The synthesis of methyl 5-O-benzoyl-2-chloro-2,3-dideoxy-3-fluoro-beta-d-ribofuranoside (5) and its use as a glycosylating agent for persilylated thymine, N(6)-benzoyladenine, and N(4)-benzoylcytosine are described (Scheme 1). The 2'-chloro-2',3'-dideoxy-3'-fluoro-d-ribonucleosides 10-12 synthesized were transformed to 2',3'-dideoxy-3'-fluoro-alpha- and -beta-d-erythro-pentofuranoside nucleosides of thymine (13a,b), adenine (14a,b), and cytidine (15a,b) by treatment with tributyltin hydride in the presence of alpha,alpha'-azobisisobutyronitrile (Scheme 2). Treatment of 2'-chloro-2',3'-dideoxy-3'-fluoro-d-ribonucleosides with 1 M MeONa/MeOH under reflux for 1-5 h afforded 2',3'-didehydro-2',3'-dideoxy-2'-chloro-d-pentofuranosyl nucleosides as the principal products (47-81%) of the reaction, along with recovered starting nucleoside (11-33%) (Scheme 3). Easy HF elimination was also observed in the case of the 2'-azido-2',3'-dideoxy-3'-fluoro-beta-d-ribofuranosides of thymine (17) and adenine (20) (Scheme 3). The role of conformational peculiarities of 2'-chloro-2',3'-dideoxy-3'-fluoro-d-ribonucleosides as well as of 17 and 20 in the observed exclusive elimination of HF is discussed. The conformational analysis of a rather broad palette of 2,3-dideoxy-3-fluoro-2-(X-substituted)-d-ribofuranosides was performed with the aid of the PSEUROT (version 6.3) program, using (i) the recently reparametrized Karplus-type relation (Chattopadhyaya and co-workers. J. Org. Chem. 1998, 63, 4967) and (ii) empirical bond angle correction terms suggested by us. The predictive power of the Brunck and Weinhold model (J. Am. Chem. Soc. 1979, 101, 1700) of the gauche effect between atoms and groups as a conformational driving force acting upon the pentofuranose ring is explored. Their model invokes maximum antiperiplanar sigma <--> sigma stabilization when the donating bond is the least polar one and the acceptor orbital is at the most polarized bond and is found at least as satisfactory, and in various specific cases more so than, as rationalizations on the basis of the preference of the gauche vs the trans conformation of two vicinal electronegative substituents (Wolfe. Synthetic studies are presented addressing the oxidative decarbonylation of molybdenum and tungsten complexes supported by the encumbering m-terphenyl isocyanide ligand CNAr(Dipp2) (Ar(Dipp2) = 2,6-(2,6-(i-Pr)(2)C(6)H(3))(2)C(6)H(3)). These studies represent an effort to access halide or pseudohalide M/CNAr(Dipp2) species (M = Mo, W) for use as precursors to low-coordinate, low-valent group 6 isocyanide complexes. The synthesis and structural chemistry of the tetra- and tricarbonyl tungsten complexes trans-W(CO)(4)(CNAr(Dipp2))(2) and trans-W(NCMe)(CO)(3)(CNAr(Dipp2))(2) are reported. The acetonitrile adducts trans-M(NCMe)(CO)(3)(CNAr(Dipp2))(2) (M = Mo, W) react with I(2) to form divalent, diiodide complexes in which the extent of decarbonylation differs between Mo and W. In the molybdenum example, the diiodide, dicarbonyl complex MoI(2)(CO)(2)(CNAr(Dipp2))(2) is generated, which has an S = 1 ground state in solution. Paramagnetic group 6 MX(2)L(4) complexes are rare, and the structure of MoI(2)(CO)(2)(CNAr(Dipp2))(2) is discussed in relation to other diamagnetic and C(2v)-distorted MX(2)L(4) complexes. Diiodide MoI(2)(CO)(2)(CNAr(Dipp2))(2) reacts further with I(2) to effect complete decarbonylation, producing the paramagnetic tetraiodide complex trans-MoI(4)(CNAr(Dipp2))(2). The reactivity of the trans-M(NCMe)(CO)(3)(CNAr(Dipp2))(2) (M = Mo, W) complexes toward benzoyl peroxide is also surveyed, and it is shown that dicarboxylate complexes can be obtained by oxidative or salt-elimination routes. The reduction behavior of the tetraiodide complex trans-MoI(4)(CNAr(Dipp2))(2) toward Mg metal and sodium amalgam is studied. In benzene solution under N(2), trans-MoI(4)(CNAr(Dipp2))(2) is reduced by Na/Hg to the η(6)-arene-dinitrogen complex, (η(6)-C(6)H(6))Mo(N(2))(CNAr(Dipp2))(2). The diiodide-η(6)-benzene complex (η(6)-C(6)H(6))MoI(2)(CNAr(Dipp2))(2) is an isolable intermediate in this reduction reaction, and its formation and structure are discussed in context of putative low-coordinate, low-valent molybdenum isocyanide complexes. 相似文献

