Described herein is the synthesis of BMS‐986001 by employing two novel organocatalytic transformations: 1) a highly selective pyranose to furanose ring tautomerization to access an advanced intermediate, and 2) an unprecedented small‐molecule‐mediated dynamic kinetic resolution to access a variety of enantiopure pyranones, one of which served as a versatile building block for the multigram, stereoselective, and chromatography‐free synthesis of BMS‐986001. The synthesis required five chemical transformations and resulted in a 44 % overall yield.  相似文献

Starting from 3,5-dimethylisoxazole the carboxylic acids I and V, the amino acids VIII (L-) and IX (D-), and the ureido acids X (L-) and XI (D-) were prepared, which were used for the synthesis of the new cephalosporins XVIIb, XXa-c (L-), and XXIb (D-). Thein vitro antibacterial activity of these semi-synthetic antibiotics was studied. The sustainable development and consumption need more efficient use of natural resources. As a consequence, the use of industrial solvents demands their recovery instead of end-of-pipe treatment. It is not always clear, however, which treatment alternative should be applied. Based on an industrial case study, the environmental and economic evaluation and comparison of the treatment alternatives of a non-ideal solvent mixture containing azeotropes is investigated for determining the preferable option. For the recovery of the industrial solvent mixture, two different separation alternatives are evaluated: a less effective alternative and a novel design based on hybrid separation tools. An end-of-pipe treatment alternative, incineration, is also considered and the split of the solvent mixtures between recovery and incineration is investigated. The environmental evaluation of the alternatives is carried out using ‘Eco-indicator 99 life-cycle impact assessment methodology’. Economic investigation is also accomplished. The economic features clearly favour the total recovery, however, the environmental evaluation detects that if a recovery process of low efficiency is applied, its environmental burden can be similar or even higher than that of the incineration. This motivates engineers to design more effective recovery processes and reconsider the evaluation of process alternatives at environmental decision making.  相似文献

Sensor arrays are useful for many purposes. Our interests include quasi-distributed intrinsic fiber optic arrays, those distributed along the length of an optical fiber. We have demonstrated an optical time-of-flight approach to distinguishing the fluorescence output of such arrays, as well as a synthesis of combinatorial libraries that takes advantage of a support of linear morphology to make numerous compounds in a simple manner without information loss in the synthesis. To unite these research areas, we needed an optical fiber cladding material that meets demanding synthetic and optical requirements. We have chosen the Meldal SPOCC polymer support as the best candidate for such a material and report here our initial results with this material.  相似文献

The solvent-detergent (S/D) method was applied for inactivation of lipid-enveloped viruses during the production of immunoglobulins. Amberlite XAD-7 resin was used for removal of solvent (tri-n-butyl phosphate, TnBP) and detergent (Triton X-100) after the performed S/D inactivation procedure. The S/D reagents from the immunoglobulin preparation were adsorbed on Amberlite XAD-7, while immunoglobulins passed through the column and retained their biological activity. Using the method developed here, the final immunoglobulin preparation contains less than 1 ppm of Triton X-100 and less than 2 ppm TnBP.  相似文献

We report two crystal structures of a synthetic porphyrin molecule which was programmed for self-assembly. The same groups which ensure that bacteriochlorophylls c, d, and e can self-assemble into the chlorosomal nanorods, the photosynthetic antenna system of some green bacteria, have been engineered into desired positions of the tetrapyrrolic macrocycle. In the case of the 5,15-meso-substituted anchoring groups, depending upon the concentration, by using the same crystallization solvents, either a tetragonal or a layered structure of porphyrin stacks were encountered. Surprisingly, pi-pi interactions combined with extensive dispersive interactions, which also encompass cyclohexane, one of the crystallization solvents, win over putative hydrogen bonding. We are aware that our compounds differ considerably from the natural bacteriochlorophylls, but based upon our findings, we now question the hydrogen-bonding network, previously proposed to organize stacks of bacteriochlorophylls. Transmission electron microscopy (TEM), atomic force microscopy (AFM), and small-angle X-ray scattering (SAXS) on various isomeric compounds support our challenge of current models for the chlorosomal antenna as these show structures, astonishingly similar to those of chlorosomes.  相似文献

In order to continue our previous studies concerning Geranium pyrenaicum Burm. (Geraniaceae), we have performed spectrophotometric determinations and a HPLC study of some polyphenols. We have analyzed the dried Geranii pyrenaici herba (harvested from Cluj-Napoca, district of Cluj, Romania). We have established the content in flavonoids (0.316%), phenolic acids (0.099%), tannins (5.295%), and anthocyanins (12.030 mg/100 g vegetal product). We have identified and measured by HPLC the following compounds: hyperoside (21.61 μg/100 mg), ellagic acid (1810.44 μg/100 mg), isoquercitrine (11.197 μg/100 mg), and caftaric acid (76.83 μg/100 mg). We have also analyzed by HPLC a hydrolyzed sample of the same drug in which we have identified and measured: ellagic acid (4139.33 μg/100 mg), quercetol (29.65 μg/100 mg), kaempherol (41.48 μg/100 mg), and caftaric acid (20.721 μg/100 mg). __________ Published in Khimiya Prirodnykh Soedinenii, No. 4, pp. 322–324, July–August, 2005.  相似文献

For inactivation of lipid-enveloped viruses during the immunoglobulin production, the solvent-detergent (S/D) method was applied. Tri-n-butyl phosphate (solvent) and Triton X-100 (detergent) were removed from S/D treated immunoglobulins by ion-exchange chromatography on Q-Sepharose Fast Flow (FF). During the chromatographic procedure immunoglobulins remained bound on a Q-Sepharose FF, whereas solvent and detergent were eluted by washing with starting buffer. Elution of immunoglobulins was achieved by increasing the ionic strength of the starting buffer. The final immunoglobulin preparation contained less than 10 microg/ml of Triton X-100 and less than 2 microg/ml tri-n-butyl phosphate. It was confirmed that the S/D procedure did not cause a significant change in polymers and specific antibodies content. Immunoglobulin classes were also not affected by the same procedure.  相似文献
