A study was made for purification of cephamycin C from fermentation of Streptomyces clavuligerus. Initially, the culture broth was clarified by microfiltration and ultrafiltration, after which the resulting permeates were subjected to nonspecific adsorption and ion-exchange chromatography on resin columns. The antibiotic activity was measured by the biological method at each stage by assaying its activity against the Escherichia coli ESS, super sensitive to β-lactam antibiotic. The purification processes were assessed in relation to the variables affecting each step. The purification efficiency by nonspecific adsorption was monitored by UV spectrophotometry, while the ion-exchange adsorption fractions were assessed by NMR spectroscopy. Some of the fractions obtained during purification were also analyzed by mass spectrometry (LC/MS and LC/MS/MS) to identify the cephamycin C molecule. These preliminary results proved the process feasibility.  相似文献

Background

The subjective haptic vertical (SHV) task requires subjects to adjust the roll orientation of an object, mostly in the roll plane, in such a way that it is parallel to perceived direction of gravity. Previously we found a tendency for clockwise rod rotations to deviate counter-clockwise and vice versa, indicating hysteresis. However, the contributing factors remained unclear. To clarify this we characterized the SHV in terms of handedness, hand used, direction of hand rotation, type of grasping (wrap vs. precision grip) and gender, and compared findings with perceived straight-ahead (PSA). Healthy subjects repetitively performed adjustments along SHV (n?=?21) and PSA (n?=?10) in complete darkness.

Results

For both SHV and PSA significant effects of the hand used and the direction of rod/plate rotation were found. The latter effect was similar for SHV and PSA, leading to significantly larger counter-clockwise shifts (relative to true earth-vertical and objective straight-ahead) for clockwise rotations compared to counter-clockwise rotations irrespective of the handedness and the type of grip. The effect of hand used, however, was opposite in the two tasks: while the SHV showed a counter-clockwise bias when the right hand was used and no bias for the left hand, in the PSA a counter-clockwise bias was obtained for the left hand without a bias for the right hand. No effects of grip and handedness (studied for SHV only) on accuracy were observed, however, SHV precision was significantly (p?<?0.005) better in right-handed subjects compared to left-handed subjects and in male subjects.

Conclusions

Unimanual haptic tasks require control for the hand used and the type of grip as these factors significantly affect task performance. Furthermore, aligning objects with the SHV and PSA resulted in systematic direction-dependent deviations that could not be attributed to handedness, the hand used, or the type of grip. These deviations are consistent with hysteresis and are likely not related to gravitational pull, as they were observed in both planes tested, i.e. parallel and perpendicular to gravity. Short-term adaptation that shifts attention towards previous adjustment positions may provide an explanation for such biases of spatial orientation in both the horizontal and frontal plane.  相似文献

The photophysical behavior of BCECF [2',7'-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein]--currently the most widely used fluorescent pH indicator for near-neutral intracellular pH measurements--has been explored by using absorption and steady-state and time-resolved fluorescence measurements. The influence of ionic strength as well as total buffer concentration on the absorbance and steady-state fluorescence has been investigated. The apparent acidity constant of the pH indicator determined by absorbance and fluorescence titration is dependent on the added buffer and salt concentrations. A semiempirical model is proposed to rationalize the variations in the apparent pKa values. The excited-state proton exchange of BCECF at physiological pH becomes reversible upon addition of phosphate buffer, inducing a pH-dependent change of the fluorescence decay times. Fluorescence decay traces collected as a function of total buffer concentration and pH were analyzed by global compartmental analysis yielding the following values of the rate constants describing excited-state dynamics of BCECF: k01 = 3.4 x 10(8) s(-1), k02 = 2.6 x 10(8) s(-1), k21 approximately 1 x 10(6) M(-1) s(-1), k12(B) = 1.4 x 10(8) M(-1) s(-1), and k21(B) = 4.3 x 10(7) M(-1) s(-1).  相似文献