The subject of proton transfer between carbon acids and nitrogen bases in aprotic solvents is reviewed. Equilibrium and rate constants that characterize such reactions are most often determined utilizing UV-visible spectrophotometry. At ambient temperature reaction rates are sufficiently rapid that fast reaction methods, for example, the stopped-flow and temperature-jump techniques are required in many cases. Variation of the properties of the donor and acceptor reaction pairs enables electronic and steric effects upon thermodynamic and kinetic parameters of proton transfer to be assessed. Determination of the kinetic isotope effect (KIE), i.e. k(protium)/k(deuterium) led to the conclusion that, under certain circumstances and when the KIE is greater than seven, the proton undergoes reaction with a significant degree of quantum mechanical tunneling, consistent with a theoretical prediction advanced several decades earlier. In fact this aspect may be one of the most significant outgrowths of these studies. Many reactions have been characterized (by tunneling) but rarely are the reacting systems experimentally amenable to obtaining all the experimental criteria that support tunneling. Controversy that has arisen regarding treatment of experimental data and resulting conclusions from them is visited in this review. The structural nature of the product state of reaction is formulated based on spectroscopic evidence, in favorable cases, and probable structures of the transition state can be inferred. 相似文献

Inter- and intramolecular hydrogen bonding of an N-H group in pyrazole complexes was studied using ligands with two different groups at pyrazole C-3 and C-5. At C-5, groups such as methyl, i-propyl, phenyl, or tert-butyl were present. At C-3, side chains L-CH(2)- and L-CH(2)CH(2)- (L = thioether or phosphine) ensured formation of chelates to a cis-dichloropalladium(II) fragment through side-chain atom L and the pyrazole nitrogen closest to the side chain. The significance of the ligands is that by placing a ligating side chain on a ring carbon (C-3), rather than on a ring nitrogen, the ring nitrogen not bound to the metal and its attached proton are available for hydrogen bonding. As desired, seven chelate complexes examined by X-ray diffraction all showed intramolecular hydrogen bonding between the pyrazole N-H and a chloride ligand in the cis position. In addition, however, intermolecular hydrogen bonding could be controlled by the substituent at C-5: complexes with either a methyl at C-5 or no substituent there showed significant intermolecular hydrogen bonding interactions, which were completely avoided by placing a tert-butyl group at C-5. The acidity of two complexes in acetonitrile solutions was estimated to be closer to that of pyridinium ion than those of imidazolium or triethylammonium ions. 相似文献

The intramolecular benzannulations of carbene complexes with alkynes are examined where the alkyne is tethered to the alpha-carbon of the vinyl carbene complex. These reactions are sensitive to the length of the tether and to the nature of the solvent. With a tether length of 16 methylenes, the reaction occurs in the same fashion as the intermolecular reactions to give a p-cyclophane. With intermediate tether lengths (n = 10, 13), the reaction gives an additional p-cyclophane in which the two oxygen substituents are meta on the arene ring. This type of product is unprecedented from the reaction of carbene complexes and alkynes and is quite surprising because the formation of this product requires that the carbon-carbon bond between the alpha- and beta-carbons of the vinyl carbene complex is broken. A mechanism is proposed to account for this process which involves the crossed intramolecular [2 + 2] cycloaddition of the alkene and a ketene in a conjugated dienyl ketene to give a benzvalenone paddalane intermediate. 相似文献

