whereas some other products were formed in solution, and the explanation is given as to why this occurred. This conformity supports the hypothesis based on the results of kindred reactions with ketones and organomercurials: the results of EI-initiated homolytic reactions between fullerenes and other compounds in an ionization chamber can predict the reactivity of the fullerenes toward them in solution. 相似文献

Electrocatalytic generation of nickel catalysts in low oxidation states by reduction of nickel complexes with various ligands (2,2'-bipyridine, 2,2':6',2'-terpyridine, (S,S)-2,6-bis(4-phenyl-2-oxazolin-2-yl)-pyridine) in the presence of olefinic substrates and fluoroalkyl halides leads to new organic products derived from addition-dimerization processes. Due to the presence of two stereocenters in the dimerization products two diastereomers were characterized by a variety of analytical techniques including multi-dimensional NMR methods and X-ray single crystal diffraction. The formation of dimers was prevented by the inclusion of the hydrogen atom donor tributyltin hydride. Dihydrogen sulfide recently emerged as a biological signaling molecule with important physiological roles and significant pharmacological potential. Chemically plausible explanations for its mechanisms of action have remained elusive, however. Here, we report that H(2)S reacts with S-nitrosothiols to form thionitrous acid (HSNO), the smallest S-nitrosothiol. These results demonstrate that, at the cellular level, HSNO can be metabolized to afford NO(+), NO, and NO(-) species, all of which have distinct physiological consequences of their own. We further show that HSNO can freely diffuse through membranes, facilitating transnitrosation of proteins such as hemoglobin. The data presented in this study explain some of the physiological effects ascribed to H(2)S, but, more broadly, introduce a new signaling molecule, HSNO, and suggest that it may play a key role in cellular redox regulation. 相似文献

Polyphosphoinositides (PPIs) and in particular phosphatidylinositol-(4,5)-bisphosphate (PI4,5P2), control many cellular events and bind with variable levels of specificity to hundreds of intracellular proteins in vitro. The much more restricted targeting of proteins to PPIs in cell membranes is thought to result in part from the formation of spatially distinct PIP2 pools, but the mechanisms that cause formation and maintenance of PIP2 clusters are still under debate. The hypothesis that PIP2 forms submicrometer-sized clusters in the membrane by electrostatic interactions with intracellular divalent cations is tested here using lipid monolayer and bilayer model membranes. Competitive binding between Ca(2+) and Mg(2+) to PIP2 is quantified by surface pressure measurements and analyzed by a Langmuir competitive adsorption model. The physical chemical differences among three PIP2 isomers are also investigated. Addition of Ca(2+) but not Mg(2+), Zn(2+), or polyamines to PIP2-containing monolayers induces surface pressure drops coincident with the formation of PIP2 clusters visualized by fluorescence, atomic force, and electron microscopy. Studies of bilayer membranes using steady-state probe-partitioning fluorescence resonance energy transfer (SP-FRET) and fluorescence correlation spectroscopy (FCS) also reveal divalent metal ion (Me(2+))-induced cluster formation or diffusion retardation, which follows the trend: Ca(2+) ? Mg(2+) > Zn(2+), and polyamines have minimal effects. These results suggest that divalent metal ions have substantial effects on PIP2 lateral organization at physiological concentrations, and local fluxes in their cytoplasmic levels can contribute to regulating protein-PIP2 interactions. 相似文献

Postponement strategies are becoming increasingly important in light of a global trend in which products’ life-cycles are decreasing, such that even products that are not traditionally considered seasonal become “obsolete” within a short period of time (e.g., electronic devices, new cars). Our work addresses postponed-pricing and ordering decisions for a retailer who sells a newsvendor-type inventoried product, in a selling season that is divided into two sub-periods. The division of the selling season enables the retailer to on-line adjust her decisions when faced with a scenario (one that is highly prevalent in reality) in which potential demand changes (increases or decreases) following consumers’ experiences of the product in early stages of the selling season. We assume that the retailer has two opportunities for receiving shipments: prior to the first sub-period and prior to the second one. The retailer determines each order quantity (base-stock level) on the basis of the demand distribution for the corresponding sub-period. In each sub-period, after observing additional market signals, the retailer determines the price of the product for that sub-period. With the aid of a stochastic programming approach, we develop optimization problems and solution methods in order to obtain pricing and ordering decisions that maximize the expected profit of the retailer. We present an extensive numerical example that compares the suggested strategy to three alternative strategies, and conclude that price postponement and responsiveness to demand changes can each reduce leftovers and lost sales as well as substantially increase expected profit. 相似文献

The isomerization of M+ ˙ of o-carboxy- and o-carboxamidocyclopropylbenzenes into five- and six-membered heterocycles due to the ortho effect has been demonstrated by means of electron impact mass spectra, high-resolution mass spectrometry data and metastable ion spectra. The criteria for the quantitative estimation of the degree of cyclopropane cyclization into these isomeric cycles were formulated. Orthorhombic (s.g. P 21 21 2, a = 28.6145 Å, b = 27.4115 Å, c = 6.9536 Å, V = 5454.1 Å3) betulin (C30H50O2) was found in this study to melt at 245 °C with the enthalpy of fusion 40.3 J mol?1. The shape of the peak of melting gives rise to the belief that there are several polymorphs of betulin. 相似文献