A 2-D native-PAGE/SDS-PAGE method for detecting the subunit components of protein oligomers at low picomole sensitivity is presented. IgG was electrophoresed in a native acidic polyacrylamide gel in amounts ranging from 51 pmol to 60 fmol. Silver-staining (native fast silver stain, ammoniacal silver stain, permanganate silver stain), Coomassie-staining (R-250, G-250), metal ion-reverse-staining (zinc, copper), and fluorescent chromophore-staining (SYPRO Ruby) methods were used to visualize the IgG oligomers. The protein zones were then excised, separated by SDS-PAGE, and subunits visualized with a permanganate silver stain. The Coomassie R-250/permanganate silver-staining combination detected IgG subunits using 2 pmol of sample. Coomassie G-250 and native fast silver staining in the first-dimensional gel produced detectable subunits in the second-dimensional separation at 3 and 13 pmol, respectively. Staining with silver (ammoniacal, permanganate), copper, zinc, or SYPRO Ruby in the first-dimensional gel did not produce discernible subunits in the second-dimensional gels due to protein streaking or protein immobilization in the native gel. When using a 2-D native-PAGE/SDS-PAGE system, Coomassie staining of the first-dimensional native gel combined with permanganate silver staining of the second-dimensional denaturing gel provides the most sensitive method (2-3 pmol) for visualizing constituent subunits from their oligomeric assemblies.  相似文献

Combining the Berends-Giele and on-shell recursion relations we obtain an extremely compact expression for the scattering amplitude of a complex massive scalar-antiscalar pair and an arbitrary number of positive helicity gluons. This is one of the basic building blocks for constructing other helicity configurations from recursion relations. We also show explicitly that the scattering amplitude of massive fermions to gluons, all with positive helicity, is proportional to the scalar one, confirming in this way the recently advocated SUSY-like Ward identities relating both amplitudes.  相似文献

We report a single-mode, highly stable, continuous-wave Ho3+: ZBLAN fiber laser. The system was pumped by a 1175-nm fiber Raman laser and emitted at ~2950 nm. The optical-to-optical conversion efficiency was 43% that is the highest reported in the literature. Characterizations revealed a quasigaussian intensity profile with a power stability of less than 1%. This high stability relies on the single-wavelength emission of the system because the system does not require simultaneous oscillation at 2100 nm. Coincidence between the emission wavelength and the absorption peak of water makes the system suitable to replace a high-power (~9.34-W) Er3+: ZBLAN fiber laser for cutting materials with high moisture content.  相似文献

In the theory of nonlinear elasticity universal relations are relationships connecting the components of stress and deformation tensors that hold independently of the constitutive equation for the considered class (or sub-class) of materials. They are classified as linear or nonlinear according as the components of the stress appear linearly or nonlinearly in the relations. In this paper a general scheme is developed for the derivation of nonlinear universal relations and is applied to the constitutive law of an isotropic Cauchy elastic solid. In particular, we consider examples of quadratic and cubic universal relations. In respect of universal solutions our results confirm the general result of Pucci and Saccomandi [1] that nonlinear universal relations are necessarily generated by the linear ones. On the other hand, for non-universal solutions we develop a general method for generating nonlinear universal relations and illustrate the results in the case of cubic relations. X-ray diffraction data for four different liquid crystalline side group methacrylic polymers based on the ω-hexyloxysalicylaldimine moiety with p-alkyl or p-alkyloxy terminal substituents are presented. For the decyloxy, dodecyloxy and octyl derivatives, a smectic C2 phase occurs over a broad temperature range, while for the tetradecyl derivative a complex behaviour was observed on cooling from the isotropic state. In this last case a smectic Ad phase arises, changing to a smectic C2 phase and a further smectic Cd phase. A possible explanation for the observed phenomena is presented.  相似文献

This report is about the X-ray diffraction and Mössbauer studies of three impactite samples denominated PMe-8, PMe-9 and PMe-11 from Huamalies Province in Huánuco Region, Peru. When terrestrial rocks are submitted to pressures higher than 60 GP, the majority is completely melted, forming a kind of glass called impactites. X-ray diffraction indicates the presence of quartz as the principal mineralogical phase in all samples. The 57Fe Mössbauer spectra at room temperature of samples PMe-8 and PMe-9 show broadened spectra that were fitted using a distribution model. The most probable field of the magnetic component is 34 T, corresponding to the presence of small particles of goethite, confirmed by the 4.2 K spectrum. For the sample PMe-11, the MS showed the presence of well crystallized hematite.  相似文献
