Evaluation of the adsorption affinity of proteins to calcium hydroxyapatites by desorption and pre-adsorption methods

The adsorption affinity of bovine serum albumin (BSA) and lysozyme (LSZ) to calcium hydroxyapatite (CaHAP) was evaluated by desorption and two step adsorption methods. These experiments were carried out at 15°C in a 1×10⁻⁴ mol dm⁻³ KCl solution of pH 6.0. BSA molecules were scarcely desorbed, exhibiting an irreversible adsorption of BSA, though LSZ slightly desorbed. This result supports our previous findings that LSZ adsorbs weakly onto phosphate ions exposed on ac or bc faces of CaHAP while BSA adsorbs strongly onto positively charged sites on ac or bc faces of CaHAP. The amount of adsorbed LSZ was markedly increased by the pre-adsorption of BSA, where LSZ was adsorbed onto BSA-covered CaHAP. On the other hand, the amount of adsorbed BSA was not changed by the pre-adsorption of LSZ. In both pre-adsorption systems it was confirmed by an HPLC method that no protein molecule pre-adsorbed was desorbed after the post-adsorption procedure. Therefore, it was interpreted that the enhancement of adsorption of positively charged LSZ is induced by an electrostatic attractive force through pre-adsorption of negatively charged BSA molecules with a high coverage. However, since the coverage of LSZ onto CaHAP is considerably low, no stimulation of BSA adsorption occurred on the LSZ-covered surface. The formation of double protein adsorbed layers consisting of pre- and post-adsorbed proteins was proposed.     

The photochemical isomerization of cross-linked 1,3,5-tristyrylbenzene dendrimer with hula-twist mechanism

A cross-linked 1,3,5-tristyrylbenzene dendrimer 5 was synthesized to study the photochemical isomerization mechanism. On irradiation, 5 isomerized with the quantum yields (Φ_E→Z) of 0.063, which was not very much different from that for a model compound 1 (Φ_E→Z 0.080), supporting a volume-conserving hula-twist (HT) mechanism during the photochemical E-Z isomerization.     

Studies of nucleosides and nucleotides—LXXIV: Purine cyclonucleosides—34 a new method for the synthesis of 2'-substituted 2'-deoxyadenosines

8,2'-O-Cycloadenosine was protected at 3' and 5'-OHs with acetyl groups and cleaved using liq. H₂S. Subsequent dethiolation and mesylation gave 2'-O-mesyl-3',5'-di-O-acetyl-arabinosyladenine (6). When 6 or its deacetylated parent compound (7) was heated with sodium azide in DMF, 3'-azido-3'-deoxyxylofuranosyladenine (9) was the only product. The cyclonucleoside was then protected with tetrahydropyranyl groups and subjected to a similar series of reactions as above to give 2'-O-mesyl-3',5'-di-O-tetrahydropyranylarabinosyladenine (14). The compound 14 was heated with sodium azide after which acidic deprotection afforded 2'-azido-2'-deoxyadenosine (16). Hydrogenation of 16 gave 2'-amino-2'-deoxyadenosine (18). 2'-Chloro-2'-deoxyadenosine (19) was also obtained by treatment of 14 with lithium chloride and subsequent deprotection. UV, IR and NMR spectral data of these compounds are described.     

Emission of 2-phenylethanol from its β-d-glucopyranoside and the biogenesis of these compounds from [H8] l-phenylalanine in rose flowers

The hydrolysis of 2-phenylethyl β-d-glucopyranoside (3) was found to be partially inhibited by feeding with 2-phenyl-N-glucosyl-acetamidiumbromide (8), a β-glucosidase inhibitor, resulting in a decrease in the diurnal emission of 2-phenylethanol (2) from Rosa damascena Mill. flowers. Detection of [1,1,2,2′,3′,4′,5′,6′-²H₈]-2 and [1,2,2′,3′,4′,5′,6′-²H₇]-2 from R. ‘Hoh-Jun’ flowers fed with [1,1,2,2′,3′,4′,5′,6′-²H₈]-3 suggested that β-glucosidase, alcohol dehydrogenase, and reductase might be involved in scent emission. Comprehensive GC-SIM analyses revealed that [1,2,2,2′,3′,4′,5′,6′-²H₈]-2 and [1,2,2,2′,3′,4′,5′,6′-²H₈]-3 must be biosynthesized from [1,2,2,2′,3′,4′,5′6′-²H₈] l-phenylalanine ([²H₈]-1) with a retention of the deuterium atom at α-position of [²H₈]-1.     

