Fibrin glue increases the cell survival and the transduced gene product secretion of the ceiling culture-derived adipocytes transplanted in mice

The development of clinically applicable scaffolds is important for the application of cell transplantation in various human diseases. The aims of this study are to evaluate fibrin glue in a novel protein replacement therapy using proliferative adipocytes and to develop a mouse model system to monitor the delivery of the transgene product into the blood and the fate of the transduced cells after transplantation. Proliferative adipocytes from mouse adipose tissue were transduced by a retroviral vector harboring the human lecithin-cholesterol acyltransferase (lcat) gene, and were subcutaneously transplanted into mice combined with fibrin glue. The lcat gene transduction efficiency and the subsequent secretion of the product in mouse adipocytes were enhanced using a protamine concentration of 500 μg/ml. Adipogenesis induction did not significantly affect the lcat gene-transduced cell survival after transplantation. Immunohistochemistry showed the ectopic enzyme production to persist for 28 days in the subcutaneously transplanted gene- transduced adipocytes. The increased viability of transplanted cells with fibrin glue was accompanied with the decrease in apoptotic cell death. The immunodetectable serum LCAT levels in mice implanted with the fibrin glue were comparable with those observed in mice implanted with Matrigel, indicating that the transplanted lcat gene-transduced adipocytes survived and functioned in the transplanted spaces with fibrin glue as well as with Matrigel for 28 days. Thus, this in vivo system using fibrin is expected to serve as a good model to further improve the transplanted cell/scaffold conditions for the stable and durable cell-based replacement of defective proteins in patients with LCAT deficiency.     

Singular values of some modular functions

For an integer N greater than 5 and a triple \({\mathfrak{a}}=[a_{1},a_{2},a_{3}]\) of integers with the properties 0<a _i ≤N/2 and a _i ≠a _j for i≠j, we consider a modular function \(W_{\mathfrak{a}}(\tau)=\frac{\wp (a_{1}/N;L_{\tau})-\wp (a_{3}/N;L_{\tau})}{\wp (a_{2}/N;L_{\tau})-\wp(a_{3}/N;L_{\tau})}\) for the modular group Γ ₁(N), where ?(z;L _τ ) is the Weierstrass ?-function relative to the lattice L _τ generated by 1 and a complex number τ with positive imaginary part. For a pair of such triples \({\mathfrak{A}}=[{\mathfrak{a}},{\mathfrak{b}}]\) and a pair of non-negative integers F=[m,n], we define a modular function \(T_{{\mathfrak{A}},F}\) for the group Γ ₀(N) as the trace of the product \(W_{\mathfrak{a}}^{m}W_{\mathfrak{b}}^{n}\) to the modular function field of Γ ₀(N). In this article, we study the integrality of singular values of the functions \(W_{\mathfrak{a}}\) and \(T_{{\mathfrak{A}},F}\) by using their modular equations. We prove that the functions \(T_{{\mathfrak{A}},F}\) for suitably chosen \({\mathfrak{A}}\) and F generate the modular function field of Γ ₀(N), and from Shimura reciprocity and Gee–Stevenhagen method we obtain that singular values \(T_{{\mathfrak{A}},F}(\tau)\) for suitably chosen \({\mathfrak{A}}\) and F generate ring class fields. Further, we study the class polynomial of \(T_{{\mathfrak{A}},F}\) for Schertz N-system.

Effects of pressure and temperature on the survival rate of adherent A-172 cells

Preservation of cells under high pressure is an important alternative to cryopreservation. We studied the effect of temperature (4, 25, 37°C) and pressure (0.1–350 MPa) on the survival rate of A-172 glioblastoma cells. The survival rate was not changed by brief (10 min) pressurization of up to 150 MPa, but the survival rate began to decrease from 150 MPa, and most of the A-172 cells died when treated with over 200 MPa. Lengthy pressurization (4 days) at lower pressure (upto 20.1 MPa) without medium exchange showed complex results. The survival rate of cells preserved at 25°C showed two maxima at 1.6 and 20.1 MPa. After preservation, cells adhered and proliferated in the same way as normal cells when cultured at 37°C in a CO₂ incubator. The other two temperatures, 4° and 37°C, showed no maximum survival rate. Therefore, a high survival rate can be maintained with high pressure treatment.     

AC transport current loss of assembled conductor of HTS tapes

We study experimentally and theoretically the AC transport current loss characteristics of a tape in multiple tapes assembled in single layer and subject to external field produced by transport currents of adjacent tapes. We measured the AC transport current losses of a Bi2223 silver-sheathed tape in a single layer arrangement of three tapes using our newly developed potential leads arrangement to avoid spurious loss components caused by the magnetization in the adjacent tapes. In the paper, the influence of the external AC field produced by adjacent tapes on the loss characteristics is studied based on the experimental results and theoretical analysis.

A new class of chiral calix[4]arenes as receptors with planar chirality

The modification of title compounds to chiral receptors and their characterization are reported. The two synthetic methods were developed. The racemates obtained could be resolved to each stable enantiomer by a chiral HPLC column. Chiral calixarenes were designed as the receptors with planar chirality. The (−)-receptor strongly forms 1:1 complex with (R)-(+)--phenylethylammonium picrate.     

Analytical application of porphyrins-I Spectrophotometric determination of ultramicro amounts of copper with alpha,beta,gamma,delta-tetra-(3-N-methylpyridyl)porphine

A new water-soluble porphyrin, alpha,beta,gamma,delta-tetra(3-N-methylpyridyl)porphine [T(3-MPy)P], has been synthesized, and its reaction with copper(II) studied spectrophotometrically. Sensitive methods for the determination of copper at ppM level have been established. The spectrophotometric sensitivity and relative standard deviation of the recommended procedure for copper are 0.18 ng/cm(2) (for A = 0.001) and 1.0%, respectively. The interference of 19 ions and 20 salts has been examined, and found to be minimal. The methods established could be applied to the determination of copper in water samples with satisfactory results.