Elemental white phosphorus (P4) is well recognized as a critical precursor to organophosphorus compounds. However, regulatory constraints stemming from the toxic and pyrophoric nature of white phosphorus have significantly limited its accessibility. Herein is described a new approach to white phosphorus storage and release based on a unique example of photolytic reductive elimination of the tetrahedral P4 molecule from a mononuclear cyclo‐P4 molybdenum complex. The latter functions as an air‐stable, chemically‐deactivated source of white phosphorus. The system features efficient photo‐release of white phosphorus using inexpensive violet LED sources. Additionally, high‐yield recapture of unspent white phosphorus by the molybdenum center can be achieved by post‐photolysis heating at convenient temperatures. 相似文献

Spatiotemporal control over biochemical signaling processes involving G protein‐coupled receptors (GPCRs) is highly desired for dissecting their complex intracellular signaling. We developed sixteen photoswitchable ligands for the human histamine H3 receptor (hH3R). Upon illumination, key compound 65 decreases its affinity for the hH3R by 8.5‐fold and its potency in hH3R‐mediated Gi protein activation by over 20‐fold, with the trans and cis isomer both acting as full agonist. In real‐time two‐electrode voltage clamp experiments in Xenopus oocytes, 65 shows rapid light‐induced modulation of hH3R activity. Ligand 65 shows good binding selectivity amongst the histamine receptor subfamily and has good photolytic stability. In all, 65 (VUF15000) is the first photoswitchable GPCR agonist confirmed to be modulated through its affinity and potency upon photoswitching while maintaining its intrinsic activity, rendering it a new chemical biology tool for spatiotemporal control of GPCR activation. 相似文献

The varying coordination modes of the title ligand, L [5-methyl-1-(pyridin-2-yl)-N′-[pyridin-2-ylmethylidene]pyrazole-3-carbohydrazide] towards the different metal centers is reported by preparation and characterization of Cu(II), Zn(II) and Cd(II) complexes, [Cu(L)NO3.H2O](NO3) (1) [Zn(L)2](ClO4)2·2DMF (2) and [Cd(L)(I)2] (3) respectively. In 1, the neutral ligand serves as tetradentate 4 N donor where both pyridine and pyrazole nitrogen atoms of pyridyl–pyrazole part are coordinatively active, leaving the carbonyl oxygen of the carbohydrazide part inactive. The same pyridine and pyrazole N atoms remain abstained from the coordination process towards the Zn(II) and Cd(II) metal centers. For 2 and 3 the ligand behaves as a tridentate NNO donor where the two nitrogen atoms come from azomethine, pyridine of pyridine-2-carbaldehyde parts and O from carbonyl oxygen atoms (carbohydrazide part). The complex 1 and 2 are distorted octahedral while complex 3 adopts distorted square pyramidal geometry. All the complexes are X-ray crystallographically characterized. 相似文献

A new ternary polar intermetallic, Ca(18)Li(5)In(25.07), was obtained from high-temperature reactions of the elements in welded Nb tubes. Its crystal structure, established by single-crystal X-ray diffraction, was found to crystallize in the orthorhombic space group Cmmm (No. 65). Unit cell parameters are a = 9.9151(6) A, b = 26.432(2) A, and c = 10.2116(6) A; Z = 2. The structure of Ca(18)Li(5)In(25.07) features two distinct types of indium anionic layers. An "electron-deficient" layer is made up of Li-centered In(12) icosahedra that are interconnected by bridging planar In(4) units and In atoms. A second In(3)(5-) layer is an electron-precise Zintl layer formed by fused four-, five-, and six-membered rings of three- and four-bonded indium atoms. The two distinct layers are alternately stacked and linked into a complex three-dimensional network. Vacancies are observed to occur only at the In(12) icosahedral and the bridging indium units within the "electron-deficient" layers. Magnetic property measurements indicate that Ca(18)Li(5)In(25.07) exhibits temperature-independent paramagnetism consistent with metallic behavior. Band structure calculations were performed to elucidate the role of defects and vacancies in the electronic structure of the electron-deficient "metallic" Zintl phase. 相似文献