Effect of molecular weight in amphipathic polyethyleneglycol on prolonging the circulation time of large unilamellar liposomes

The effect in mice of the molecular weight of polyethyleneglycol on prolonging the circulation time of large unilamellar liposomes (LUVs) was examined using four different distearoyl N-(monomethoxy polyethyleneglycol succinyl) phosphatidylethanolamines (DSPE-PEGs). The molecular weights tested were 1000, 2000, 5000 and 12000. Incorporation of 6 mol% of DSPE-PEG in LUV composed of distearoylphosphatidylcholine (DSPC) / cholesterol (CH) (1:1 in molar ratio) increased the blood circulation half-life significantly more than those without DSPE-PEG derivatives. DSPE-PEGs with molecular weights of 1000 and 2000 prolonged the circulation time of liposomes more than other DSPE-PEGs with higher molecular weights, such as 5000 and 12000. Their effects are also higher than ganglioside GM1, a well described glycolipid with this effect. DSPC/CH LUV-incorporated DSPE-PEG with a molecular weight of 2000 displayed a high concentration in the blood, approximately 40% of the dose, 6 h after the injection.     

Analysis by partial reflection spectrometry of protonated tetraphenylporphyrin adsorbed at a liquid-liquid interface

Visible reflection spectra of diprotonated meso-tetraphenylporphyrin adsorbates spontaneously formed at a dodecane-aqueous sulfuric acid interface have been measured using a home-made device comprising a prism-cell and variable-angle optics. The tilt angle of the pyrrole ring plane was estimated to be 47 degrees from the interface normal by use of an experimentally evaluated molecular density (1.20x10(-10) mol cm(-2)) of the diprotonated molecule in a monolayer form at the liquid-liquid interface. Positive and negative bands have been observed in the p-polarized partial internal reflection (p-PIR) spectra, whose band locations correspond to those in p-polarized external reflection (p-ER) spectra. Nevertheless, the bands in the p-PIR exhibited reversed sign to those of p-ER spectra. These suggest that the surface selection rule of the p-PIR spectrometry has a reversal rule of p-ER and p-PIR can also be used for the analysis of molecular orientation.     

Conformation of poly(S-(2-N-carbazolylethyl)-L-cysteine) in the solid state

The β-forming characteristics of poly(S-(2-N-carbazolylethyl)-L -cysteine), (CELC)_n, were examined by infrared (IR) spectroscopy. The film of (CELC)₃₉ cast from tetrahydrofuran (THF) showed the typical spectrum of the antiparallel β-form with the amide I band at 1630 cm^?1, whereas that of (CELC)₂₀₀ from dimethylformamide (DMF) or pyridine exhibited the amide I band at 1640 cm^?1 which shifted to 1630 cm^?1 on heating at 100°C for 5 min. As a result of the examination of the spectral variation due to the degree of polymerization, casting solvents, casting temperatures, and heat treatment, together with the evaluation of interaction constants for intermolecular hydrogen bonds, the band at 1640 cm^?1 was attributed to the antiparallel-chain pleated sheet (designated as β′-form) with weaker hydrogen bond strength than that of the usual antiparallel β-form. The β′-β-transformation is discussed in terms of the rigidity of the side-chain region.     

Preparation and characterization of carbonated barium-calcium hydroxyapatite solid solutions

Particles of carbonated barium-calcium hydroxyapatite solid solutions (BaCaHap) with different Ba/(Ba+Ca) (X(Ba)) atomic ratios were prepared by a wet method at 100 degrees C and characterized by various means. The crystal phases and structures of the products strongly depended on the composition of the starting solution, that is, the Ba/(Ba+Ca) atomic ratio ([X(Ba)]) and H3PO4 concentration ([H3PO4]) in the solution. BaCaHap with X(Ba)0.43 could be prepared at [X(Ba)]0.7 by changing [H3PO4], but could never be obtained at [X(Ba)]=0.8-0.95 regardless of [H3PO4]. The carbonated calcium hydroxyapatite particles prepared at [X(Ba)]=0 were fine and short rod-shaped particles (ca. 14x84 nm). With increasing [X(Ba)] from 0 to 0.8, the particles obtained became large spherical agglomerates. The carbonated barium hydroxyapatite particles formed at [X(Ba)]=1 were long rod-shaped agglomerates (ca. 0.2x2 microm) of fine primary particles. The amount of CO2 adsorbed irreversibly on a series of BaCaHaps showed a minimum at (Ba+Ca)/(P+C) atomic ratio of around 1.56, which agreed well with the minimum cation/P ratio obtained for the other hydroxyapatites, as already reported.     

Differential calculus on the quantum superspace and deformation of phase space

We investigate non-commutative differential calculus on the supersymmetric version of quantum space in which quantum supergroups are realized. Multiparametric quantum deformation of the general linear super-group,GL _q(m|n), is studied and the explicit form for the \(\hat R - matrix\) is presented. We apply these results to the quantum phase-space construction ofOSp _q(2n|2m) and calculate their \(\hat R - matrices\) .
