The quantitative evaluation on measurement of liver volume and the hepatic accumulation rate of the radioactive agent were studied with respect to body constitution, background activity and splenic consumption rate, using single photon emission computed tomography (SPECT). The optical cut-off level for liver volume determination was proved to be reasonable as 35%. The background less than 5% had no influence to their clinical estimation with the error of less than 1.6%. The estimation of liver volume was mostly influenced by the splenic activity. When the splenic activity was increased two times of the hepatic activity, the liver volume was determined as 4.2% larger. Calculated counts by SPECT was well correlated with the hepatic activity. This study proved that SPECT was useful for the quantitative evaluation of the liver volume and the hepatic accumulation rate.  相似文献

In the present study, a new (E)‐rich‐enyne π‐conjugated polymer containing a carbazole was designed and synthesized. Two different synthesis methods of poly[N‐(2‐ethylhexyl)‐3,6‐carbazolyleneethynylene‐(E)‐vinylene] (PCZEV) have been prepared from 3,6‐diethynyl‐9(2‐ethylhexyl)carbazole by using the palladium‐carbene‐catalyzed reaction and/or by using the organolanthanide‐catalyzed reaction leading to well‐defined polymer, and their general properties were studied. Compared to poly[N‐(2‐ethylhexyl)‐3,6‐carbazolyleneethynylene] (PCE), the UV‐vis absorption and photoluminescence of the PCZEV was red‐shifted, which indicates the extension of conjugation length. © 2009 Wiley Periodicals, Inc. Blue phases (BPs) obtained by doping a commercially available liquid crystalline compound (4-butyl-N-(4-ethoxybenzylidene)aniline (EBBA)) with (2R,3R)-2,3-bis(4-octyloxyphenylbenzoyloxy)-N-(hydroxydecyl)succinimide (1) were highly stabilised by the addition of a small amount of alkane diols. Especially, addition of only 0.5 mol% of octane-1,8-diol increased the BPIso transition temperature by 10 K and widened the BP temperature range up to 35 K. A model stabilisation mechanism based on the construction of a hydrogen bond network in each disclination zone in BPs was proposed.  相似文献

Protein trisulfide linkages are generated by the post-translational insertion of a sulfur atom into a disulfide bond. Molecular heterogeneity was detected in a recombinant IgG1 monoclonal antibody (mAb) and attributed to the presence of a protein trisulfide moiety. The predominant site of trisulfide modification was the bond between the heavy and light chains. The trisulfide was eliminated during purification of the IgG1 mAb via a cysteine wash step incorporated into Protein A affinity column chromatography. Analysis of the cysteine-treated mAb by electrophoresis and peptide mapping indicated that the trisulfide linkages were efficiently converted to intact disulfide bonds (13% trisulfide decreased consistently to 1% or less) without disulfide scrambling or an increase in free sulfhydryls. The on-column trisulfide conversion caused no change in protein folding detectable by hydrogen/deuterium exchange or differential scanning calorimetry. Consistent with this, binding of the mAb to its antigen in vitro was insensitive to the presence of the trisulfide modification and to its removal by the on-column cysteine treatment. Similar, high efficiency trisulfide conversion was achieved for a second IgG1 mAb using the column wash strategy (at least 7% trisulfide decreased to 1% or less). Therefore, trisulfide/disulfide heterogeneity can be eliminated from IgG1 molecules via a convenient and inexpensive procedure compatible with routine Protein A affinity capture.  相似文献

Nano-sized magnetic Y3Fe5O12 ferrite having a high heat generation ability in an AC magnetic field was prepared by bead milling. A commercial powder sample (non-milled sample) of ca. 2.9 μm in particle size did not show any temperature enhancement in the AC magnetic field. The heat generation ability in the AC magnetic field improved with a decrease in the average crystallite size for the bead-milled Y3Fe5O12 ferrites. The highest heat ability in the AC magnetic field was for the fine Y3Fe5O12 powder with a 15-nm crystallite size (the samples were milled for 4 h using 0.1 mm? beads). The heat generation ability of the excessively milled Y3Fe5O12 samples decreased. The main reason for the high heat generation property of the milled samples was ascribed to an increase in the Néel relaxation of the superparamagnetic material. The heat generation ability was not influenced by the concentration of the ferrite powder. For the samples milled for 4 h using 0.1 mm? beads, the heat generation ability (W g−1) was estimated using a 3.58×10−4 fH2 frequency (f/kHz) and the magnetic field (H/kA m−1), which is the highest reported value of superparamagnetic materials.  相似文献

Conditions for the synthesis of aluminum organophosphonate (AOP) and aluminophosphate (AlPO) spheres containing periodic mesopores were optimized and demonstrated to be general morphological controls for the surfactant‐assisted synthesis of mesoporous materials. High‐quality AOP and AlPO spheres with uniform mesopores were obtained at low and high temperatures, respectively. The aerosol‐assisted synthesis of materials with uniform mesopores was categorized by using the difference in relative density of soluble AOP and AlPO oligomers that interact with ethylene oxide (EO) units in EOnPOmEOn triblock copolymer (PO=propylene oxide). Then, ordered mesoporous structures are constructed with the adequate amount of species in resultant frameworks, and the number of interactive points in soluble species determines the resultant density of the frameworks after self‐assembly. Consequently, temperature‐dependent synthesis, which allows controlled infiltration of soluble species to match the density of resultant frameworks, is required for the formation of ordered mesoporous structures under morphological control.  相似文献

Phoborhodopsin (pR; also called sensory rhodopsin II, SRII) is a photoreceptor of negative phototaxis of halobacteria. The studies of photochemical properties of this pigment are not many because the amount of the pigment is small and the stability is low. Recently an expression system of phoborhodopsin from Halobacterium salinarum (called salinarum phoborhodopsin, spR; also HsSRII) in Escherichia coli and purification method has been developed (Mironova et al. [2005] FEBS Lett., 579, 3147–3151), which enables detailed studies on the photochemical properties of spR. In the present work, the photoreaction cycle of E. coli-expressed spR was studied by low-temperature spectroscopy and flash photolysis. Formations of K-, M-, O-like intermediates and P480 were reconfirmed as reported previously. New findings are as follows. (1) The K-like intermediate (P500) was a mixture of two photoproducts. (2) Formation of L-like intermediate (P482) was observed by low-temperature spectroscopy and flash photolysis at room temperature. (3) On long irradiation of spR at 20°C, formation of a new photoproduct P370 was observed and it decayed to the original spR in the dark with a decay half time of 190 min. Based on these results the similarities and dissimilarities between spR and ppR are discussed.  相似文献

Novel unsymmetrical vanadyl and zinc dodecakis(2,2,2‐trifluoroethoxy)phthalocyaninates with an iodo group were synthesized by a statistical condensation route. The palladium‐catalyzed coupling reaction between such monoiodinated metallophthalocyanines and terminal acetylenic derivatives gave various new unsymmetrically dodecakis(2,2,2‐trifluoroethoxy)‐substituted metallophthalocyanines with extended exocyclic π‐conjugation. Unsymmetrical zinc dodecakis(2,2,2‐trifluoroethoxy)phthalocyaninate with a nitro group was also prepared for comparison. All the target phthalocyanines were separated by common col umn chromatography and characterized by elemental analysis, ir and 1H‐nmr, uv‐visible and fast‐atombombardment mass spectroscopy.  相似文献
