Five cytotoxic macrolides, aplyronines D–H (48), were isolated from the Japanese sea hare Aplysia kurodai. They are new congeners of the antitumor compound aplyronine A (1), which was previously isolated from the same organism. Their structures were determined by spectroscopic analysis (NMR and MS). The cytotoxicity of these new compounds was evaluated in comparison with that of aplyronines A–C (13), suggesting the importance of the 7-O-seryl ester group for mediating the potent cytotoxicity of aplyronines.  相似文献

Efficient methods for the synthesis of aminomethylated azaindole derivatives via domino copper-catalyzed multicomponent coupling and cyclization have been developed. Using various secondary amines and aldehydes, N-substituted 3-ethynyl-4-aminopyridine was converted to substituted azaindoles in moderate to excellent yields. By use of a 3,4-diaminopyridine derivative bearing two alkynyl groups, the corresponding pyrrole-fused azaindoles were synthesized by controlled stepwise cyclization.  相似文献

Phoborhodopsin (pR; also called sensory rhodopsin II, SRII) is a photoreceptor of negative phototaxis of halobacteria. The studies of photochemical properties of this pigment are not many because the amount of the pigment is small and the stability is low. Recently an expression system of phoborhodopsin from Halobacterium salinarum (called salinarum phoborhodopsin, spR; also HsSRII) in Escherichia coli and purification method has been developed (Mironova et al. [2005] FEBS Lett., 579, 3147–3151), which enables detailed studies on the photochemical properties of spR. In the present work, the photoreaction cycle of E. coli-expressed spR was studied by low-temperature spectroscopy and flash photolysis. Formations of K-, M-, O-like intermediates and P480 were reconfirmed as reported previously. New findings are as follows. (1) The K-like intermediate (P500) was a mixture of two photoproducts. (2) Formation of L-like intermediate (P482) was observed by low-temperature spectroscopy and flash photolysis at room temperature. (3) On long irradiation of spR at 20°C, formation of a new photoproduct P370 was observed and it decayed to the original spR in the dark with a decay half time of 190 min. Based on these results the similarities and dissimilarities between spR and ppR are discussed.  相似文献

This review overviews recent reports on the electroanalytical applications of boron-doped diamond (BDD) electrodes. Because BDD electrodes have excellent features for electroanalysis, such as wide potential window, low background current, electrochemical stability, and fouling resistance, they can be useful for sensitive and stable detection of various substances, including drugs, bio-related substances, metal ions, and organic pollutants. Many articles have reported high-sensitivity detection of real samples, demonstrating that this electrode material is practically applicable. Surface modification of the BDD electrodes using metal nanoparticles, nanocarbons, and polymers can increase the sensitivity of the electrochemical detection. Furthermore, research on the electroanalytical device equipped with BDD electrodes will be expanded by combining peripheral technologies related to the device fabrication.  相似文献

The chemical modification of proteins is an effective technique for manipulating the properties and functions of proteins, and for creating protein-based materials. The N-terminus is a promising target for single-site modification that provides modified proteins with uniform structures and properties. In this paper, a copper(II)-mediated aldol reaction with 2-pyridinecarboxaldehyde (2-PC) derivatives is proposed as an operationally simple method to selectively modify the N-terminus of peptides and proteins at room temperature and physiological pH. The copper(II) ion activates the N-terminal amino acids by complexation with an imine of the N-terminal amino acid and 2-PCs, realizing the selective formation of the nucleophilic intermediate at the N-terminus. This results in a stable carbon-carbon bond between the 2-PCs and the α-carbon of various N-terminal amino acids. The reaction is applied to four different proteins, including biopharmaceuticals such as filgrastim and trastuzumab. The modified trastuzumab retains the human epidermal growth factor receptor 2 recognition activity.  相似文献

Fibroadenomas (FAs) and phyllodes tumors (PTs) are major benign breast tumors, pathologically classified as fibroepithelial tumors. Although the clinical management of PTs differs from FAs, distinction by core needle biopsy diagnoses is still challenging. Here, a combined technique of label-free imaging with multi-photon microscopy and artificial intelligence was applied to detect quantitative signatures that differentiate fibroepithelial lesions. Multi-photon excited autofluorescence and second harmonic generation (SHG) signals were detected in tissue sections. A pixel-wise semantic segmentation method using a deep learning framework was used to separate epithelial and stromal regions automatically. The epithelial to stromal area ratio and the collagen SHG signal strength were investigated for their ability to distinguish fibroepithelial lesions. An image segmentation analysis with a pixel-wise semantic segmentation framework using a deep convolutional neural network showed the accurate separation of epithelial and stromal regions. A further investigation, to determine if scoring the epithelial to stromal area ratio and the SHG signal strength within the stromal area could be a marker for differentiating fibroepithelial tumors, showed accurate classification. Therefore, molecular and morphological changes, detected through the assistance of computational and label-free multi-photon imaging techniques, enable us to propose quantitative signatures for epithelial and stromal alterations in breast tissues.  相似文献

Anionic ring-opening polymerization (ROP) behavior of trans-cyclohexene carbonate (CHC) using metal alkoxides as initiators was investigated. As a result, lithium tert-butoxide-initiated ROP of CHC with a high-monomer concentration (10 M) at low temperature (−15 to −10°C) proceeded to afford a poly(trans-cyclohexene carbonate) (PCHC) without undesired side reactions such as mainly backbiting. The suppression of side reactions enables the control of the molecular weight (Mn = 2400–6100) of PCHC with low molar-mass dispersity values (Mw/Mn = 1.16–1.22). Furthermore, by increasing the feed ratio of the monomer to the initiator, the molecular weight increases proportionally, indicating a controllable polymerization. The results of a matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis, a kinetic study, and a chain extension experiment suggested a living nature of this ROP using lithium tert-butoxide.  相似文献

In order to clarify the mechanism conferring heat resistance on phthalate esters, those with a substituent on the β-carbon atom, such as bis(2-aminobutyl) phthalate, bis(2-nitrobutyl) phthalate, bis(2,4-diphenylbutyl) phthalate and dineopentyl phthalate, were synthesized and their thermal stabilities were studied by thermogravimetry and differential thermal analysis. The analytical results for these phthalate esters were compared with those for dibutyl phthalate, with a straight alkyl chain. As the temperatures required for a 3% weight loss of phthalate esters with a substituent, an electron-donating group (amino group) or an electron-accepting group (nitro group) on the β-carbon atom move to the higher end of the range, the effect of the adjacent group was recognized. The presence of a phenyl group in phthalate esters considerably improved the heat resistance. It is considered that the high heat resistance of bis(2,4-diphenylbutyl) phthalate is due to the obstruction of the planar configuration for cis elimination by the phenyl group and hindrance by the phenyl group of the formation of the six-membered cyclic transition state owing to the interaction between non-bondable molecules.  相似文献
